Mechanism of ubiquitin ligation and lysine prioritization by a HECT E3

TL;DR: This work reports that the second step of ligation is mediated by a distinct catalytic architecture established by both the HECT E3 and its covalently linked ubiquitin, and shows how HECT-specific ubiquit in interactions driving multiple reactions are repurposed by a major E3 conformational change to promote ligation.

TL;DR: The findings suggest that the combinatorial readout of the H3R2/K14ac by PHD12 might represent an important epigenetic regulatory mechanism that governs transcription and also provide a clue of cross-talk between the MOZ complex and histone H3 modifications.
