Abstract 606: Targeting the immune checkpoint PTPN2 with ABBV-CLS-484 inflames the tumor microenvironment and unleashes potent CD8+ T cell immunity
Arvin Iracheta-Vellve,Hakimeh Ebrahimi-Nik,Thomas Davis,Kira E Olander,Sarah Kim,Mitchell Yeary,James C. Patti,Ian C. Kohnle,Christina K. Baumgartner,Keith M. Hamel,Kathleen A. McGuire,Cun Lan Chuong,Zhaoming Xiong,Elliot P. Farney,Jennifer M. Frost,Matthew G. Rees,Andrew S Boghossian,Melissa M. Ronan,Jennifer M. Roth,Todd R. Golub,Gabriel K. Griffin,Clay Beauregard,Philip R. Kym,Kathleen B. Yates,Robert T. Manguso +24 more
TL;DR: ABBV-CLS-484 is a highly effective immunotherapy with monotherapy efficacy across mouse tumor models and small molecule inhibitors of PTPN2 offer a promising new strategy for cancer immunotherapy by targeting an IFN signaling checkpoint.
Journal ArticleDOI
Single duplex DNA sequencing with CODEC detects mutations with high sensitivity
Jin H. Bae,Ruolin Liu,Eugenia M. Roberts,Erica Nguyen,Shervin Tabrizi,Justin Rhoades,T. M. Blewett,K. Xiong,Gregory Gydush,D. Shea,Zhenyi An,Sahil B. Patel,S. Sridhar,Mei Hong Liu,Emilie Lassen,Anne-Bine Skytte,Marta Grońska-Pęski,Jonathan Shoag,Gilad D. Evrony,Heather A. Parsons,Erica L. Mayer,G. Mike Makrigiorgos,Todd R. Golub,Viktor A. Adalsteinsson +23 more
TL;DR: Concatenating original duplex for error correction (CODEC) as mentioned in this paper was proposed to detect biologically significant mutations from single DNA molecules using up to 100-fold fewer reads than duplex sequencing.
Proceedings ArticleDOI
Abstract 105: Integrative analysis of genomic and pharmacologic data from the Cancer Cell Line Encyclopedia
Nicolas Stransky,Kavitha Venkhatesan,Michael Morrissey,Jordi Barretina,Pichai Raman,Gad Getz,Mike Berger,Cory M. Johannessen,Adam Callahan,Paula Morais,Scott Mahan,Supriya Gupta,Robert C. Onofrio,Carrie Sougnez,Wendy Winckler,Lili Niu,Sarah M. Kehoe,Charlie Hatton,Ted Liefeld,Yan Ding,Dmitriy Sonkin,Michael D. Jones,Giordiano Caponigro,Christine D. Wilson,Sungjoon Kim,John Che,Andrew I. Su,Laura E. MacConaill,Stacey Gabriel,Kristin G. Ardlie,Michael R. Reich,Markus Warmuth,Matthew Meyerson,Todd R. Golub,Jennifer L. Harris,William R. Sellers,Bob Schlegel,Levi A. Garraway +37 more
TL;DR: Significant regions of genomic gains and losses have been linked to expression and mutation data to find significant correlations at both single-gene and pathway levels and suggest that this integrative approach applied to a robust cancer cell line collection has considerable power to discover novel associations that augment ongoing basic research into cancer biology and drug discovery.
Journal ArticleDOI

An intravenous DNA-binding priming agent protects cell-free DNA and improves the sensitivity of liquid biopsies
Shervin Tabrizi,Carmen Martín-Alonso,Kan Xiong,T. M. Blewett,S. Sridhar,Zhenyi An,Sahil B. Patel,Sergio A. Rodriguez-Aponte,C. A. Naranjo,Shi-Ting Wang,D. Shea,Todd R. Golub,Sangeeta N. Bhatia,Viktor A. Adalsteinsson,J. Christopher Love +14 more
TL;DR: In this article , an intravenous DNA-binding priming agent given 2 hours prior to a blood draw was used to attenuate the clearance of cfDNA in vivo, which improved the sensitivity of existing DNA-based cancer testing using blood biopsies.
