Abstract: The principles and techniques of primary refractive-index gas thermometry (RIGT) are reviewed. Absolute primary RIGT using microwave measurements of helium-filled quasispherical resonators has been implemented at the temperatures of the triple points of neon, oxygen, argon and water, with relative standard uncertainties ranging from 9.1 × 10-6 to 3.5 × 10-5. Researchers are now also using argon-filled cylindrical microwave resonators for RIGT near ambient temperature, with relative standard uncertainties between 3.8 × 10-5 and 4.6 × 10-5, and conducting relative RIGT measurements on isobars at low temperatures. RIGT at optical frequencies is progressing, and has been used to perform a Boltzmann constant measurement at room temperature with a relative standard uncertainty of 1.2 × 10-5. Uncertainty budgets from implementations of absolute primary microwave RIGT, relative primary microwave RIGT and absolute primary optical RIGT are provided.

Abstract: In this paper I review covariance-based Partial Least Squares (PLS) methods, focusing on common features of their respective algorithms and optimization criteria. I then show how these algorithms can be ad- justed for use as optimal scaling tools. Three new PLS-type algorithms are proposed for the analysis of one, two or several blocks of variables: the Non- Metric NIPALS, the Non-Metric PLS Regression and the Non-Metric PLS Path Modeling, respectively. These algorithms extend the applicability of PLS methods to data measured on different measurement scales, as well as to variables linked by non-linear relationships.

Abstract: Microglia are the resident macrophages of the brain. Over the past decade, our understanding of the function of these cells has significantly improved. Microglia do not only play important roles in the healthy brain but are involved in almost every brain pathology. Gene expression profiling allowed to distinguish microglia from other macrophages and revealed that the full microglia signature can only be observed in vivo. Thus, animal models are irreplaceable to understand the function of these cells. One of the popular models to study microglia is the zebrafish larva. Due to their optical transparency and genetic accessibility, zebrafish larvae have been employed to understand a variety of microglia functions in the living brain. Here, we performed RNA sequencing of larval zebrafish microglia at different developmental time points: 3, 5, and 7 days post fertilization (dpf). Our analysis reveals that larval zebrafish microglia rapidly acquire the core microglia signature and many typical microglia genes are expressed from 3 dpf onwards. The majority of changes in gene expression happened between 3 and 5 dpf, suggesting that differentiation mainly takes place during these days. Furthermore, we compared the larval microglia transcriptome to published data sets of adult zebrafish microglia, mouse microglia, and human microglia. Larval microglia shared a significant number of expressed genes with their adult counterparts in zebrafish as well as with mouse and human microglia. In conclusion, our results show that larval zebrafish microglia mature rapidly and express the core microglia gene signature that seems to be conserved across species.

Abstract: Three groups of weanling female mice were fed one of two iron-deficient diets (5 and 12 mg Fe/kg diet) or a normal diet (30 mg Fe/kg diet) for 6 wk. A control pair-fed group was included. Seven mice received the 5 mg Fe/kg diet for 6 wk, then were rehabilitated using the 30 mg Fe/kg diet for 10 d. Mice fed the 5 mg Fe/kg diet were moderately iron-deficient, as shown by indices of iron status. No significant differences were observed in thymus weight or in the proportion and number of thymocyte subsets in thymuses of anemic, moderately iron-deficient and control mice. Thymus weight was decreased in pair-fed mice. No significant difference was found in lymph node subsets. In the spleen of anemic mice, the proportions and total number of Thy-1+ splenocytes, CD4-8+ and CD4+8- cells were very low compared with control (P less than 0.01) and iron-deficient (P less than 0.02) mice. The decrease was not only observed for the percentage of subsets but also for the absolute number of cell subtypes per spleen. Thy-1+ splenocyte subpopulations were normalized after rehabilitation. These quantitative modifications could explain alterations in the blastogenic response of splenic lymphocytes described by other authors.
