The current study investigated the effects of deltamethrin, fluoride (F(-)) and their combination on the hepatic oxidative stress and consequent alterations in blood biochemical markers of hepatic damage in rats. Significant hepatic oxidative stress and hepatic damage were observed in the toxicant exposed groups. These changes were higher in the deltamethrin-F(-) co-exposure treatment group, depicting a positive interaction between the two chemicals.

Sub-acute Deltamethrin and Fluoride Toxicity Induced Hepatic Oxidative Stress and Biochemical Alterations in Rats

Pesticides are widespread synthesized substances used for public health protection and agricultural programs. However, they cause environmental pollution and health hazards. This study aimed to examine the protective effects of sesame oil (SO) on the genetic alterations induced by cypermethrin (CYP) in the liver and kidney of Wistar rats. Male rats were divided into four groups, each containing 10 rats: the control group received vehicle, SO group (5 mL/kg b.w), CYP group (12 mg/kg b.w), and protective group received SO (5 mL/kg b.w) plus CYP (12 mg/kg b.w). Biochemical analysis showed an increase in albumin, urea, creatinine, GPT, GOT, and lipid profiles in the CYP group. Co-administration of SO with CYP normalized such biochemical changes. CYP administration decreased both the activity and mRNA expression of the examined antioxidants. SO co-administration recovered CYP, downregulating the expression of glutathione-S-transferase (GST), catalase, and superoxide dismutase. Additionally, SO co-administration with CYP counteracted the CYP- altering the expression of renal interleukins (IL-1 and IL-6), tumor necrosis factor alpha (TNF-α), heme oxygenase-1 (HO-1), anigotensinogen (AGT), AGT receptors (AT1), and genes of hepatic glucose and fatty acids metabolism. CYP induced degenerative changes in the kidney and liver histology which are ameliorated by SO. In conclusion, SO has a protective effect against alterations and pathological changes induced by CYP in the liver and kidney at genetic and histological levels.

Genetic and histopathological alterations induced by cypermethrin in rat kidney and liver: Protection by sesame oil.

Recently, melatonin has attracted attention because of their strong free radical scavenging and antioxidant properties. The present study was designed to investigate the role of melatonin against cypermethrin induced hepatotoxicity in rats using qualitative, quantitative and biochemical approaches. Adult female Wistar rats were randomly divided into four groups; each group consists of six animals. Control group received vehicle, corn oil; Cypermethrin (CM) (250mg/kg bw); Melatonin (MEL) (10 mg/kg bw); CM+MEL treated group. Animals were treated orally, daily for a period of 4 weeks. Animals were sacrificed by decapitation and liver was used for various biochemical and histopathological examinations. The result of this study shows that in vivo administration of cypermethrin caused a significant induction of oxidative damage in liver tissue as evidenced by depletion of proteins, glutathione (GSH) and increased lipid peroxidation in hepatic tissue. Cypermethrin administration significantly enhanced the activities of enzymatic antioxidants (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) glutathione reductase (GR) and glutathione-S- transferase (GST). Cypermethrin intoxication exhibited elevation in the activities of liver marker enzymes such as serum glutamate pyruvate transaminase (SGPT) serum glutamate oxaloacetate transaminase (SGOT) and Lactate dehydrogenase (LDH). Furthermore, the co-administration of melatonin mitigates the hepatotoxicity of cypermethrin by normalizing the biochemical and enzymatic parameters. The biochemical observations were supported by histopathological examination of liver. The findings of this investigation suggest that the melatonin may play a protective role against cypermethrin induced hepatotoxicity and oxidative damage in female albino rats.

Ameliorative role of melatonin against cypermethrin induced hepatotoxicity and impaired antioxidant defense system in Wistar rats

Alpha-cypermethrin (α-CYP) is one of the most widely used insecticides. It may become an air pollutant and adversely affect the health. The present study was designed to determine whether treatment with N-acetyl cysteine (NAC), a well-known antioxidant, can be useful for the management of the deleterious effects of α-CYP on lung tissues. For this purpose, thirty two male rats were divided into four different groups (eight rats for each). Group (I) gavaged with corn oil (control group), group (II) gavaged daily with NAC (150 mg kg−1 body weight), group (III) gavaged with α-CYP (14.5 mg kg−1 body weight/day, dissolved in corn oil), group (IV) gavaged with NAC then with α-CYP 2 h later for 12 weeks. α-CYP significantly increased serum lactate dehydrogenase (LDH) and pulmonary malondialdehyde (MDA) levels, while decreased the activities of catalase (CAT) and superoxide dismutase (SOD) as well as reduced glutathione (GSH) content in lung. It also provoked higher levels of serum nitric oxide (NO), lung interleukin-1 beta (IL-1β), tumor necrosis factor-alpha (TNF-α), hydroxyproline (Hyp) as well as heme oxygenase-1 (HO-1), inducible nitric oxide synthase (iNOS) and nuclear factor-kappa B (NF-КB) gene expression in lung tissues. Histopathological alterations in lung with congestion, cellular infiltration, necrotic changes and thickening of inter-alveolar septa were observed following α-CYP administration. NAC reduced the adverse effects of α-CYP on lung tissues and improved the histological architecture of lung since it showed antioxidant, anti-inflammatory and antifibrotic effects on lung tissues. Our results indicate that NAC exerts a potent protective effect against α-CYP-induced oxidative damage and inflammation in lung tissues. © 2013 Wiley Periodicals, Inc. Ameliorative effect of N‐acetyl cysteine on alpha‐cypermethrin‐induced pulmonary toxicity in male rats

Bifenthrin is an insecticide, used extensively on some crops, including corn. Roughly 70 % of all US-grown hops and raspberries are treated with bifenthrin. The current study on bifenthrin was undertaken to investigate the potential alterations in biochemical parameters, induced by sub-acute oral exposure of bifenthrin, in goats. The animals were randomly divided into two groups. Group A (n = 3) received only tap water and served as control, whereas, Group B goats (n = 4) received bifenthrin by gavage (Telstar 10 EC) at the dose rate of 5 mg Kg−1 body weight for 28 consecutive days. Bifenthrin exposed group showed significant alterations in the enzyme biochemical parameters. The activities of erythrocyte cholinesterase and plasma cholinesterase reduced significantly from 7th day and 28th day of bifenthrin exposure, respectively. The activity of plasma aspartate aminotransferase and alanine aminotransferase increased significantly from the 21st day of bifenthrin exposure. The activity of acid phosphatase and alkaline phosphatase increased significantly from the 14th day of bifenthrin exposure. All these parameters returned to the non-significant levels by 7th day of post-exposure period. Non-significant alterations in blood urea nitrogen, creatinine and plasma proteins were also observed. The current study thus reveals that sub-acute oral administration of bifenthrin produces reversible liver damage.

Effect of Sub-Acute Oral Exposure of Bifenthrin on Biochemical Parameters in Crossbred Goats

Pyrethrins are obtained from the flowers of Chrysanthemum cinerarifolium. These are strong insecticides with low mammalian toxicity. The toxic effects of pyrethroid cypermethrin were studied using various biochemical parameters along with histopathological changes in a 30-day study in Wistar rats. The rats were divided into two groups. Rats of the test group were given sublethal doses of cypermethrin (14.5 mg/kg) by gavage once daily for 30 days and the control rats were given an equal volume of the vehicle. The animals were sacrificed on day 0, 10, 20 and 30 of the study. The results showed that cypermethrin caused a significant increase in the levels of serum aspartate transaminase, alanine transaminase, lactate dehydrogenase and plasma creatinine. It significantly lowered the levels of total proteins. The histopathological studies on various organs like liver, lungs, kidneys and heart were carried out. The changes in various biochemical parameters correlated well with the histopathological changes in various organs. In conclusion, the results of this study demonstrate that subacute oral administration of cypermethrin, even in low doses such as 1/10 LD50 for 30 days induces toxic effects on different vital organs.
