Photodynamic therapy for leishmaniasis: recent advances and future trends.

Nanomaterials (NMs) are the leading edge as an amazing class of materials that consists of at least one dimension in the range of 1–100 nm. NMs can be made with exceptional magnetic, electrical, and catalytic properties different from their bulk counterparts. We summarized unique features of NMs, their synthesis, and advances in agri-nanotechnology and cutting-edge nanobionics. The review describes advances in NMs including their applications, dosimetry to ensure biosafety, remote sensing of agro-forestry fields, nanofertilizers, and nanopesticides, and avoid post-harvest losses, gene delivery, and nanobionics. Tree nanobionics has enabled the synthesis and delivery of nanosensors, which enhance the rate of photosynthesis, detection of pathogens, and poisonous residues to ensure biosafety and biomass accumulation. Finally, we conclude by discussing challenges, future perspectives, and agro-ecological risks of using NMs. Graphical Abstract

Agri-Nanotechnology and Tree Nanobionics: Augmentation in Crop Yield, Biosafety, and Biomass Accumulation

Leishmaniasis is a serious parasitic disease caused by Leishmania spp. transmitted through sandfly bites. This disease is a major public health concern worldwide. It can occur in 3 different clinical forms: cutaneous, mucocutaneous, and visceral leishmaniasis (CL, MCL, and VL, respectively), caused by different Leishmania spp. Currently, licensed vaccines are unavailable for the treatment of human leishmaniasis. The treatment and prevention of this disease rely mainly on chemotherapeutics, which are highly toxic and have an increasing resistance problem. The development of a safe, effective, and affordable vaccine for all forms of vector-borne disease is urgently needed to block transmission of the parasite between the host and vector. Immunological mechanisms in the pathogenesis of leishmaniasis are complex. IL-12-driven Th1-type immune response plays a crucial role in host protection. The essential purpose of vaccination is to establish a protective immune response. To date, numerous vaccine studies have been conducted using live/attenuated/killed parasites, fractionated parasites, subunits, recombinant or DNA technology, delivery systems, and chimeric peptides. Most of these studies were limited to animals. In addition, standardization has not been achieved in these studies due to the differences in the virulence dynamics of the Leishmania spp. and the feasibility of the adjuvants. More studies are needed to develop a safe and effective vaccine, which is the most promising approach against Leishmania infection.

Immunotherapy for visceral leishmaniasis: A trapeze of balancing counteractive forces.

Amphotericin B (AmpB) is a polyene macrolide antibiotic used in the treatment of blood-borne parasitic and fungal infections. However, its use, particularly in the developing world, has been limited by dose-dependent kidney toxicity, other systemic-related toxicity issues following injection, the inconvenience of parenteral administration, and accessibility. Oral formulation approaches have focused on the dual problem of solubility and permeability of AmpB, which is poorly water soluble, amphoteric and has extremely low oral bioavailability. Therefore, to enhance oral absorption, researchers have employed micellar formulations, polymeric nanoparticles, cochleates, pro-drugs, and self-emulsifying drug delivery systems (SEDDS). This paper will highlight current uses of AmpB against parasitic infections such as leishmaniasis, preclinical and clinical formulation strategies, applications in veterinary medicine and the importance of developing a cost-effective and safe oral AmpB formulation.
