The aim of microfluidic mixing is to achieve a thorough and rapid mixing of multiple samples in microscale devices. In such devices, sample mixing is essentially achieved by enhancing the diffusion effect between the different species flows. Broadly speaking, microfluidic mixing schemes can be categorized as either “active”, where an external energy force is applied to perturb the sample species, or “passive”, where the contact area and contact time of the species samples are increased through specially-designed microchannel configurations. Many mixers have been proposed to facilitate this task over the past 10 years. Accordingly, this paper commences by providing a high level overview of the field of microfluidic mixing devices before describing some of the more significant proposals for active and passive mixers.

Microfluidic Mixing: A Review

The recent introduction of surface acoustic wave (SAW) technology onto lab-on-a-chip platforms has opened a new frontier in microfluidics. The advantages provided by such SAW microfluidics are numerous: simple fabrication, high biocompatibility, fast fluid actuation, versatility, compact and inexpensive devices and accessories, contact-free particle manipulation, and compatibility with other microfluidic components. We believe that these advantages enable SAW microfluidics to play a significant role in a variety of applications in biology, chemistry, engineering and medicine. In this review article, we discuss the theory underpinning SAWs and their interactions with particles and the contacting fluids in which they are suspended. We then review the SAW-enabled microfluidic devices demonstrated to date, starting with devices that accomplish fluid mixing and transport through the use of travelling SAW; we follow that by reviewing the more recent innovations achieved with standing SAW that enable such actions as particle/cell focusing, sorting and patterning. Finally, we look forward and appraise where the discipline of SAW microfluidics could go next.

Surface acoustic wave microfluidics

Acoustic tweezers are a versatile set of tools that use sound waves to manipulate bioparticles ranging from nanometer-sized extracellular vesicles to millimeter-sized multicellular organisms. Over the past several decades, the capabilities of acoustic tweezers have expanded from simplistic particle trapping to precise rotation and translation of cells and organisms in three dimensions. Recent advances have led to reconfigured acoustic tweezers that are capable of separating, enriching, and patterning bioparticles in complex solutions. Here, we review the history and fundamentals of acoustic-tweezer technology and summarize recent breakthroughs. This review discusses and contrasts different acoustic-tweezer technologies and their applications in biology.

Acoustic tweezers for the life sciences

In part 14 of the tutorial series “Acoustofluidics – exploiting ultrasonic standing wave forces and acoustic streaming in microfluidic systems for cell and particle manipulation”, we provide a qualitative description of acoustic streaming and review its applications in lab-on-a-chip devices. The paper covers boundary layer driven streaming, including Schlichting and Rayleigh streaming, Eckart streaming in the bulk fluid, cavitation microstreaming and surface-acoustic-wave-driven streaming.

Flow characteristics in microfluidic devices is naturally laminar due to the small channel dimensions. Mixing based on molecular diffusion is generally poor. In this article, we report the fabrication and characterization of active surface-acousticwave-driven micromixers which exploit the acoustic streaming effect to significantly improve the mixing efficiency. A side-by-side flow of water and fluorescent dye solution was driven by a syringe pump. Surface wave with a frequency of 13 MHz was launched perpendicular to the flow. The wave was generated by two designs of interdigitated electrodes on LiNbO3 substrate: parallel electrodes and focusing electrodes. The mixing efficiency was observed to be proportional to the square of the applied voltage. Under the same applied voltage, the focusing type offers a better mixing efficiency. The fabrication of the micromixer is compatible to current technology such as soft lithography and deep reactive ion etching. Despite the high throughput and fast mixing time, the mixer design is simple and could be integrated into any microfluidic platform.

High-throughput micromixers based on acoustic streaming induced by surface acoustic wave

his paper presents a systematic overview on the recent works on surface acoustic wave (SAW) driven microfluidics. SAW microfluidics is based on acoustic streaming induced by leaky SAW radiation into a liquid. The development of this field attracts attention from microfluidic research community due to its rapid actuation, programmable capability, simple and yet efficient operation. In our paper, SAW microfluidic applications are categorized into droplet-based applications and continuous-flow applications. Droplet is actuated into unique behaviours depending on the applied SAW power. A wide range of droplet based applications have been employed utilizing these behaviours. In continuous-flow system, applications are further categorized based on the interaction of travelling SAW and standing SAW with the bulk liquid. Finally, future perspectives of SAW driven microfluidics are discussed

Surface Acoustic Wave Driven Microfluidics – A Review

This paper reports the fabrication and characterization of an adhesive-based liquid-metal microcoil for magnetic resonance relaxometry (MRR). Conventionally, microcoils are fabricated by various techniques such as electroplating, microcontact printing and focused ion beam milling. These techniques require considerable fabrication efforts and incur high cost. In this paper, we demonstrate a novel technique to fabricate three-dimensional multilayer liquid-metal microcoils together with the microfluidic network by lamination of dry adhesive sheets. One of the unique features of the adhesive-based technique is that the detachable sample chamber can be disposed after each experiment and the microcoil can be reused without cross-contamination multiple times. The integrated microcoil has a low direct-current (DC) resistance of 0.3 Ω and a relatively high inductance of 67.5 nH leading to a high quality factor of approximately 30 at 21.65 MHz. The microcoil was characterized for ∼0.5 T proton MRR measurements. The optimal pulse duration, amplitude, and frequency for the 90° pulse were 131 μs, −30 dB (1.56 W) and 21.6553 MHz, respectively. In addition, we used the liquid-metal microcoil to perform a parametric study on the transverse relaxation rate of human red blood cells at different hematocrit levels. The transverse relaxation rate increases quadratically with the hematocrit level. The results from the liquid-metal microcoil were verified by measurements with a conventional solenoid coil.

Bjerknes Forces in Motion: Long-Range Translational Motion and Chiral Directionality Switching in Bubble-Propelled Micromotors via an Ultrasonic Pathway

Flow cytometers are widely applied to environmental monitoring, industrial testing, and biochemical studies. Integrating a flow cytometer into microfluidic networks helps to miniaturize the system and make it portable for field use. The integration of optical components, such as lenses, further improves the compactness and thus has been intensively studied recently. However, the current designs suffer from severe light scattering due to the roughness of the solid-based lens interface. In this Letter, we propose a flow cytometer using an optofluidic lens to focus the light beam. Benefiting from the smooth liquid-liquid lens interface and the refractive-index matching liquid as cladding streams, a light beam can be well focused without scattering. The variations of the signal peak values are reduced, owing to the small beam width at the beam waist. The device presents an efficient and accurate performance on both the counting and sizing of particles.
