Abstract: Evidence is presented for the homogeneity of a preparation of hypothalamic TRF of ovine origin and its molecular structure has been established as 2-pyrrolidone-5-carboxylyl-histidyl-proline amide. Synthetic L-2-pyrrolidone-5-carboxylyl-L-histidyl-L-proline amide has physicochemical and biological characteristics which are quantitatively and qualitatively indistinguishable from those of the natural substance isolated in this laboratory.

Abstract: WE have observed lymphocytotoxic antibodies in sera of patients with connective tissue disorders. The antibodies are cytotoxic to lymphocytes from unrelated random donors using the microcytotoxicity technique1,2. The initial studies were done on 161 coded stored sera submitted to the clinical laboratory for routine immuno-electrophoresis, and confirmatory studies were done on forty-seven coded sera selected from patients followed in the clinic for connective tissue disorders. Results of testing of both series of sera are summarized in Table 1 and show that 74 per cent of all sera tested from patients with systemic lupus erythematosus and 14 per cent of sera from patients with rheumatoid arthritis contained lymphocytotoxic reactivity. Appropriate controls indicated that the cytotoxic effect of lupus sera was dependent on complement; fractionation of one lupus serum by anion exchange chromatography indicated that the cytotoxic factor resides among the IgG globulins. Because cytotoxic antibodies are not present in sera of normal persons except in some cases of immunization by multiple transfusions3,4, pregnancy5,6 or organ transplantation7,8, the results suggest that lymphocytotoxic antibodies develop during the course of systemic lupus erythematosus.

Abstract: Intravenous administration of synthetic TRF in doses of 250–750 μg stimulates a rise in plasma HTSH levels in normal individuals. No rise in plasma HTSH occurs following 500–750 μg of TRF in subjects on 0.3 mg of Lthyroxine or in patients with panhypopituitarism. One of two euthyroid patients with a sella tumor appeared to respond suboptimally to TRF. It is condluded that synthetic TRF is a convenient means for testing the integrity of the human pituitary for TSH secretion.

Abstract: THE Pictet–Spengler reaction is a special example of the Mannich reaction involving condensation of β-aryl-ethylamines with carbonyl compounds1. In the case of phenylethylamines the resulting products would be 1,2,3,4-tetrahydroisoquinolines, which are particularly interesting because they could be formed from endogenous biogenic amines. The synthesis of simple tetra-hydroisoquinolines in physiological conditions which occur in plants was described in 19342. The facility of this chemical reaction between dopamine and acetaldehyde in conditions which exist in both plants3 and animals4,5 has been confirmed. Because acetaldehyde is the primary metabolite of ethanol, it has been suggested that tetra-hydroisoquinolines are formed in mammals after the ingestion of alcohol4–6. Indeed, this reaction forms the basis for the histochemical localization of catecholamines in nerve tissue for which formaldehyde vapour is used as the carbonyl agent. Reaction products of this type have been demonstrated in animals after administration of methanol7.

Abstract: The family, reported on, have a syndrome of prolonged Q-T interval, potentially fatal episodes of ventricular arrhythmias, and normal hearing. The direct familial transmission demonstrated in this pedigree is consistent with an autosomal dominant mode of inheritance. To protect other members of the family from the consequences of ventricular arrhythmia experienced by the irreversibly brain-damaged index patient, a series of physiologic investigations was undertaken on this patient in a search for a suitable prophylactic medical regimen. The attacks of ventricular arrhythmia were found to be precipitated by increase in systemic pressure, sinus tachycardia, and extrasystoles occurring in the supernormal phase of repolarization (R in T syndrome). Propranolol was demonstrated to be an effective agent in preventing the ventricular arrhythmia in the index case and in other family members. Abstract: The seroepidemiology of the defective adenovirus-associated satellite viruses was studied by use of a neutralization test that has been shown to be much more sensitive than the complement-fixation (CF) test. Less than 25% of the subjects in any age group were positive for any type of satellite virus CF antibody. The neutralization test revealed serological evidence of satellite infection in almost 80% of the subjects in some age groups. Studies of geometric mean antibody titers by neutralization showed that antibody against type 1 and the type 2-3 complex reached peak levels at 2 to 3 years of age; antibody against types 2 and 3 reached mean titers of 500 to 800, whereas mean antibody levels of only 200 were found for type 1. Type 4 antibodies were rarely noted in human sera. In a companion study in monkeys, monospecific responses to types 1 and 4 were found only in rhesus and green monkey sera, respectively. These data suggest that the type 2-3 complex may be the only naturally occurring satellite serotype which infects man and that antibodies noted against types 1 and 4 may be the result of cross-reactions. No satellite viruses were isolated from throat or rectal swabs of 100 children between 6 months and 3 years of age. However, the patterns of adenovirus and enterovirus isolations from these specimens were typical of previous studies in this laboratory.

Abstract: Forty patients with unequivocal Turner stigmata and cardiovascular disease were studied. Every patient had complete documentation of both his cardiovascular disease and chromosomal constitution by physical examination, electrocardiogram, roentgenogram and cardiac catheterization, chromosome preparation and buccal mucosal smear. Two clusters of cardiovascular lesions were observed. Coarctation of the aorta occurred in 11 of 16 patients with XO Turner syndrome but not in any of the patients with Turner phenotype or Turner mosaic constitution. Pulmonic stenosis was present in 21 of 24 patients with Turner phenotype and Turner mosaic constitution but was not encountered in those with XO Turner syndrome.

Abstract: In a double-blind comparison of the therapeutic effect of amantadine in a natural outbreak of Hong Kong influenza in January 1969 near Houston, Texas, the decrease in titers of virus in throat swabs from 12 treated patients during the first 10 hr of treatment was appreciably greater than similar titers of virus from 16 untreated patients (P = 0.06). The titer of shed virus decreased significantly more rapidly (P < 0.05) among amantadine-treated patients ill more than 48 hr before treatment than among five control patients who had been ill more than 48 hr. Cough, sore throat, and nasal obstruction cleared more rapidly in treated patients (P < 0.05), and decline of fever in 6 treated patients sick for less than 48 hr before treatment was more rapid than among 13 untreated patients who had been ill for less than 48 hr. These findings are considered to be consistent with a limited therapeutic effect of the drug.
