Abstract: We report the investigation of composites of poly-(3-hexylthiophene) (P3HT) and CuInSe2 nanocrystals (NCs). CuInSe2 NCs were synthesized at gram scale through a colloidal route. Cyclic voltammetry was used to determine the HOMO–LUMO energy levels, as well as the energies of trap states intrinsic to CuInSe2 NCs. The results suggest that nanocrystal–polymer charge transfer is foreseeable in the P3HT and CuInSe2 system because of their type II band alignment. We studied optical properties of the hybrid composite films of P3HT and CuInSe2 NCs as a function of P3HT/CuInSe2 NCs weight ratios. The quenching of photoluminescence spectra with the increase of CuInSe2 ratio in the composite was interpreted to result from charge transfer between the polymer and CuInSe2 NCs. We further studied photodiode devices that exhibited a transition from photoconductive gain to the photovoltaic effect with an increase of the P3HT/CuInSe2 weight ratio. Abstract: The TGF-β pathway plays an essential role in regulating cell proliferation and differentiation. GWASs and candidate approaches have identified a battery of genetic variants in the TGF-β pathway contributing to colorectal cancer (CRC). However, most of the significant variants are common variants and their functions remain ambiguous. To identify causal variants with low-frequency in the TGF-β pathway contributing to CRC susceptibility in Chinese population, we performed targeted sequencing of 12 key genes in TGF-β signaling in CRC patients followed by a two-stage case-control study with a total of 5109 cases and 5169 controls. Bioinformatic annotations and biochemical experiments were applied to reveal the potential functions of significant variants. Seven low-frequency genetic variants were captured through targeted sequencing. The two stage association studies showed that missense variant rs3764482 (c. 83C>T; p. S28F) in the gene SMAD7 was consistently and significantly associated with CRC risk. Compared with the wild type, the ORs for variant allele were 1.37 (95%CI: 1.10-1.70, P = 0.005), 1.55 (95%CI: 1.30-1.86, P = 1.15 × 106 ), and 1.48 (1.29-1.70, P = 2.44 × 10;8 ) in stage 1, stage 2, and the combined analyses, respectively. Functional annotations revealed that the minor allele T of rs3764482 was more effective than the major allele C in blocking the TGF-β signaling and inhibiting the phosphorylation of receptor-regulated SMADs (R-SMADs). In conclusion, low-frequency coding variant rs3764482 in SMAD7 is associated with CRC risk in Chinese population. The rs3764482 variant may block the TGF-β signaling via impeding the activation of downstream genes, leading to cancer cell proliferation, thus contributing to CRC pathogenesis.

Abstract: To understand virus and host interactions and host responses to rotavirus infection in children, we analysed by real-time polymerase chain reaction (PCR) the expression of mRNA for five Toll-like receptors (TLRs) (TLR2, TLR3, TLR4, TLR7 and TLR8) and four T helper (Th)1 and Th2 cytokines [interleukin (IL)-2, IL-12, interferon (IFN)-gamma and IL-4) in peripheral blood mononuclear cells (PBMC) of children with acute rotavirus diarrhoea. We observed significantly higher expression of genes encoding TLR2, TLR3, TLR4, TLR7 and TLR8 in PBMC of 41% (31/75) patients within 3 days of illness onset than those in healthy children. After 3 days of illness onset, only TLR3 and TLR8 mRNA expressions were still significantly (P<0.05) increased in 59% (44/75) children with diarrhoea. We also observed significantly (P<0.05) elevated expression of IL-12p40 and IFN-gamma in PBMC of patients during the entire period of illness and the first 3 days of illness, respectively. We further demonstrated a weak but significant association between elevated levels of gene expression of four TLRs (TLR2, TLR3, TLR4 and TLR8) and IFN-gamma. Our results suggest that multiple TLRs may modulate the immune response in the acute phase of rotavirus infection and play a role in the activation of IFN-gamma.

Abstract: A series of intramolecular charge transfer (ICT) molecules with multiple dicyanovinyl substitutes on triphenylamine were synthesized in good yields by Knoevenagel condensation of corresponding triphenylamine aldehydes and malonodinitrile. Photophysical property of these triphenylamine malonitrile dyes were studied, along with their intriguing solvatochromic behavior. The fluorescence emission of most dyes was highly sensitive to solvent polarity, yielding blue to yellow and even red fluorescence in different organic solvents. Both the fluorescence and the Stokes shifts were linearly dependent on the orientation polarizability (f) and empirical polarity parameter E T(30) according to Reichardt-Dimroth equation. Further analysis suggested that the solvatochromic behavior of these dyes was caused by dipole moment change upon excitation from ground to excited state. 2013 Elsevier B.V. All rights reserved.

Abstract: Three parallel algorithms for classical molecular dynamics are presented. The first assigns each processor a fixed subset of atoms; the second assigns each a fixed subset of inter-atomic forces to compute; the third assigns each a fixed spatial region. The algorithms are suitable for molecular dynamics models which can be difficult to parallelize efficiently—those with short-range forces where the neighbors of each atom change rapidly. They can be implemented on any distributed-memory parallel machine which allows for message-passing of data between independently executing processors. The algorithms are tested on a standard Lennard-Jones benchmark problem for system sizes ranging from 500 to 100,000,000 atoms on several parallel supercomputers--the nCUBE 2, Intel iPSC/860 and Paragon, and Cray T3D. Comparing the results to the fastest reported vectorized Cray Y-MP and C90 algorithm shows that the current generation of parallel machines is competitive with conventional vector supercomputers even for small problems. For large problems, the spatial algorithm achieves parallel efficiencies of 90% and a 1840-node Intel Paragon performs up to 165 faster than a single Cray C9O processor. Trade-offs between the three algorithms and guidelines for adapting them to more complex molecular dynamics simulations are also discussed.
