Abstract: Abstract. This review provides a community's perspective on air quality research focusing mainly on developments over the past decade. The article provides perspectives on current and future challenges as well as research needs for selected key topics. While this paper is not an exhaustive review of all research areas in the field of air quality, we have selected key topics that we feel are important from air quality research and policy perspectives. After providing a short historical overview, this review focuses on improvements in characterizing sources and emissions of air pollution, new air quality observations and instrumentation, advances in air quality prediction and forecasting, understanding interactions of air quality with meteorology and climate, exposure and health assessment, and air quality management and policy. In conducting the review, specific objectives were (i) to address current developments that push the boundaries of air quality research forward, (ii) to highlight the emerging prominent gaps of knowledge in air quality research, and (iii) to make recommendations to guide the direction for future research within the wider community. This review also identifies areas of particular importance for air quality policy. The original concept of this review was borne at the International Conference on Air Quality 2020 (held online due to the COVID 19 restrictions during 18–26 May 2020), but the article incorporates a wider landscape of research literature within the field of air quality science. On air pollution emissions the review highlights, in particular, the need to reduce uncertainties in emissions from diffuse sources, particulate matter chemical components, shipping emissions, and the importance of considering both indoor and outdoor sources. There is a growing need to have integrated air pollution and related observations from both ground-based and remote sensing instruments, including in particular those on satellites. The research should also capitalize on the growing area of low-cost sensors, while ensuring a quality of the measurements which are regulated by guidelines. Connecting various physical scales in air quality modelling is still a continual issue, with cities being affected by air pollution gradients at local scales and by long-range transport. At the same time, one should allow for the impacts from climate change on a longer timescale. Earth system modelling offers considerable potential by providing a consistent framework for treating scales and processes, especially where there are significant feedbacks, such as those related to aerosols, chemistry, and meteorology. Assessment of exposure to air pollution should consider the impacts of both indoor and outdoor emissions, as well as application of more sophisticated, dynamic modelling approaches to predict concentrations of air pollutants in both environments. With particulate matter being one of the most important pollutants for health, research is indicating the urgent need to understand, in particular, the role of particle number and chemical components in terms of health impact, which in turn requires improved emission inventories and models for predicting high-resolution distributions of these metrics over cities. The review also examines how air pollution management needs to adapt to the above-mentioned new challenges and briefly considers the implications from the COVID-19 pandemic for air quality. Finally, we provide recommendations for air quality research and support for policy.

Abstract: Elections define representative democracies but also produce spikes in physical mobility if voters need to travel to polling places. In this paper, we examine whether large-scale, in-person elections propagate the spread of COVID-19. We exploit a natural experiment from the Czech Republic, which biannually renews mandates in one-third of Senate constituencies that rotate according to the 1995 election law. We show that in the second and third weeks after the 2020 elections (held on October 9-10), new COVID-19 infections grew significantly faster in voting compared to non-voting constituencies. A temporarily related peak in hospital admissions and essentially no changes in test positivity rates suggest that the acceleration was not merely due to increased testing. The acceleration did not occur in the population above 65, consistently with strategic risk-avoidance by older voters. Our results have implications for postal voting reforms or postponing of large-scale, in-person (electoral) events during viral outbreaks.

Abstract: Abstract In this paper I discuss how to consistently incorporate higher-order corrections to the bubble-nucleation rate at finite temperature. Doing so I examine the merits of different approaches, with the goal of reducing uncertainties for gravitational-wave calculations. To be specific, the region of applicability and accuracy of the derivative expansion is discussed. The derivative expansion is then compared to a numerical implementation of the Gelfand–Yaglom theorem. Both methods are applied to popular first-order phase transition models, like a loop-induced barrier and a SM-EFT tree-level barrier. The results of these calculations are presented in easy-to-use parametrizations that can directly be used in gravitational-wave calculations. In addition, higher-order corrections for models with multiple scalar fields, such as singlet/triplet extensions, are studied. Lastly, the main goal of this paper is to investigate the convergence and uncertainty of all calculation. Doing so I argue that current calculations for the Standard Model with a tree-level barrier are inaccurate.

Abstract: Abstract. Biogenic volatile organic compounds (BVOCs) emitted from the terrestrial vegetation into the Earth's atmosphere play an important role in atmospheric chemical processes. Gridded information of their temporal and spatial distribution is therefore needed for proper representation of the atmospheric composition by the air quality models. Here we present three newly developed high-resolution global emission inventories of the main BVOC species including isoprene, monoterpenes, sesquiterpenes, methanol, acetone and ethene. Monthly mean and monthly averaged daily profile emissions were calculated by the Model of Emission of Gases and Aerosols from Nature (MEGANv2.1) driven by meteorological reanalyses of the European Centre for Medium-Range Weather Forecasts for the period of 2000–2019. The dataset CAMS-GLOB-BIOv1.2 is based on ERA-Interim meteorology (0.5∘ × 0.5∘ horizontal spatial resolution); the datasets CAMS-GLOB-BIOv3.0 and v3.1 were calculated with ERA5 (both 0.25∘ × 0.25∘ horizontal spatial resolution). Furthermore, European isoprene emission potential data were updated using high-resolution land cover maps and detailed information of tree species composition and emission factors from the EMEP MSC-W model system. Updated isoprene emissions are included in the CAMS-GLOB-BIOv3.1 dataset. The effect of annually changing land cover on BVOC emissions is captured by the CAMS-GLOB-BIOv3.0 as it was calculated with land cover data provided by the Climate Change Initiative of the European Space Agency (ESA-CCI). The global total annual BVOC emissions averaged over the simulated period vary between the datasets from 424 to 591 Tg (C) yr−1, with isoprene emissions from 299.1 to 440.5 Tg (isoprene) yr−1. Differences between the datasets and variation in their emission estimates provide the emission uncertainty range and the main sources of uncertainty, i.e. meteorological inputs, emission potential data and land cover description. The CAMS-GLOB-BIO time series of isoprene and monoterpenes were compared to other available data. There is a general agreement in an interannual variability in the emission estimates, and the values fall within the uncertainty range. Abstract: Abstract Purpose: The successful implementation of immune checkpoint inhibitors (ICI) in the clinical management of various solid tumors has raised considerable expectations for patients with epithelial ovarian carcinoma (EOC). However, EOC is poorly responsive to ICIs due to immunologic features including limited tumor mutational burden (TMB) and poor lymphocytic infiltration. An autologous dendritic cell (DC)-based vaccine (DCVAC) has recently been shown to be safe and to significantly improve progression-free survival (PFS) in a randomized phase II clinical trial enrolling patients with EOC (SOV01, NCT02107937). Patients and Methods: We harnessed sequencing, flow cytometry, multispectral immunofluorescence microscopy, and IHC to analyze (pretreatment) tumor and (pretreatment and posttreatment) peripheral blood samples from 82 patients enrolled in SOV01, with the aim of identifying immunologic biomarkers that would improve the clinical management of patients with EOC treated with DCVAC. Results: Although higher-than-median TMB and abundant CD8+ T-cell infiltration were associated with superior clinical benefits in patients with EOC receiving standard-of-care chemotherapy, the same did not hold true in women receiving DCVAC. Conversely, superior clinical responses to DCVAC were observed in patients with lower-than-median TMB and scarce CD8+ T-cell infiltration. Such responses were accompanied by signs of improved effector functions and tumor-specific cytotoxicity in the peripheral blood. Conclusions: Our findings suggest that while patients with highly infiltrated, “hot” EOCs benefit from chemotherapy, women with “cold” EOCs may instead require DC-based vaccination to jumpstart clinically relevant anticancer immune responses.

Abstract: Background: While the gender/sex differences in neuropsychiatric symptoms in dementia population are well described, gender/sex differences in mild behavioral impairment (MBI) in dementia-free populations and the relationship to cognitive performance and to subsequent cognitive decline have not been studied. Objective: We aimed to explore gender/sex differences in the association of MBI with the level of cognitive performance and its rate of decline in a dementia-free cohort. Methods: We studied 8,181 older adults enrolled in the online PROTECT UK Study. MBI was assessed using the MBI Checklist and cognition was measured by digit span, paired associate learning, spatial working memory, and verbal reasoning. Statistical analysis was conducted using linear regression models and linear mixed-effects models. Results: Out of 8,181 individuals (median age 63 years, 73% females), 11% of females and 14% of males had MBI syndrome. Females exhibited less often symptoms of decreased motivation (45% versus 36% in males), impulse dyscontrol (40% versus 44% in males; p = 0.001) and social inappropriateness (12% versus 15%; p < 0.001), while they showed more often symptoms of emotional dysregulation (45% versus 36%; p < 0.001). The associations of MBI domains with some measures of cognitive performance and decline were stronger in males than females, with the exception of the association of emotional dysregulation with the rate of cognitive decline in verbal reasoning, which was present exclusively in females. Conclusion: MBI may influence cognition to a greater extent in males than in females. We propose that predictors and biomarkers of dementia should consider gender/sex as an effect modifier.

Abstract: In Brief We report a case of monkeypox and herpes simplex type 2 coinfection in an HIV-positive male patient who has sex with men. This case report describes a diagnostic approach for papular rash in the anal area of the male patient who has sex with men with a history of sexually transmitted disease. This is also the first documented case of monkeypox in the Czech Republic, which was confirmed after a retrospective review of swab samples from a previously hospitalized HIV-positive patient. Monkeypox is a newly emerging disease that should be considered in the differential diagnosis of genital and/or anal papular rashes in men who have sex with men, seen in coinfection with other sexually transmitted disease.

Abstract: Turquoise killifish, Nothobranchius furzeri, is a naturally short-lived fish with applications across biomedical, ecotoxicological, and evolutionary research. In this chapter, we describe the challenges for their captive breeding and discuss current approaches to mitigate them. We first present the availability of different strains of N. furzeri (and of other Nothobranchius species) for experimental work and outline their differences. We then provide updates on the existing laboratory maintenance and breeding protocols, describe the course, peculiarities, and potential pitfalls of N. furzeri embryo development, summarize the current stage of approach to diet standardization, and define major health issues associated with keeping N. furzeri and other Nothobranchius species.

Abstract: During past periods of advance, Arctic glaciers and ice sheets overrode soil, sediments, and vegetation and buried significant stores of organic matter (OM); these glaciers are now shrinking rapidly due to climate warming. Little is known about the biogeochemical processing of the OM buried beneath glacier ice which makes the processes associated with deglaciation difficult to predict. Subglacial sediments exposed at receding glacier fronts may represent a legacy of past biogeochemical processes. Here, we analyzed sediments from retreating fronts of 19 Arctic glaciers for their mineralogical and elemental composition, contents of major nutrients, OM biomarkers (aliphatic lipids and lignin-derived phenols), 14C age, and microbial community structure. We show the character of the sediments is mostly determined by local glaciation history and bedrock lithology. Most subglacial sediments offer high amounts of readily bioavailable phosphorus (i.e., loose, labile, and Fe/Al P fractions) but lack readily accessible carbon substrates. The subglacial OM originated mainly from overridden terrestrial vascular plants. The results of OM biomarker analysis and 14C dating suggest the OM substrates degrade in the subglacial environment and are reworked by the resident microbial communities. We argue the biogeochemical legacy of the perishing subglacial environments is an important determinant for the early processes of proglacial ecological succession.

Abstract: Heterogeneity of effects between economic variables has been a frequently discussed topic for many years now. However, the estimation of such scale-dependent effects has proved challenging. Here, we propose a multivariate multiscale regression approach based on the combination of detrended fluctuation analysis and detrended cross-correlation analysis, but the idea can be easily translated into other time and frequency domain frameworks. As illustrations, we pick two classic economic models – the Taylor’s rule and the money demand function for the USA and Japan – and we uncover evident scale-dependence in the individual effects not visible by the simple regression tools. Importantly, the proposed framework can be used in any discipline where studying the effects at various scales is of interest. Further applications are thus certainly at hand.

Abstract: Cancer cells hijack developmental growth mechanisms but whether tissue morphogenesis and architecture modify tumorigenesis is unknown. Here, we characterized a new mouse model of sporadic thyroid carcinogenesis based on inducible expression of BRAF carrying a Val600 Glu (V600E) point mutation (BRAFV600E) from the thyroglobulin promoter (TgCreERT2). Spontaneous activation of this Braf-mutant allele due to leaky activity of the Cre recombinase revealed that intrinsic properties of thyroid follicles determined BRAF-mutant cell fate. Papillary thyroid carcinomas developed multicentrically within a normal microenvironment. Each tumor originated from a single follicle that provided a confined space for growth of a distinct tumor phenotype. Lineage tracing revealed oligoclonal tumor development in infancy and early selection of BRAFV600E kinase inhibitor-resistant clones. Somatic mutations were few, non-recurrent and limited to advanced tumors. Female mice developed larger tumors than males, reproducing the gender difference of human thyroid cancer. These data indicate that BRAFV600E-induced tumorigenesis is spatiotemporally regulated depending on the maturity and heterogeneity of follicles. Moreover, thyroid tissue organization seems to determine whether a BRAF-mutant lineage becomes a cancerized lineage. The TgCreERT2;BrafCA/+ sporadic thyroid cancer mouse model provides a new tool to evaluate drug therapy at different stages of tumor evolution.
