Abstract: The effect of bile on the development of 1,2-dimethylhydrazine (DMH)-induced colon cancer was studied in male Wistar rats Experimental operative models were created, in which in Group 1, the half intestinal tract, the ileum, and the right side of colon were released from bile In Group 2, both sides of the colon contained bile The sham operated animals formed Group 3 These techniques changed the concentration of bile acid in different parts of the colon, and the daily total fecal bile acid excretion as well After DMH treatment, the relationship between these changes of bile acid level and the development of colon cancer was studied Significantly more tumors than in the control group were found if the daily total bile acid level and the bile acid concentration in the left side of the colon were increased Our findings show an unambiguous connection between the fecal bile acid level and the incidence of DMH-induced colon cancer

Abstract: Immobilized pH gradients (IPG), introduced as early as 1982, have been plagued by several problems that have hampered their use over the years. We review here recent technical advances that have led to the solution of these problems and to the development of a successful and trouble-free IPG technique. In particular, it was discovered that the buffers used (acrylamido weak acids and bases) were prone to two major degradation pathways, hydrolysis at the amido bond and autopolymerization. A new generation of these buffers is described, in which both degradation routes are completely inhibited

Abstract: Between 1983 and 1988 we observed altogether 222 cases of neonatal septicemia and/or meningitis in our Department of Neonatology. The incidence was 8.46 per 1,000 liveborn infants. The case fatality rate amounted to 45.9%. The most frequently isolated causative agents were Escherichia coli (23.4%) followed by group B Streptococci (16.7%), Staphylococcus aureus (9.9%), Klebsiella pneumoniae species (8.8%), Serratia marcescens (7.9%), Pseudomonas aeruginosa and coagulase-negative Staphylococci each 5.9%. The report includes information about serotypes of Escherichia coli, group B Streptococci and plasmid patterns of Serratia marcescens. The latter was responsible for an outbreak of septicemia and meningitis with high mortality. The changing infection pattern reflects changes in the newborn population, especially in the patient structure of the neonatal intensive care unit, changes in the antibiotic policy and organizational problems.

Abstract: The present paper gives a review of German studies concerning the AMP and AMDP system that have been published since 1983. The test manual to the AMDP-system, published 1983, is complemented herewith. The following topics are discussed: Syndromes/scales on the basis of psychological and somatic outcomes, studies on reliability (especially interrater reliability); capability to differentiate diagnoses; comparison with other assessment instruments, especially biological parameters; results of several diagnoses (schizophrenia, affective disorders, depression etc.); range of application (psychopharmacological studies, diagnostic questions, etc.). The review points out that the AMDP system covers a very wide and elaborate range. Some topics are pointed out that would merit additional study.

Abstract: Two inherited cases of osteomesopyknosis are reported. This is an autosomal dominant osteosclerosis described by Maroteaux in 1980; its radiological abnormalities consist of increased density of the vertebral plates, pelvis, and sometimes of the upper part of the femur. Osteomesopyknosis, usually discovered incidentally on radiographic examination, is a mild form of familial osteosclerosis and must be distinguished from osteopetrosis which carries a worse prognosis.

Abstract: Elemental concentrations in different compartments of cryosections of isolated rat liver cells cryotransferred and freeze-dried were compared with those obtained after storage under vacuum for 12 or 60 h and after exposure to room air for 2 min. Poorer image contrast and segregation artefacts are frequently found in air-exposed sections, together with a slight but significant decrease of the K concentration in the cytoplasm and an increase of the S concentration in the liver cell nuclei and the extracellular medium. Extreme distortions of both ultrastructure and elemental distributions are observed if the sections are even slightly colder than the surrounding atmosphere. While storage of frozen-dried cryosections under vacuum for less than 12 h does not lead to alterations in the sections, gross changes are found both in morphology and elemental distribution in sections stored under vacuum for about 60 h. Long-time vacuum storage of frozen-dried cryosections is, therefore, not recommended.

Abstract: Ion concentrations in isolated lymphocyte nuclei subjected to KCl or MgCl2 media of varying ionic strength were measured by X-ray microanalysis. Values were corrected for the contribution of free ions by estimating the volume fraction of the water space morphometrically. The amount of bound cations and Cl was constant and independent of the widely varying free ion concentration. It is concluded that the mechanism of binding is counterion condensation but with limited cooperativity. In contrast to classical counterion condensation theory, the binding of ions occurs at oppositely charged clusters at the surface of the chromatin. Therefore, both cations and anions are bound and binding cannot be completely delocalized. The bound ions stabilize the basic chromatin fibre but are not involved in the regulation of the transition between the condensed and decondensed state. Using earlier data, we estimated the concentration of free cations in rat liver nuclei under in-vivo conditions to be in the order of about 80 mM.

Abstract: Sarcocystis gigantea extract (SGE) was separated by affinity chromatography into one lectin-containing fraction (SGL) that was mitogenic to mononuclear cells (MNC) and another that lacked this lectin activity. The SGL-depleted Sarcocystis extract (SGTF) contained the so-called Sarcotoxin, inducing only a slight increase in MNC proliferation. Furthermore, preincubation of MNC with SGTF for 60 min suppressed the mitogenic capacity of SGL by 60%-90%. The results presented indicate that SGTF interacts with human MNC differently than SGL, particularly by interfering with the mitogenic lectin. These findings suggest that SGL and SGTF may be involved in different immunomechanisms induced by the parasite.

Abstract: Ahnlich der Allotransplantatrejektion findet die autoaggressive Zerstorung der pankreatischen B-Zelle, die zum insulinabhangigen Diabetes fuhrt, unter Betei ligung mononuklearer lymphatischer Zellen statt. Tierexperimentell wurden unter Nutzung der BB-Ratte, die den insulinabhangigen menschlichen Diabetes weitgehend simuliert, eine Reihe von Beweisen fur die Beteiligung zellularer Effektorpopulationen erarbeitet, wie die erfolgreiche Diabetespravention durch Behandlung pradiabetischer BB-Ratten mit Antilymphozytenserum, Bestrahlung, monoklonale Anti-T-Zellantikorper oder Cyclosporin A [1]. Auch die Ubertragung der Erkrankung durch Milzzellentransfer von diabetischen BB-Ratten bzw. die Verhinderung des Krankheitsausbruches durch Transfusion von T-Lymphozyten von diabetesresistenten BB-Ratten unterstreichen die dominierende Rolle zellularer Immunreaktionen, die sich morphologisch als Insulitis verifizieren last [1].

Abstract: Methodische Fortschritte, wie die Patch-clamp-Technik [4, 16, 22], der Einsatz radiomarkierter [15] und fluoreszenzoptischer [9, 24] Tracer und die Zytofluorometrie [1, 10, 14, 17, 18, 23], haben eine breite Anwendung der Transmembran potential-(TMP-)Messung an Einzelzellsuspensionen eingeleitet. Sie liefern zunehmend Hinweise fur ein universelles Prinzip fein abgestimmter Ionenkanalaktivierung [3, 4, 7] bei der transmembranalen Signalvermittlung in sogenannten nichterregbaren Zellen. Die selektive Anderung von Ionenkanalkonduktivitaten und damit des TMP der Lymphozyten ist nach neueren Befunden essentiell fur die initiale Triggerung [1, 8, 10, 16, 24], die I a-Antigenexpression [5, 18], IL 2 Sekretion [9] und zytotoxische Effektorfunktion [22]. Dabei konnen die aktivierten Zellrezeptoren selber als Ionenkanale, z. B. fur Ca+ +, fungieren oder funktioneil eng mit ihnen verknupft sein, wie es Alcover etal. [1, 2] fur den CD3-Ti-und CD 2-Komplex diskutieren. Andererseits kann die Rezeptoraktivierung sekundar, z. B. uber die Freisetzung intrazellularer Messenger aus dem Phosphatidylinosi tolstoffwechsel [5, 8, 13, 16, 19], zu Permeabilitatsanderungen fur Na + /H+ [2, 8, 13], K + [8] und Ca + + [1, 9, 16, 19] fuhren mit dem Endresultat der Erhohung des intrazellularen pH [8] und freien Ca + + [1, 5, 8, 13, 16, 19, 20, 21, 24]. In jedem Fall sind die zu registrierenden TMP-Anderungen als fruhe Membranereignisse nach funktioneller Rezeptor-Ligand-Wechselwirkung anzusehen [3, 8, 10, 16, 17, 24].
