Abstract: The interaction of new dinuclear copper(II) complex 1; [Cu2(glygly)2(ppz)(H2O)4]·2H2O, derived from dipeptide (glycyl glycine) and piperazine as a metallopeptide drug with human serum albumin (HSA) was examined by means of fluorescence spectroscopy which revealed that complex 1 has a strong ability to quench the intrinsic fluorescence of HSA through a static quenching procedure. The alterations of HSA secondary structure in the presence of complex 1 were confirmed by UV-visible, FT-IR, CD and 3D fluorescence spectroscopy. The binding constants (K), and binding site number (n), corresponding thermodynamic parameters ΔG, ΔH and ΔS at different temperatures were calculated. The molecular docking technique was utilized to ascertain the mechanism and mode of action towards the molecular target HSA indicating that complex 1 was located at the entrance of site I by electrostatic and hydrophobic forces, consistent with the corresponding experimental results. Complex 1 shows efficient photo-induced HSA cleavage activity, indicating the involvement of hydroxyl radicals as the reactive species. Furthermore, the cytotoxicity of 1 was examined on a panel of human tumor cell lines of different histological origins showing significant GI50 values specifically towards MIAPACA2, A498 and A549 tumor cell lines. These results complement previous biological studies of new specific target metallopeptides, providing additional information about possibilities of their transport and disposition in blood plasma.

Abstract: Ascorbate (AsA)-glutathione (GSH) cycle metabolism is an essential mechanism for the resistance of plants under stress conditions In a greenhouse pot experiment, the influence of cadmium (Cd) (25, 50, and 100 mg/kg soil) on plant dry weight and leaf area, photosynthetic parameters (net photosynthetic rate (PN) and chlorophyll (Chl) content) and oxidative stress, and the possible protective role of AsA-GSH cycle metabolism was studied in two mung bean (Vigna radiata (L) Wilczek) cvs Pusa 9531 (Cd-tolerant) and PS 16 (Cd-susceptible) at 30 days after sowing The contents of thiobarbituric acid-reactive substances (TBARS), H2O2, and the leakage of ions were the highest at 100 mg Cd/kg soil, and the effect was more pronounced in cv PS 16 than in cv Pusa 9531 This was concomitant with the strongest decreases in PN, plant dry weight, and leaf area The changes in the AsA-GSH redox state and an increase in AsA-GSH-regenerating enzymes, such as glutathione reductase, monodehydroascorbate reductase, dehydroascorbate reductase, and other antioxidant enzymes, such as superoxide dismutase and ascorbate peroxidase, strongly supported over-utilization of AsA-GSH in Cd-treated plants However, the oxidative stress caused by Cd toxicity was partially overcome by AsA-GSH-based detoxification mechanism in the two genotypes studied because an increases in lipid peroxidation (TBARS, ion leakage) and H2O2 content were accompanied by a corresponding decrease in reduced AsA and GSH pools Thus, changes in AsA-GSH pools and the coordination between AsA-GSH-regenerating enzymes and other enzymatic antioxidants of the leaves suggest their relevance to the defense against Cd stress

Abstract: The aim of this study was to evaluate the effect of selected herbicides (quizalafop-p-ethyl, clodinafop, metribuzin and glyphosate) at one, two and three times the recommended field rates on the plant-growth-promoting (PGP) traits of Pseudomonas putida strain PS9 isolated from the mustard rhizosphere. P. putida strain PS9 was selected for its high herbicide-tolerance and its production of substantial amounts of indole acetic acid, siderophores (salicylic acid and 2,3 dihydroxy benzoic acid) and exo-polysaccharides. It also exhibits HCN and ammonia releasing ability. A progressive, herbicide-concentration-dependent, decline in PGP properties (except exo-polysaccharide production) of P. putida strain PS9 was observed when the strain was grown in the presence of increasing concentrations of each herbicide. Generally, among the selected herbicides, the maximum toxicity to PGP traits of P. putida strain PS9 was shown by quizalafop-p-ethyl at three times the recommended dose. This study concludes that herbicides should be screened before field application for their degree of toxicity to PGP traits of beneficial microorganisms.
