Journal ArticleDOI
Metabolic biomarker signature to differentiate pancreatic ductal adenocarcinoma from chronic pancreatitis
Julia Mayerle,Holger Kalthoff,Regina Reszka,Beate Kamlage,Erik Peter,Bodo Schniewind,Sandra González Maldonado,Christian Pilarsky,Claus-Dieter Heidecke,Philipp Schatz,Marius Distler,Jonas A. Scheiber,Ujjwal M. Mahajan,F. Ulrich Weiss,Robert Grützmann,Markus M. Lerch +15 more
TL;DR: A biomarker signature (nine metabolites and additionally CA19-9) was identified for the differential diagnosis between PDAC and CP and results in a negative predictive value of 99.9% (95% CI 99.7%–99.9%) in patients with CP with an increased risk for pancreatic cancer.
Journal ArticleDOI
Cathepsin B-Mediated Activation of Trypsinogen in Endocytosing Macrophages Increases Severity of Pancreatitis in Mice.
Matthias Sendler,Frank-Ulrich Weiss,Janine Golchert,Georg Homuth,Cindy van den Brandt,Ujjwal M. Mahajan,Lars-Ivo Partecke,Paula Döring,Ilya Gukovsky,Anna S. Gukovskaya,Preshit R. Wagh,Markus M. Lerch,Julia Mayerle,Julia Mayerle +13 more
TL;DR: Studying pancreatitis in mice, activation of digestive proteases to occur not only in acinar cells but also in macrophages that infiltrate pancreatic tissue is found, and contributes to systemic inflammation and severity of pancreatitis.
Journal ArticleDOI
NLRP3 Inflammasome Regulates Development of Systemic Inflammatory Response and Compensatory Anti-Inflammatory Response Syndromes in Mice With Acute Pancreatitis
Matthias Sendler,Cindy van den Brandt,Juliane Glaubitz,Anika Wilden,Janine Golchert,Frank Ulrich Weiss,Georg Homuth,Laura L. De Freitas Chama,Neha Mishra,Ujjwal M. Mahajan,Lukas Bossaller,Uwe Völker,Barbara M. Bröker,Julia Mayerle,Julia Mayerle,Markus M. Lerch +15 more
TL;DR: In mice with severe pancreatitis, SIRS and CARS are found to develop in parallel and inhibition of NLRP3 reduces SIRs and CARs, and might be used to treat patients with severe Pancreatitis.
Journal ArticleDOI
Immune Cell and Stromal Signature Associated With Progression-Free Survival of Patients With Resected Pancreatic Ductal Adenocarcinoma.
Ujjwal M. Mahajan,Eno Langhoff,Elisabetta Goni,Eithne Costello,William Greenhalf,Christopher Halloran,Steffen Ormanns,Stephan Kruger,Stefan Boeck,Silvia Ribback,Georg Beyer,Georg Beyer,Frank Dombroswki,Frank-Ulrich Weiss,John P. Neoptolemos,Jens Werner,Jan G. D’Haese,Alexandr V. Bazhin,Julian Peterhansl,Svenja Pichlmeier,Markus W. Büchler,Jörg Kleeff,Paula Ganeh,Matthias Sendler,Daniel H. Palmer,Thomas Kohlmann,Roland Rad,Ivonne Regel,Markus M. Lerch,Julia Mayerle,Julia Mayerle +30 more
TL;DR: In an analysis of PDAC tissue microarray specimens, a histologic signature is identified and validated that associates with PFS times of patients with resected PDACs and provides new insights into the immune response to PDAC.
Journal ArticleDOI
Complement Component 5 Mediates Development of Fibrosis, via Activation of Stellate Cells, in 2 Mouse Models of Chronic Pancreatitis
Matthias Sendler,Georg Beyer,Ujjwal M. Mahajan,Vivien Kauschke,Sandrina Maertin,Claudia Schurmann,Georg Homuth,Uwe Völker,Henry Völzke,Walter Halangk,Thomas Wartmann,Frank-Ulrich Weiss,Péter Hegyi,Markus M. Lerch,Julia Mayerle +14 more
TL;DR: In mice, loss of C5 or injection of a C5a-receptor antagonist significantly reduced the level of fibrosis of chronic pancreatitis, but this was not a consequence of milder disease in early stages of pancreatitis.
