Structure−Activity Relationship of 6-Methylidene Penems Bearing Tricyclic Heterocycles as Broad-Spectrum β-Lactamase Inhibitors: Crystallographic Structures Show Unexpected Binding of 1,4-Thiazepine Intermediates
Aranapakam M. Venkatesan,Yansong Gu,Osvaldo Dos Santos,T. Abe,Atul Agarwal,Youjun Yang,Petersen Peter J,William J. Weiss,Tarek S. Mansour,Michiyoshi Nukaga,Andrea M. Hujer,Robert A. Bonomo,James R. Knox +12 more
TL;DR: The design and synthesis of a series of seven tricyclic 6-methylidene penems as novel class A and C serine beta-lactamase inhibitors is described and the formation of the 1,4- thiazepine ring structures is proposed based on a 7-endo-trig cyclization.
Journal ArticleDOI
Highly diastereoselective alkylation onto 4-acetoxy-2-azetidinones employing tin(II) enolates of 3-acyl C-4-chiral 1,3-thiazolidine-2-thiones
Yoshimitsu Nagao,T. Kumagai,S. Tamai,T. Abe,Y. Kuramoto,Tooru Taga,Sakae Aoyagi,Y. Nagase,Masahito Ochiai,Y. Inoue,Eiichi Fujita +10 more
Journal ArticleDOI
Inhibition of Class A and Class C Beta-Lactamases by Penems: Crystallographic Structures of a Novel 1,4-Thiazepine Intermediate
Michiyoshi Nukaga,T. Abe,Aranapakam M. Venkatesan,Tarek S. Mansour,Robert A. Bonomo,James R. Knox +5 more
TL;DR: A new beta-lactamase inhibitor, a methylidene penem having a 5,6-dihydro-8H-imidazo[2,1-c][1,4]oxazine heterocyclic substituent at the C6 position with a Z configuration, irreversibly inhibits both class A and class C serine beta- lactamases.
Journal ArticleDOI
Structure-Activity Relationship of 6-Methylidene Penems Bearing 6,5 Bicyclic Heterocycles as Broad-Spectrum β-Lactamase Inhibitors: Evidence for 1,4-Thiazepine Intermediates with C7 R Stereochemistry by Computational Methods
Aranapakam M. Venkatesan,Atul Agarwal,T. Abe,Hideki Ushirogochi,Itsuka Yamamura,Mihira Ado,Takasaki Tsuyoshi,Osvaldo Dos Santos,Yansong Gu,Fuk-Wah Sum,Li Zhong,Gerry Francisco,Yang-I. Lin,Petersen Peter J,Youjun Yang,T. Kumagai,William J. Weiss,David M. Shlaes,James R. Knox,Tarek S. Mansour +19 more
TL;DR: The design and synthesis of a series of 6-methylidene penems containing [6,5]-fused bicycles as novel class A, B, and C beta-lactamase inhibitors is described, and penems proposed to form a seven-membered 1,4 thiazepine ring in both class A and C Beta- lactamases are proposed.
Journal ArticleDOI
Inhibition of OXA-1 β-Lactamase by Penems
Christopher R. Bethel,Anne M. Distler,Mark W. Ruszczycky,Marianne P. Carey,Paul R. Carey,Andrea M. Hujer,Magda Taracila,Marion S. Helfand,Marion S. Helfand,Jodi M. Thomson,Matthew Kalp,Vernon E. Anderson,David A. Leonard,Kristine M. Hujer,T. Abe,Aranapakam M. Venkatesan,Tarek S. Mansour,Robert A. Bonomo +17 more
TL;DR: Penem β-lactamase inhibitors which demonstrate high affinities and which form long-lived acyl intermediates may prove to be extremely useful against the broad range of inhibitor-resistant serine β- lactamases present in gram-negative bacteria.
