Abstract: We present the angular power spectrum derived from the first-year Wilkinson Microwave Anisotropy Probe (WMAP) sky maps We study a variety of power spectrum estimation methods and data combinations and demonstrate that the results are robust The data are modestly contaminated by diffuse Galactic foreground emission, but we show that a simple Galactic template model is sufficient to remove the signal Point sources produce a modest contamination in the low frequency data After masking approximately 700 known bright sources from the maps, we estimate residual sources contribute approximately 3500 mu sq Kappa at 41 GHz, and approximately 130 mu sq Kappa at 94 GHz, to the power spectrum [iota(iota + 1)C(sub iota)/2pi] at iota = 1000 Systematic errors are negligible compared to the (modest) level of foreground emission Our best estimate of the power spectrum is derived from 28 cross-power spectra of statistically independent channels The final spectrum is essentially independent of the noise properties of an individual radiometer The resulting spectrum provides a definitive measurement of the CMB power spectrum, with uncertainties limited by cosmic variance, up to iota approximately 350 The spectrum clearly exhibits a first acoustic peak at iota = 220 and a second acoustic peak at iota approximately 540, and it provides strong support for adiabatic initial conditions Researchers have analyzed the CT(sup Epsilon) power spectrum, and present evidence for a relatively high optical depth, and an early period of cosmic reionization Among other things, this implies that the temperature power spectrum has been suppressed by approximately 30% on degree angular scales, due to secondary scattering

Abstract: The Trans-Omics for Precision Medicine (TOPMed) programme seeks to elucidate the genetic architecture and biology of heart, lung, blood and sleep disorders, with the ultimate goal of improving diagnosis, treatment and prevention of these diseases The initial phases of the programme focused on whole-genome sequencing of individuals with rich phenotypic data and diverse backgrounds Here we describe the TOPMed goals and design as well as the available resources and early insights obtained from the sequence data The resources include a variant browser, a genotype imputation server, and genomic and phenotypic data that are available through dbGaP (Database of Genotypes and Phenotypes)1 In the first 53,831 TOPMed samples, we detected more than 400 million single-nucleotide and insertion or deletion variants after alignment with the reference genome Additional previously undescribed variants were detected through assembly of unmapped reads and customized analysis in highly variable loci Among the more than 400 million detected variants, 97% have frequencies of less than 1% and 46% are singletons that are present in only one individual (53% among unrelated individuals) These rare variants provide insights into mutational processes and recent human evolutionary history The extensive catalogue of genetic variation in TOPMed studies provides unique opportunities for exploring the contributions of rare and noncoding sequence variants to phenotypic variation Furthermore, combining TOPMed haplotypes with modern imputation methods improves the power and reach of genome-wide association studies to include variants down to a frequency of approximately 001% The goals, resources and design of the NHLBI Trans-Omics for Precision Medicine (TOPMed) programme are described, and analyses of rare variants detected in the first 53,831 samples provide insights into mutational processes and recent human evolutionary history

Abstract: An accuracy analysis of a new class of integration algorithms for finite deformation elastoplastic constitutive relations recently proposed by the authors, is carried out in this paper. For simplicity, attention is confined to infinitesimal deformations. The integration rules under consideration fall within the category of return mapping algorithms and follow in a straightforward manner from the theory of operator splitting applied to elastoplastic constitutive relations. General rate-independent and rate-dependent behaviour, with plastic hardening or softening, associated or non-associated flow rules and nonlinear elastic response can be efficiently treated within the present framework. Isoerror maps are presented which demonstrate the good accuracy properties of the algorithm even for strain increments much larger than the characteristic strains at yielding.

Abstract: This paper proposes the use of repetitive broadcast as a way of augmenting the memory hierarchy of clients in an asymmetric communication environment. We describe a new technique called "Broadcast Disks" for structuring the broadcast in a way that provides improved performance for non-uniformly accessed data. The Broadcast Disk superimposes multiple disks spinning at different speeds on a single broadcast channel--in effect creating an arbitrarily fine-grained memory hierarchy. In addition to proposing and defining the mechanism, a main result of this work is that exploiting the potential of the broadcast structure requires a re-evaluation of basic cache management policies. We examine several "pure" cache management policies and develop and measure implementable approximations to these policies. These results and others are presented in a set of simulation studies that substantiates the basic idea and develops some of the intuitions required to design a particular broadcast program.

Abstract: Drug concentrations associated with protection from HIV-1 acquisition have not been determined. We evaluated drug concentrations among men who have sex with men in a substudy of the iPrEx trial (1). In this randomized placebo-controlled trial, daily oral doses of emtricitabine/tenofovir disoproxil fumarate were used as pre-exposure prophylaxis (PrEP) in men who have sex with men. Drug was detected less frequently in blood plasma and in viable cryopreserved peripheral blood mononuclear cells (PBMCs) in HIV-infected cases at the visit when HIV was first discovered compared with controls at the matched time point of the study (8% versus 44%; P < 0.001) and in the 90 days before that visit (11% versus 51%; P < 0.001). An intracellular concentration of the active form of tenofovir, tenofovir-diphosphate (TFV-DP), of 16 fmol per million PBMCs was associated with a 90% reduction in HIV acquisition relative to the placebo arm. Directly observed dosing in a separate study, the STRAND trial, yielded TFV-DP concentrations that, when analyzed according to the iPrEx model, corresponded to an HIV-1 risk reduction of 76% for two doses per week, 96% for four doses per week, and 99% for seven doses per week. Prophylactic benefits were observed over a range of doses and drug concentrations, suggesting ways to optimize PrEP regimens for this population.
