Abstract: An optical heterodyne detection system includes a tunable optical pre-selector that is adjusted to track the frequency of a swept local oscillator signal. The tunable optical pre-selector is adjusted in response to a measure of the frequency of the swept local oscillator signal and in response to a measure of a portion of the swept local oscillator signal after the portion of the swept local oscillator signal has optically interacted with the optical pre-selector. In an embodiment, at least some portion of the swept local oscillator signal is modulated before it interacts with the optical pre-selector. In an embodiment, the portion of the swept local oscillator signal that interacts with the pre-selector is detected and used in a feedback control circuit to generate a control signal which causes the error between the center frequency of the pre-selector and the frequency of the swept local oscillator signal to be small.

Abstract: The present invention provides a method for making a large nucleic acid having a defined sequence in vivo. The method combines recombineering techniques with a CRISPR/Cas system to permit multiple insertions of defined sequences into a target nucleic acid at one time, double stranded cleavage of target nucleic acids in which the defined sequences were not successfully inserted, and selection of successful recombinant cells. The method further includes repeating the process one or more times, using a successful recombinant from one round as the host cell for the next round.

Abstract: An ICP-MS instrument fitted with an octopole reaction system (ORS) was used to directly measure the inorganic contents of several biofuel materials. Following sample preparation by simple dilution in kerosene, the biofuels were analysed directly. The ORS effectively removed matrix- and plasma-based spectral interferences to enable measurement of all important analytes, including sulfur, at levels below those possible by ICP-OES. A range of commonly produced biofuels was analysed, and spike recovery and long-term stability data was acquired. Suitably configured ICP-MS has been shown to be a fast and very sensitive technique for the elemental analysis of biofuels.

Abstract: A quantitation method for 18 aroma compounds reported to contribute to "oxidative" flavor in wines was developed. The method allows quantitation of the (E)-2-alkenals ((E)-2-hexenal, (E)-2-heptenal, (E)-2-octenal, and (E)-2-nonenal), various Strecker aldehydes (methional, 2-phenylacetaldehyde, 3-methylbutanal, and 2-methylpropanal), aldehydes (furfural, 5-methylfurfural, hexanal, and benzaldehyde), furans (sotolon, furaneol, and homofuraneol), as well as alcohols (methionol, eugenol, and maltol) in the same analysis. The aldehydes were determined after derivatization directly in the wine with O-(2,3,4,5,6-pentafluorobenzyl)hydroxylamine hydrochloride; the formed oximes along with the underivatized aroma compounds were isolated by solid-phase extraction and analyzed by means of GC-MS/MS. The method was used to investigate the effect of different closures (synthetic closures, natural corks, and screw cap) on the formation of oxidation-related compounds in 14 year old white wine. Results showed a significant increase in the concentration of some of the monitored compounds in the wine, particularly methional, 2-phenylacetaldehyde, and 3-methylbutanal.

Abstract: A system and method for measuring flow rate within a fluid-bearing passageway include introducing heat fluctuations into the flow and then non-invasively monitoring the effects of the heat fluctuations propagating to or from one or more interrogation regions In one embodiment, the non-invasive monitoring detects fluctuations in the refractive index of the flowing fluid as a result of variations in the temperature of the fluid In another embodiment, electrical conductivity of the fluid is monitored The heat fluctuations may be introduced using an optical heat generator, such as an infrared laser, or may be introduced using an electrical member, such as a heater coil Determining the refractive index along the interrogation region may be achieved by monitoring characteristics on an interference pattern, but other optical arrangements may be utilized
