Abstract: Xanthophyllomyces dendrorhous (Phaffia rhodozyma) is the only yeast or fungus that synthesizes the commercially attractive carotenoid astaxanthin. For a suitable bioprocess, the wild type has to be modified for increasing biomass content. To achieve this, a two step strategy has been followed. At first, random mutagenesis was applied leading to colonies with substantially higher astaxanthin content. Then, the resulting strain was genetically engineered by targeting limiting reactions for further enhancement of astaxanthin biosynthesis. This combinatorial approach together with selection of an appropriate growth medium resulted in highest astaxanthin biomass contents reported to date for X. dendrorhous. In a fermenter culture, its maximum content was 9.7 mg/g dry weight.

Abstract: The acoustic-emission (AE) technique has been applied to study scale-damage processes during thermal cycling of a tube, preferentially between 600 and 300°C in air, oxygen, and air + 0.5% SO2. The AE measurements were accompanied by optical and electron-optical investigations on tube rings exposed to the same cycling conditions. During the first period of cycling, a scale rich in hematite is formed. It suffers compressive stresses during cooling. The result is a buckled multilayered scale with separated lamellae. The scaling rate is lower than under isothermal conditions. AE signals start after 175°C cooling. After longer exposure times, the scale contains an increasing amount of magnetite and becomes more compact. The scaling rate increases and is comparable to that under isothermal conditions. AE signals are already observed after 50°C cooling and are correlated with crack formation in the magnetite caused by tensile stresses there. The addition of SO2 to air enhances the crack-healing process due to higher Fe diffusion in FeS. The scale is more compact.

Abstract: Bond coat oxidation as well as bond coat depletion of Al are still believed to be a major degradation mechanism with respect to the lifetime of thermal barrier coating (TBC) systems. In this study the top coat lifetime is described as being limited by both bond coat depletion of Al and mechanical failure of the top coat. The empirical results are introduced by considering three spallation cases, namely, Al depletion failure, thermal fatigue failure, and thermal aging failure. Al depletion failure occurs when the Al content within the bond coat reaches a critical value. In this paper bond coat depletion of Al is modeled by considering the diffusion of Al into both the thermally grown oxide (TGO) and substrate. The diffusion model results are compared to Al concentration profiles measured with an electron beam microprobe. These measured results are from oxidized air plasma sprayed TBC systems (APS-TBC) with vacuum plasma sprayed (VPS) bond coats for exposures up to 5000 h in the temperature range of 950-1100 °C. This paper focuses on the Al depletion failure and how it relates to top coat spallation.
