POINT technology illuminates the processing of polymerase-associated intact nascent transcripts.
Rui Sousa-Luís,Gwendal Dujardin,Inna Zukher,Hiroshi Kimura,Carika Weldon,Maria Carmo-Fonseca,Nick J. Proudfoot,Takayuki Nojima,Takayuki Nojima +8 more
TL;DR: In this paper, a new approach to characterize nascent RNA, called polymerase intact nascent transcript (POINT) technology, is presented, which maps nascent RNA 5′ ends, establishes the kinetics of co-transcriptional splicing patterns, and profiles whole transcription units.

Point Technology Illuminates The Processing Of Polymerase-Associated Intact Nascent Transcripts
Rui Sousa-Luís,Gwendal Dujardin,Inna Zukher,Hiroshi Kimura,Maria Carmo-Fonseca,Nick J. Proudfoot,Takayuki Nojima +6 more
TL;DR: This work shows by depletion of the nuclear exonuclease Xrn2 that this activity acts selectively on cleaved 5’P-RNA at polyadenylation sites, and reveals that splicing occurs either immediately after splice site transcription or is delayed until Pol II transcribes downstream sequences.
Journal ArticleDOI
Counteracting chromatin effects of a splicing-correcting antisense oligonucleotide improves its therapeutic efficacy in spinal muscular atrophy
Luciano E Marasco,Gwendal Dujardin,Rui Sousa-Luís,Ying Hsiu Liu,José N. Stigliano,Tomoki T. Nomakuchi,Nick J. Proudfoot,Adrian R. Krainer,Alberto R. Kornblihtt +8 more
TL;DR: In this paper , the histone deacetylase inhibitor VPA cooperates with an antisense oligonucleotide (ASO) to promote E7 inclusion in spinal muscular atrophy (SMA).
Journal ArticleDOI
Transcriptome profiling of human pluripotent stem cell‐derived cerebellar organoids reveals faster commitment under dynamic conditions
Teresa Silva,Teresa Silva,Rui Sousa-Luís,Tiago G. Fernandes,Evguenia Bekman,Evguenia Bekman,Carlos A V Rodrigues,Sandra H. Vaz,Sandra H. Vaz,Leonilde M. Moreira,Yas Hashimura,Sunghoon Jung,Brian Lee,Maria Carmo-Fonseca,Joaquim M. S. Cabral +14 more
TL;DR: In this article, a reproducible and scalable production of cerebellar organoids by using the single-use Vertical-Wheel bioreactors was presented, in which functional cerebella neurons were obtained.
Journal ArticleDOI
Pseudouridylation: A new player in co-transcriptional splicing regulation.
TL;DR: This paper showed that uridines at splice sites and splicing regulatory motifs in the pre-mRNA may be converted to pseudouridine during transcription and impact splicing decisions.
