Abstract: We investigated the heat shock response of the adenylate cyclase deficient mutant cr-1 (crisp) of Neurospora crassa. This strain was observed to be much more resistant to a lethal temperature of 50 °C than the wild type. This constitutive thermotolerance was absent in cr-1 conidiospores raised on cyclic AMP (cAMP, 2.5 mM) supplemented solid medium, but was partially restored when the conidiospores were germinated at 30 °C, a temperature which fails to induce thermotolerance in the wild-type strain. Two other crisp-like Neurospora mutants, cr-2 and cr-3 which, in contrast to cr-1, contain normal levels of cAMP, did not exhibit the thermotolerance phenomenon observed for cr-1. A cr-1, pe, fl (crisp-microconidial) strain also lacked the ability to tolerate a lethal heat treatment. Our results demonstrate that the adenylate cyclase deficient cr-1 mutant of Neu-rospora crassa expresses a constitutive thermotolerant phenotype as a consequence of its primary genetic defect: low levels of cAMP.

Abstract: In order to restructure the curriculum of the Nursing School in Ribeirao Preto - University of Sao Paulo, the following items were taken as indicators: 1. a survey on the expectations of nursing professionals towards the profession, functions, other members of the staff and professora of the School; 2. an appreciation of the curriculum by these professionals (former students). A hundred and fifteen nurses from five general hospitals in Ribeirao Preto were heard. The tabulation of the answers given to the questionaire applied allows us to assert that the assistence is the most emphasized function; the expectations of the individual as a student change with the practice of the profession; 60% of the nurses heard consider the contents of the subjects included in the curriculum adequate and 69% consider them unessential to the different qualifications of nursing.

Abstract: 1. Pulse labeling with [35S]-methionine, one-dimensional SDS-polyacrylamide gel electrophoresis and fluorography were used to study the pattern of protein synthesis in Neurospora crassa mycelia undergoing sexual development. 2. Contact of sexually-competent mycelium with cells of the opposite mating type elicited a rapid and transient increase in the synthesis of two predominant proteins of 58 kDa and 40 kDa localized in the cytosol fraction. 3. Marked changes in the pattern of protein synthesis were also observed in the 12,000 g particulate fraction, predominantly mitochondrial, where the synthesis of a 34 kDa polypeptide was most prominent among others. 4. Poly(A)+ RNA extracted from mycelia 2 h after sexual stimulation supported the in vitro synthesis of the 58 kDa and 40 kDa major polypeptides synthesized in vivo. 5. No differences were observed in the pattern of protein synthesis of treated cultures and controls 24 h after the sexual stimulus.

Abstract: In order to investigate whether conditioned circling interferes with recovery from turning induced by unilateral substantia nigra (SN) lesion, rats were trained to turn either ipsi- or contralateral to the lesioned side before and after the lesion was made. Two yoked groups served as controls. The number of turns contralateral to the trained side was significantly lower in relation to the pre-lesion value for the ipsilateral trained group and remained so until the end of the experiment. A partial recovery was observed on the 19th post-lesion day for the contralateral trained group. The results are discussed in terms of additive effects from training and the lesion symptoms.

Abstract: 1. NAD(P)ase activity was stimulated when 1% sorbose was present in the culture medium of A. nidulans, and this effect was partially reversed by 1% glucose. 2. The level of extracellular NAD(P)ase was more affected by sorbose in the culture medium than the intracellular enzyme and no morphological changes were obtained. 3. The sorbose effect on NAD(P)ase activity appears to be specific since two other exoenzymes tested (beta-glucosidase and alkaline protease) show normal secretion patterns. 4. These findings suggest that the sorbose effect on NAD(P)ase production may be the consequence of metabolic disorders not necessarily linked with the morphological changes induced by the ketohexose.
