Abstract: Signalling by all-trans retinoic acid is mediated through RXR-RAR retinoid receptor heterodimers, in which RXR has been considered to act as a transcriptionally silent partner However, we show here that in cultured NB4 (ref 6) human acute promyelocytic leukaemia cells treated with either an RAR-alpha-selective agonist alone, or certain RAR-alpha antagonists in combination with an RXR agonist, receptor-DNA binding is induced in vivo, resulting in expression of the target genes of retinoic acid as well as acute promyelocytic leukaemia protein (PML) relocation to nuclear bodies and differentiation before apoptosis These results indicate that RAR-alpha ligands can induce two separate events: one enables RXR-RAR-alpha heterodimers to bind to DNA in vivo and allows RXR agonists to act; the other induces transcriptional activity of RAR-alpha The availability of receptor-specific synthetic retinoids that can induce distinct receptor functions has potential in extending the therapeutic repertoire of retinoids

Abstract: The formation of the endoderm has been investigated in chimaeric embryos resulting from the combination of the lower and upper germ layers taken from chick and quail embryos at stages 2–6 of Vakaet (1962). The ability to recognize quail from chick cells made it possible to follow the fate of each germ layer during development. It appeared that the primitive hypoblast participates in the formation of the anterolateral extra-embryonic endoderm while the embryonic endoderm is formed later by migration of cells of the ectomesoblast through Hensen9s node and the primitive streak. Further interspecific combinations were carried out between ectoderm and endoderm + mesoderm from quail and chick embryos at stages 5–7 of Hamburger and Hamilton. The explants were grafted into chick embryos for several days and the intestinal structures which developed were observed. No contribution of cells from the neurectoderm to the endoderm was found. In contrast, cells coming from the neural crest colonized the intestinal structures and gave rise to the enteric ganglia. It was concluded from these observations that the enterochromaffin and endocrine cells of the gut epithelium do not originate from the neurectoderm.

Abstract: Brain macrophages are transiently present in different regions of the central nervous system during development or in the course of tissue remodelling following various types of injuries. To investigate the influence of these phagocytes on neuronal growth and survival, brain macrophages stemming from the cerebral cortex of rat embryos were added to neuronal primary cultures. A neurotoxic effect of brain macrophages was demonstrated by the reduction of the number of neurons bearing neurites within two days of contact between the two cell types. Neuronal death and phagocytosis were also directly observed in video recordings of living cultures. This toxicity involved the production by brain macrophages of reactive oxygen intermediates, as shown by the protective effect of catalase, a scavenger of H2O2. In addition, the respiratory bursts of brain macrophages were stimulated in the presence of neurons. These results suggest that brain macrophages could favour the appearance of neuroregressive events which occur either during neurogenesis or in neurodegenerative diseases, implying intracerebral recruitment of mononuclear phagocytes.
