Abstract: The active transport of glycolytic pyruvate across the inner mitochondrial membrane is thought to involve two mitochondrial pyruvate carrier subunits, MPC1 and MPC2, assembled as a 150 kDa heterotypic oligomer. Here, the recombinant production of human MPC through a co-expression strategy is first described; however, substantial complex formation was not observed, and predominantly individual subunits were purified. In contrast to MPC1, which co-purifies with a host chaperone, we demonstrated that MPC2 homo-oligomers promote efficient pyruvate transport into proteoliposomes. The derived functional requirements and kinetic features of MPC2 resemble those previously demonstrated for MPC in the literature. Distinctly, chemical inhibition of transport is observed only for a thiazolidinedione derivative. The autonomous transport role for MPC2 is validated in cells when the ectopic expression of human MPC2 in yeast lacking endogenous MPC stimulated growth and increased oxygen consumption. Multiple oligomeric species of MPC2 across mitochondrial isolates, purified protein and artificial lipid bilayers suggest functional high-order complexes. Significant changes in the secondary structure content of MPC2, as probed by synchrotron radiation circular dichroism, further supports the interaction between the protein and ligands. Our results provide the initial framework for the independent role of MPC2 in homeostasis and diseases related to dysregulated pyruvate metabolism.

Abstract: A new molecularly imprinted sol–gel polymer (MIP) was developed for microdisc solid phase extraction of parabens from human milk samples, followed by liquid chromatography analysis with a photometric detector (MISPE/LC-UV). The molecularly imprinted ormosil was synthesized by the sol–gel method using benzylparaben as a template. After template removal, selective cavities (benzylparaben) were revealed for parabens' extraction. The process of molecular recognition was based on the intermolecular interactions (hydrogen bonds) between the parabens and functional groups present in the MIP selective cavities. Fourier Transform-Infrared Spectroscopy (FTIR) spectra of the MIP and non-imprinted polymer (NIP) displayed feature bands of the 3-aminopropyltrimethoxysilane (APTMS) bonds. The use of Magic Angle Spinning-Nuclear Magnetic Resonance (MAS-NMR) attested to the presence of covalent Si–O–Si–R bonds in the structure of MIPs and NIPs. The effects of the microdisc SPE variables (pH of the sample, washing step, nature and volume of the elution solvent) on MISPE performance were evaluated. The linear range of the MISPE/LC-UV method ranges from the LOQ values (10–20 ng mL−1) to 150 ng mL−1, with a correlation coefficient higher than 0.992. The interday precision of the MISPE/LC-UV method presented a coefficient of variation lower than 13%; the accuracy ranged from 86 to 117%. According to the evaluated analytical validation parameters, the MISPE/LC-UV method is suitable for the analysis of parabens in human milk samples. Analysis of the milk samples from lactating women demonstrated the effectiveness of the proposed method.

Abstract: The objective of this study was to characterize the pattern of shell utilization by Paguristes tortugae, from infralittoral areas of Anchieta Island (Ubatuba, Sao Paulo State, Brazil) using the percentage of the different shell types occupied and the morphometric relationship between hermit crabs and occupied shells Specimens were collected monthly (January to December 1998) by SCUBA diving The animals and the shells were measured and weighed A total of 2,429 hermit crabs were captured occupying 21 species of gastropod shells The shells more occupied were Pisania auritula (3549%), Cerithium atratum (2783%), Morula nodulosa (1270%) and Leucozonia nassa (683%), respectively There was differential shell occupation between sexes: males and ovigerous females occupied in higher percentage P auritula shells while the non-ovigerous females occupied C atratum Regression analysis showed best correlation between the size of the crabs (shield length) and shell dry weight This study showed that shell occupation by P tortugae involves inter- and intraspecific competition as well as shell type availability in the field

Abstract: Objective to verify depressive symptoms and adherence to chemotherapy among women with breast cancer who are served by the Pharmacy of the Chemotherapy Center of a university hospital.METHOD: cross-sectional study with quantitative approach conducted with 112 women receiving chemotherapy. Structured interviews guided by a script addressing socio-demographic, clinical and therapeutic information, the Morisky Test, and the Beck Depression Inventory were used to collect data.RESULTS: 12.50% and 1.78% of the patients experienced "moderate" and "severe" depression, respectively, while 10.59% did not use antidepressant medication. A statistically significant association was found between levels of depression and the use of antidepressants. Lack of adherence was identified in 46.43% of the participants.CONCLUSION: these findings show the need to regularly screen for depressive symptoms and for adherence to chemotherapy treatment among women with breast cancer, in order to provide early detection and appropriate treatment centered on patients, and to improve their quality of life.
