Abstract: This study describes a novel analytical system integrating in vivo microdialysis sampling with a radial thin-layer flow cell with a single-walled carbon nanotube (SWNT)-modified glassy carbon electrode as working electrode for continuous and on-line monitoring of ascorbate depletion in the rat striatum induced by global ischemia. The SWNTs, especially those after vacuum heat treatment at 500 °C, are found to be able to enhance the electron-transfer kinetics of ascorbate oxidation at a low potential (ca. −50 mV) and possess a strong ability against electrode fouling. These properties essentially make it possible to determine ascorbate with a good stability and high selectivity against catecholamines and their metabolites and other electroactive species of physiological levels. While being integrated with in vivo microdialysis to assemble an on-line analytical system, the electrode is proved useful for continuous and sensitive monitoring of the basal dialysate level of ascorbate and its depletion in the rat...

Abstract: Magnesium-aluminum hydrotalcite compounds have been synthesized without possible contamination by alkali cations. Pure hydrotalcite compounds with Mg/Al ratios from 3 to 12 were synthesized without the formation of separate phases. These materials were converted into magnesium-aluminum mixed oxides upon calcination at 773 K, showing the X-ray diffraction pattern of MgO. Microcalorimetric and infrared spectroscopic measurements of NH[sub 3] and CO[sub 2] adsorption were used to characterize the number, strength, and nature of acid/base sites on the surface of the calcined hydrotalcites. These materials do not possess acid sites and base sites with differential heats higher than 120 kJ/mol, respectively. In contrast, [gamma]-Al[sub 2]O[sub 3] possesses acid sites with differential heats from about 60 to kJ/mol, and MgO has base sites with differential heats as high as 170 kJ/mol. Infrared spectroscopy revealed that the acid sites on calcined hydrotalcites are mainly Lewis acids sites, while the base sites are oxygen anions. Calcination of hydrotalcites can be used to prepare Mg-Al-O mixed oxides with high surface are and moderate acidity and basicity. 35 refs., 11 figs., 2 tabs.

Abstract: Aims. A transient astrophysical event observed in both gravitational wave (GW) and electromagnetic (EM) channels would yield rich scientific rewards. A first program initiating EM follow-ups to possible transient GW events has been developed and exercised by the LIGO and Virgo community in association with several partners. In this paper, we describe and evaluate the methods used to promptly identify and localize GW event candidates and to request images of targeted sky locations. Methods. During two observing periods (Dec. 17, 2009 to Jan. 8, 2010 and Sep. 2 to Oct. 20, 2010), a low-latency analysis pipeline was used to identify GW event candidates and to reconstruct maps of possible sky locations. A catalog of nearby galaxies and Milky Way globular clusters was used to select the most promising sky positions to be imaged, and this directional information was delivered to EM observatories with time lags of about thirty minutes. A Monte Carlo simulation has been used to evaluate the low-latency GW pipeline's ability to reconstruct source positions correctly. Results. For signals near the detection threshold, our low-latency algorithms often localized simulated GW burst signals to tens of square degrees, while neutron star/neutron star inspirals and neutron star/black hole inspirals were localized to a few hundred square degrees. Localization precision improves for moderately stronger signals. The correct sky location of signals well above threshold and originating from nearby galaxies may be observed with similar to 50% or better probability with a few pointings of wide-field telescopes.

Abstract: Novel surface-enhanced Raman scattering (SERS) substrates with high SERS-activity are ideal for novel SERS sensors, detectors to detect illicitly sold narcotics and explosives. The key to the wider application of SERS technique is to develop plasmon resonant structure with novel geometries to enhance Raman signals and to control the periodic ordering of these structures over a large area to obtain reproducible Raman enhancement. In this work, a simple Ar(+)-ion sputtering route has been developed to fabricate silver nanoneedles arrays on silicon substrates for SERS-active substrates to detect trace-level illicitly sold narcotics. These silver nanoneedles possess a very sharp apex with an apex diameter of 15 nm and an apex angle of 20°. The SERS enhancement factor of greater than 10(10) was reproducibly achieved by the well-aligned nanoneedles arrays. Furthermore, ketamine hydrochloride molecules, one kind of illicitly sold narcotics, can be detected down to 27 ppb by using our SERS substrate within 3 s, indicating the sensitivity of our SERS substrates for trace amounts of narcotics and that SERS technology can become an important analytical technique in forensic laboratories because it can provide a rapid and nondestructive method for trace detection.

Abstract: Kyoto Encyclopedia of Genes and Genomes (KEGG) is a knowledge base for systematic analysis of gene functions in terms of the networks of genes and molecules. The major component of KEGG is the PATHWAY database that consists of graphical diagrams of biochemical pathways including most of the known metabolic pathways and some of the known regulatory pathways. The pathway information is also represented by the ortholog group tables summarizing orthologous and paralogous gene groups among different organisms. KEGG maintains the GENES database for the gene catalogs of all organisms with complete genomes and selected organisms with partial genomes, which are continuously re-annotated, as well as the LIGAND database for chemical compounds and enzymes. Each gene catalog is associated with the graphical genome map for chromosomal locations that is represented by Java applet. In addition to the data collection efforts, KEGG develops and provides various computational tools, such as for reconstructing biochemical pathways from the complete genome sequence and for predicting gene regulatory networks from the gene expression profiles. The KEGG databases are daily updated and made freely available (http://www.genome.ad.jp/kegg/).

Abstract: limma is an R/Bioconductor software package that provides an integrated solution for analysing data from gene expression experiments. It contains rich features for handling complex experimental designs and for information borrowing to overcome the problem of small sample sizes. Over the past decade, limma has been a popular choice for gene discovery through differential expression analyses of microarray and high-throughput PCR data. The package contains particularly strong facilities for reading, normalizing and exploring such data. Recently, the capabilities of limma have been significantly expanded in two important directions. First, the package can now perform both differential expression and differential splicing analyses of RNA sequencing (RNA-seq) data. All the downstream analysis tools previously restricted to microarray data are now available for RNA-seq as well. These capabilities allow users to analyse both RNA-seq and microarray data with very similar pipelines. Second, the package is now able to go past the traditional gene-wise expression analyses in a variety of ways, analysing expression profiles in terms of co-regulated sets of genes or in terms of higher-order expression signatures. This provides enhanced possibilities for biological interpretation of gene expression differences. This article reviews the philosophy and design of the limma package, summarizing both new and historical features, with an emphasis on recent enhancements and features that have not been previously described.
