Abstract: A laccase from the culture filtrate of white rot fungus Daedalea flavida MTCC-145 has been purified and characterized. The method involved concentration of the culture filtrate by ultrafiltration and an anion exchange chromatography on diethylaminoethyl (DEAE) cellulose. The sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and native polyacrylamide gel electrophoresis (native PAGE) both gave single protein bands indicating that the enzyme preparation was pure. The molecular mass of the enzyme determined from SDS-PAGE analysis was 75.0 kDa. Purification fold was 21.5 while recovery of the enzyme activity was 11.52%. Using 2,6-dimethoxyphenol, diammonium salt of 2,2'-[azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid)] and 3,5-dimethoxy-4-hydroxybenzaldehyde azine as substrates, the Km, kcat, and k
cat/K
m values of the laccase were found to be 440 µM, 6.45 s–1, 1.47 × 104 M–1 s–1; 366 µM, 6.45 s–1, 1.76 × 104 M–1 s–1; and 226 µM, 6.45 s–1, 2.85 × 104 M–1 s–1, respectively. The pH and temperature optima were 4.5 and 50°C, respectively. The enzyme was most stable at pH 5.0 when exposed for 1 h. The purified laccase has yellow color and shows no absorption band around 610 nm characteristic of blue laccases. Abstract: Mixed carboxylates and mercaptocarboxylates of mixed-valence FeIIFeIII with general formula [FeII-FeIII
2O(OOCR)3(OOCR′SH)3L3](where R = C13H27, C15H31 or C17H35; R′= CH2 or C6H4 and L = pyridine) were successfully prepared by substitution of [FeIIFeIII
2O(OAc)6(H2O)3] with straight chain carboxylic acids and mercaptocarboxylic acids. The title products have been characterized by elemental analyses, molar conductances and magnetic susceptibility measurements, i.r., electronic and Mossbauer spectral data. The i.r. spectra showed resolved bands arising from the ν
asym(OCO) and ν
sym(OCO) vibrations of the bridging carboxylate and mercaptocarboxylate ligands along with the ν
asym(Fe3O) vibrations. Mossbauer spectroscopic studies revealed two resolved quadrupole doublets due to FeII and FeIII at 80, 200 and 295 K, confirming the valence-localized type of compound. This fact has also been supported by the observed electronic spectral bands in the complexes. Magnetic susceptibility measurements showed net anti-ferromagnetic coupling related to mixed-valence pairs. A structure for the complexes has been established on the basis of physico-chemical studies.

Abstract: This paper discusses the role of information technology in business process reengineering (BPR). A conceptual model has been developed to illustrate the role of information systems in BPR and the type of information systems required to integrate functional areas in manufacturing. The managers should focus more particularly on information technology (IT) which is designed for mechanising existing processes before using it as an enabling agent of BPR. It is also a major challenge for managers to eliminate forms of work that do not add value, before using technology for mechanising it. Process simplification has been a tremendous interest for managers because information system (IS) is necessary for effective management of material flows in manufacturing. The implementation of BPR using innovative application of information technology (IT) aims at flexible, team-oriented, and cross-functionally coordinated management. A framework has been presented in the paper for the design and development of a more effective BPR system with the help of advanced IT.

Abstract: The protective potential of chelators, i.e. N-acetyl cysteine (0.6 mg /kg, intraperitoneally) and dithiothreitol (15.4 mg kg(-1) , intraperitoneally) with selenium (0.5 mg kg(-1) , pre-oral) were evaluated individually and in combination against methylmercury-induced biochemical alterations and oxidative stress consequences. Forty-two male Sprague-Dawley rats were exposed with methylmercury (1.5 mg kg(-1) , pre-oral) daily for 21 days followed by different treatments for five consecutive days. Administration of methylmercury caused significant enhancement in the release of transaminases, alkaline phosphatases and lactate dehydrogenases in serum. A significant increased was observed in lipid peroxidation level with a concomitant decreased in glutathione content after methylmercury exposure in liver, kidney and brain. Hepatic microsomal drug metabolizing enzymes (aniline hydroxylase and amidopyrine N-demethylase) of cytochrome p4502E1 showed sharp depletion after methylmercury exposure. Alterations in histological changes in liver, kidney and brain were also noted in methylmercury administered group. All treated groups showed recovery pattern, but the combined treatments with N-acetyl cysteine and dithiothreitol in combination with selenium were more effective than that with either alone treatments in recovering blood biochemical changes after methylmercury toxicity. In conclusion, the results demonstrated that combination therapy may recover all blood biochemical alterations and offer maximum protection against methylmercury-induced toxicity.

Abstract: A theoretical model for the hydration pattern and motion of ions around the triple helical DNA with mixed sequences d(GACTGGTGAC)d(GTCACCAGTC)*d(GACTGGTGAC) in solution, during MD simulation, using the particle mesh Ewald sum method, is elaborated here. The AMBER 5.0 force field has been used during the simulation in solvent. The simulation studies support a dynamically stable atmosphere around the DNA triplex in solution over the entire length of the trajectory. The results have been compared with Hoogsteen triplexes and examined in the context of the observed behaviour of hydration in crystallographic data of duplexes. The dynamical organization of counterions and water molecules around the triplex formed by mixed sequences is described here. It has been observed that cations prefer to bind between two adjoining purines of the second and the third strands. The idea of localized complexes (mobile counterions in unspecific electronegative pockets around the DNA triplex with water molecules) may have important implications for understanding the specificity of the interactions of nucleic acids with proteins and other ligands.
