Abstract: 1. The effects of intracerebroventricular administration of some α- and β-adrenoceptor stimulants and antagonists on the body temperature of rabbits were investigated.
2. Noradrenaline produced a dose dependent rise in body temperature. Other catecholamines were less active.
3. The noradrenaline response was blocked by α-adrenoceptor blocking agents while β-adrenoceptor antagonists had no effect.
4. α-Methyl-noradrenaline and metaraminol had some hyperthermic effect, but significantly reduced the response of noradrenaline.
5. The possible presence of α-adrenoceptors in the central thermoregulatory mechanisms is suggested.

Abstract: SYNOPSIS. Hartmannella (Culbertson strain A-1) was found to undergo encystment (80–90% in 72 hr) on a non-nutrient agar containing 0.015 M MgCl2 and 0.02 M taurine. Encystment was completely inhibited by 1 × 10−5 M Mitomycin C, or 1 × 10−7 M cycloheximide or 1 × 10−6 M Actinomycin D. The ability of the amoebae to consume glucose increased fourfold within 24 hr incubation in this medium. The specific activities of cellulose synthetase, hexosephosphate transaminase and uridine diphosphosphoglucose pyrophosphorylase were also stimulated. Dehydrogenases mediating electron transfer from pyruvate, malate, succinate, α-ketoglutarate and α-glycerophosphate to triphenyltetrazolium and from glucose-6-phosphate to nicotinamide-adenine dinucleotide phosphate were, however, repressed during this period of incubation in the encystment medium. The results suggested that, during encystment of Hartmannella A-1, there was a metabolic switchover and the enzyme machinery of the amoeba was oriented more towards biosynthesis of cyst wall constituents than towards the aerobic breakdown of carbohydrates.

Abstract: SYNOPSIS. A procedure is described for the isolation of cyst wall of Schizopyrenus russelli free of cytoplasmic material. It has 27.3% protein, 37.5% carbohydrate (of which 13.1% is cellulose), and 15.1% lipids. Sialic acid is absent. It has a relatively electrondense inner thick layer and a less electron-dense outer thin layer. The space between the 2 layers is 25 nm in younger cysts and 2.5 μ in fully mature cysts. This space is filled with a fibrillar spongy material likely to be cellulose.

Abstract: Publisher Summary This chapter presents information on carbazole alkaloids. Carbazole alkaloids represent a new and interesting variant in the large number of existing indole alkaloids that, in the past, have yielded several important drugs. There has been considerable progress in this field; currently, about 19 alkaloids, from simple compounds like 6-methoxy-3-methylcarbazolteo complex hexacyclic bases, are known. In most cases, their structures have been elucidated by the application of modern physical methods and confirmed by syntheses. This chapter discusses those members of this series having an extra carbon unit at C-3 or C-6. A rich and rewarding source of carbazole alkaloids has been the Indian curry leaf plant Murraya koenigii Spreng (Rutaceae). The roots of Glycosmis pentaphylla (Retz.) and Clausena heptaphylla have also yielded new carbazole alkaloids. The crucial information that murrayanine is a carbazole derivative was provided by zinc dust distillation-carbazole was obtained. Similarly, the Wolff–Kishner reduction product of murrayanine on zinc dust distillation yielded 3-methylcarbazole. The chapter also discusses C13-alkaloids, C18-alkaloids, C23-alkaloids, and others. The crude extract of M. koenigii has shown activity against Entamoeba histolytica, while girinimbine is reported to be active against Nocardia asteroides fungi.

Abstract: SYNOPSIS. Changes in macromolecular content and certain enzyme levels have been studied during excystment of Schizopyrenus russelli. Protein, RNA and DNA increase with the onset of excystment, the increase being more marked in the first 2 hr than later. Cellulose content is considerably decreased as the trophozoites start emerging from the cysts. There is no significant change in glycogen content. Enzyme activities of glutamic oxalacetic transaminase and ribonuclease increase while those of aldolase and isocitric dehydrogenase decrease as excystment proceeds.

Abstract: Intracerebral injection of vegetative myxamoebae of strains of Polysphondylium pallidum caused 80 to 100% mortality in mice, whereas a 30 to 50% mortality occurred with strains of Dictyostelium giganteum and D. mucoroides. Death is attributable to an unidentified toxic factor. Histopathologically a giant cell formation was seen in the spleen of mice. This cell was different from Langerhan's or foreign body giant cell. Rats injected intracerebrally behaved similarly. Intranasal instillation of myxamoebae in guinea pigs caused transient pneumonia. Myxamoebae do not multiply in the brain tissues of mice or rats, or in the lungs of guinea pigs.Intranasal instillation of vegetative myxamoebae is less pathogenic than free living amoebae like Hartmanella culbertsoni, H. rhysodes, Naegleria sp. (Culbertson strain HB-1), or Naegleria aerobia.
