Abstract: The concept of disposal of nuclear fuel waste in crystalline rock requires the effects of microbial action to be investigated. The Underground Research Laboratory excavated in a pluton of the Canadian Shield provides a unique opportunity to study these effects. Three biofilms kept moist by seepage through fractures in granitic rock faces of the Underground Research Laboratory have been examined. The biofilms contained a variety of gram-negative and gram-positive morphotypes held together by an organic extracellular matrix. Nutrient levels in the groundwater were low, but energy-dispersive X-ray spectroscopy has shown biogeochemical immobilization of several elements in the biofilms; some of these elements were concentrated from extremely dilute environmental concentrations, and all elements were chemically complexed together to form amorphous or crystalline fine-grained minerals. These were seen by transmission electron microscopy to be both associated with the surfaces of the bacteria and scattered throughout the extracellular matrix, suggesting their de novo development through bacterial surface-mediated nucleation. The biofilm consortia are thought to concentrate elements both by passive sorption and by energy metabolism. By Mossbauer spectroscopy and X-ray diffraction, one of the biofilms showed that iron was both oxidized and precipitated as ferrihydrite or hematite aerobically and reduced and precipitated as siderite anaerobically. We believe that some Archean banded-iron formations could have been formed in a manner similar to this, as it would explain the deposition of hematite and siderite in close proximity. This biogeochemical development of minerals may also affect the transport of material in waste disposal sites.

Abstract: Algorithms for the subtraction of backgrounds from γ-γ matrices and higher-fold data sets, obtained from in-beam HPGe coincidence experiments with heavy-ion-induced nuclear reactions, are described. The backgrounds are parameterized as the cross-products of lower-dimensional projections of the data and a one-dimensional background spectrum. A novel method of correcting for a mixture of different reaction channels in the complete data set, by making use of one or more gates on background channels in the energy region of the “E2 bump”, is presented. In many cases, this new method provides a significantly better description of the background.

Abstract: Using coarse grained models of heterogeneous vesicles we
demonstrate the potential for small-angle neutron scattering
(SANS) to detect and distinguish between two different categories
of lateral segregation: 1) unilamellar vesicles (ULV) containing a
single domain and 2) the formation of several small domains or
“clusters” (~10 nm in radius) on a ULV. Exploiting the
unique sensitivity of neutron scattering to differences between
hydrogen and deuterium, we show that the liquid ordered (lo)
DPPC-rich phase can be selectively labeled using chain deuterated
dipalymitoyl phosphatidylcholine (dDPPC), which greatly
facilitates the use of SANS to detect membrane domains. SANS
experiments are then performed in order to detect and
characterize, on nanometer length scales, lateral heterogeneities,
or so-called “rafts”, in ~30 nm radius low polydispersity
ULV made up of ternary mixtures of phospholipids and cholesterol.
For 1:1:1 DOPC:DPPC:cholesterol (DDC) ULV we find evidence for the
formation of lateral heterogeneities on cooling below 30 °C. These heterogeneities do not appear when DOPC is
replaced by SOPC. Fits to the experimental data using coarse
grained models show that, at room temperature, DDC ULV each
exhibit approximately 30 domains with average radii of ~10 nm.
