Abiotic stresses are the major environmental factors that play a significant role in decreasing plant yield and production potential by influencing physiological, biochemical, and molecular processes. Abiotic stresses and global population growth have prompted scientists to use beneficial strategies to ensure food security. The use of organic compounds to improve tolerance to abiotic stresses has been considered for many years. For example, the application of potential external osmotic protective compounds such as proline is one of the approaches to counteract the adverse effects of abiotic stresses on plants. Proline level increases in plants in response to environmental stress. Proline accumulation is not just a signal of tension. Rather, according to research discussed in this article, this biomolecule improves plant resistance to abiotic stress by rising photosynthesis, enzymatic and non-enzymatic antioxidant activity, regulating osmolyte concentration, and sodium and potassium homeostasis. In this review, we discuss the biosynthesis, sensing, signaling, and transport of proline and its role in the development of various plant tissues, including seeds, floral components, and vegetative tissues. Further, the impacts of exogenous proline utilization under various non-living stresses such as drought, salinity, high and low temperatures, and heavy metals have been extensively studied. Numerous various studies have shown that exogenous proline can improve plant growth, yield, and stress tolerance under adverse environmental factors.

Contribution of Exogenous Proline to Abiotic Stresses Tolerance in Plants: A Review

Metabolic regulation is the key mechanism implicated in plants maintaining cell osmotic potential under drought stress. Understanding drought stress tolerance in plants will have a significant impact on food security in the face of increasingly harsh climatic conditions. Plant primary and secondary metabolites and metabolic genes are key factors in drought tolerance through their involvement in diverse metabolic pathways. Physio-biochemical and molecular strategies involved in plant tolerance mechanisms could be exploited to increase plant survival under drought stress. This review summarizes the most updated findings on primary and secondary metabolites involved in drought stress. We also examine the application of useful metabolic genes and their molecular responses to drought tolerance in plants and discuss possible strategies to help plants to counteract unfavorable drought periods.

Metabolomics and Molecular Approaches Reveal Drought Stress Tolerance in Plants.

Soil salinization, which is aggravated by climate change and inappropriate anthropogenic activities, has emerged as a serious environmental problem, threatening sustainable agriculture and future food security. Although there has been considerable progress in developing crop varieties by introducing salt tolerance-associated traits, most crop cultivars grown in saline soils still exhibit a decline in yield, necessitating the search for alternatives. Halophytes, with their intrinsic salt tolerance characteristics, are known to have great potential in rehabilitating salt-contaminated soils to support plant growth in saline soils by employing various strategies, including phytoremediation. In addition, the recent identification and characterization of salt tolerance-related genes encoding signaling components from halophytes, which are naturally grown under high salinity, have paved the way for the development of transgenic crops with improved salt tolerance. In this review, we aim to provide a comprehensive update on salinity-induced negative effects on soils and plants, including alterations of physicochemical properties in soils, and changes in physiological and biochemical processes and ion disparities in plants. We also review the physiological and biochemical adaptation strategies that help halophytes grow and survive in salinity-affected areas. Furthermore, we illustrate the halophyte-mediated phytoremediation process in salinity-affected areas, as well as their potential impacts on soil properties. Importantly, based on the recent findings on salt tolerance mechanisms in halophytes, we also comprehensively discuss the potential of improving salt tolerance in crop plants by introducing candidate genes related to antiporters, ion transporters, antioxidants, and defense proteins from halophytes for conserving sustainable agriculture in salinity-prone areas.

Adaptive Mechanisms of Halophytes and Their Potential in Improving Salinity Tolerance in Plants

Climate change is predicted to increase the frequency and intensity of abiotic stress combinations that negatively impact plants and pose a serious threat to crop yield and food supply. Plants respond to episodes of stress combination by activating specific physiological and molecular responses, as well as by adjusting different metabolic pathways, to mitigate the negative effects of stress combination on plant growth, development, and reproduction. Plants synthesize a wide range of metabolites that regulate many aspects of plant growth and development, as well as plant responses to stress. Although metabolic responses to individual abiotic stresses have been studied extensively in different plant species, recent efforts have been directed at understanding metabolic responses that occur when different abiotic factors are combined. In this review we examine recent studies of metabolomic changes under stress combination in different plants and suggest new avenues for the development of stress combination-resilient crops based on metabolites as breeding targets.

Responses of plants to climate change: Metabolic changes during abiotic stress combination in plants.

Under the present era of changing climate, plants face simultaneous abiotic pressures rather than single stress. Under these unprecedented and joint environmental pressures, thorough research efforts toward controlling such major stresses should be done. A pot experiment was, therefore, conducted to unravel the salicylic acid (SA) mediated underlying defense mechanisms under concurrent stress (drought and salt) conditions in two varieties of Vicia faba L. (Assiut wardy and Assiut 84). The results revealed that separate and combined drought and salt stress decreased growth kinetic traits, photosynthetic pigments, water relations and mineral contents but increased oxidative stress biomarkers, reactive oxygen species (ROS) production, enzymatic and non-enzymatic antioxidant gadgets, osmolytes and secondary metabolites. Application of SA to drought- and/or salt-stressed plants reduced oxidative damage by triggering the modulation of the activities of antioxidants and maintaining an enhanced pool of reducing agents under drought and/or salt stress conditions differentially in two varieties of faba bean. Thus, the application of SA to drought- and/or salt-stressed faba bean varieties could be used as a potential tool to induce resistance for increasing growth and crop yield under today’s era of climate change.

Salicylic Acid Spraying-Induced Resilience Strategies Against the Damaging Impacts of Drought and/or Salinity Stress in Two Varieties of Vicia faba L. Seedlings

Plants grow on soils that not only provide support for root anchorage but also act as a reservoir of water and nutrients important for plant growth and development. However, environmental factors, such as high salinity, hinder the uptake of nutrients and water from the soil and reduce the quality and productivity of plants. Under high salinity, plants attempt to maintain cellular homeostasis through the production of numerous stress-associated endogenous metabolites that can help mitigate the stress. Both primary and secondary metabolites can significantly contribute to survival and the maintenance of growth and development of plants on saline soils. Existing studies have suggested that seed/plant-priming with exogenous metabolites is a promising approach to increase crop tolerance to salt stress without manipulation of the genome. Recent advancements have also been made in genetic engineering of various metabolic genes involved in regulation of plant responses and protection of the cells during salinity, which have therefore resulted in many more basic and applied studies in both model and crop plants. In this review, we discuss the recent findings of metabolic reprogramming, exogenous treatments with metabolites and genetic engineering of metabolic genes for the improvement of plant salt tolerance.

Enhancing Salt Tolerance of Plants: From Metabolic Reprogramming to Exogenous Chemical Treatments and Molecular Approaches.

Strigolactone and karrikin receptors, DWARF14 (D14) and KARRIKIN INSENSITIVE 2 (KAI2), respectively, have been shown to positively regulate drought resistance in Arabidopsis thaliana by modulating abscisic acid responsiveness, anthocyanin accumulation, stomatal closure, cell membrane integrity and cuticle formation. Here, we aim to identify genes specifically or commonly regulated by D14 and KAI2 under water scarcity, using comparative analysis of the transcriptome data of the A. thaliana d14-1 and kai2-2 mutants under dehydration conditions. In comparison with wild-type, under dehydration conditions, the expression levels of genes related to photosynthesis and the metabolism of glucosinolates and trehalose were significantly changed in both d14-1 and kai2-2 mutant plants, whereas the transcript levels of genes related to the metabolism of cytokinins and brassinosteroids were significantly altered in the d14-1 mutant plants only. These results suggest that cytokinin and brassinosteroid metabolism might be specifically regulated by the D14 pathway, whereas photosynthesis and metabolism of glucosinolates and trehalose are potentially regulated by both D14 and KAI2 pathways in plant response to water scarcity.
