TL;DR: This sequence was completed in two months at approximately one-hundredth of the cost of traditional capillary electrophoresis methods and demonstrated the acquisition of novel human sequence, including novel genes not previously identified by traditional genomic sequencing, which is the first genome sequenced by next-generation technologies.

TL;DR: In this paper, the authors proposed methods of sequencing both the sense and antisense strands of DNA through the use of blocked and unblocked sequencing primers, and they described the steps of annealing an unblockable primer to a first strand of nucleic acid, denoising a blocked primer to a second strand, and elongating the nucleic acids along the first strand with a polymerase.
