Journal ArticleDOI
Intratumoral IL-12 delivery empowers CAR-T cell immunotherapy in a pre-clinical model of glioblastoma
Giulia Agliardi,Anna Rita Liuzzi,Alastair Hotblack,Donatella De Feo,Nicolás Gonzalo Núñez,Cassandra L Stowe,Ekaterina Friebel,Francesco Nannini,Lukas Rindlisbacher,Thomas A. Roberts,Rajiv Ramasawmy,Iwan P. Williams,Bernard Siow,Bernard Siow,Mark F. Lythgoe,Tammy L. Kalber,Sergio A. Quezada,Martin Pule,Sonia Tugues,Karin Straathof,Karin Straathof,Burkhard Becher +21 more
TL;DR: This article showed that IL-12 not only boosts cytotoxicity of CAR-T cells, but also reshapes the TME, driving increased infiltration of proinflammatory CD4+ T cells, decreased numbers of regulatory T cells and activation of the myeloid compartment.
Journal ArticleDOI
Computational fluid dynamics with imaging of cleared tissue and of in vivo perfusion predicts drug uptake and treatment responses in tumours.
Angela d'Esposito,Paul W. Sweeney,Morium Ali,Magdy Saleh,Rajiv Ramasawmy,Thomas A. Roberts,Giulia Agliardi,Adrien E. Desjardins,Mark F. Lythgoe,R. Barbara Pedley,Rebecca J. Shipley,Simon Walker-Samuel +11 more
TL;DR: The combination of mathematical modelling of tumour tissue, high-resolution optical imaging of intact and optically cleared tumours from animal models, and in vivo imaging of vascular perfusion in tumours predicts the tumour uptake and distribution of specific therapeutic agents.
Journal ArticleDOI
Integrated and efficient diffusion-relaxometry using ZEBRA
Jana Hutter,Paddy J. Slator,Daan Christiaens,Rui Pedro A. G. Teixeira,Thomas A. Roberts,Laurence H. Jackson,Anthony N. Price,Shaihan J. Malik,Joseph V. Hajnal +8 more
TL;DR: In this paper, a fully integrated sequence simultaneously sampling the acquisition parameter spaces required for T1 and T2* relaxometry and diffusion MRI is proposed, where slice-level interleaved diffusion encoding, multiple spin/gradient echoes and slice-shuffling are combined for higher efficiency and sampling flexibility.
Journal ArticleDOI
Fluorescence characterization of clinically-important bacteria.
TL;DR: This fluorescence survey found that an emission peak of 340nm, from an excitation source at 280nm, was the cellular fluorescence signal to target for detection of bacterial contamination, and the majority of materials analysed offer a spectral window through which bacterial contamination could indeed be detected.
Journal ArticleDOI
Vaccination to improve the persistence of CD19CAR gene-modified T cells in relapsed pediatric acute lymphoblastic leukemia.
Claudia Rossig,Martin Pule,Bianca Altvater,S Saiagh,G Wright,Sara Ghorashian,Laura Clifton-Hadley,Kim Champion,Z Sattar,Bilyana Popova,Allan Hackshaw,Paul Smith,Thomas A. Roberts,Ettore Biagi,B Dreno,Raphael Rousseau,Sareetha Kailayangiri,Martina Ahlmann,Rachael Hough,Bernhard Kremens,Martin Sauer,Paul Veys,N Goulden,Michelle Cummins,Persis Amrolia,Persis Amrolia +25 more
TL;DR: Although CD19CAR CTL expansion was poor, persistence was enhanced by vaccination, demonstrating the feasibility of multi-center studies of CAR T cell therapy and the potential for enhancing persistence with vaccination.
