Abstract: Electrospray ionization mass spectrometric analysis of lapachol (2-hydroxy-3-(3-methyl-2-butenyl)-1,4-naphthoquinone) was accomplished in order to elucidate the gas-phase dissociation reactions of this important biologically active natural product. The occurrence of protonated and cationized species in the positive mode and of deprotonated species in the negative mode was explored by means of collision-induced dissociation (CID) experiments. For the protonated molecule, the H2O and C4H8 losses occur by two competitive channels. For the deprotonated molecule, the even-electron rule is not conserved, and the radicalar species are eliminated by formation of distonic anions. The fragmentation mechanism for each ion was suggested on the basis of computational thermochemistry. Atomic charges, relative energies, and frontier orbitals were employed aiming at a better understanding of the gas-phase reactivity of lapachol. Potential energy surfaces for fragmentation reactions were obtained by the B3LYP/6-31+G(d,p) model. Copyright © 2010 John Wiley & Sons, Ltd.

Abstract: A sensitive, selective, and reproducible in-tube solid-phase microextraction and liquid chromatographic (in-tube SPME/LC-UV) method for determination of lidocaine and its metabolite monoethylglycinexylidide (MEGX) in human plasma has been developed, validated, and further applied to pharmacokinetic study in pregnant women with gestational diabetes mellitus (GDM) subjected to epidural anesthesia. Important factors in the optimization of in-tube SPME performance are discussed, including the draw/eject sample volume, draw/eject cycle number, draw/eject flow rate, sample pH, and influence of plasma proteins. The limits of quantification of the in-tube SPME/LC method were 50 ng/mL for both metabolite and lidocaine. The interday and intraday precision had coefficients of variation lower than 8%, and accuracy ranged from 95 to 117%. The response of the in-tube SPME/LC method for analytes was linear over a dynamic range from 50 to 5000 ng/mL, with correlation coefficients higher than 0.9976. The developed in-tube SPME/LC method was successfully used to analyze lidocaine and its metabolite in plasma samples from pregnant women with GDM subjected to epidural anesthesia for pharmacokinetic study.

Abstract: Increased drought is forecasted for tropical regions, with severe implications for the health and function of forest ecosystems. How mature forest trees will respond to water deficit is poorly known. We investigated wood anatomy and leaf traits in lowland tropical forest trees after 24 months of experimental rainfall exclusion. Sampling sun‐exposed young canopy branches from target species, we found species‐specific systematic variation in hydraulic‐related wood anatomy and leaf traits in response to drought stress. Relative to controls, drought‐affected individuals of different tree species variously exhibited trait measures consistent with increasing hydraulic safety. These included narrower or less vessels, reduced vessel groupings, lower theoretical water conductivities, less water storage tissue and more abundant fiber in their wood, and more occluded vessels. Drought‐affected individuals also had lower leaf to twig dry mass ratios, thinner leaves, and more negative pre‐dawn or mid‐day leaf water potentials. Future studies examining both wood and leaf hydraulic traits should improve the representation of plant hydraulics within terrestrial ecosystem and biosphere models, and help fine‐tune predictions of how future climate changes will affect tropical forests globally.

Abstract: Conhecer a estrutura da familia, sua composicao, como os membros se organizam e interagem entre si e com o ambiente, os problemas de saude, as situacoes de risco, os padroes de vulnerabilidade, e vital para o planejamento do cuidado a saude da familia. Este estudo objetiva descrever a aplicacao dos instrumentos genograma e ecomapa em duas familias cadastradas no Nucleo de Saude da Familia IV do Centro de Saude Escola de Ribeirao Preto da Universidade de Sao Paulo, com vistas a reflexao sobre sua utilizacao no PSF. E um estudo descritivo-exploratorio, fundamentado na literatura e na experiencia das autoras quanto a utilizacao do genograma e ecomapa como um instrumento de coleta de dados em entrevistas com familias. A aplicacao destes instrumentos possibilitou informacoes sobre a familia e suas relacoes entre si e com a comunidade, constituindo-se em uma ferramenta para ordenar as informacoes sobre a familia, de forma pratica, para o cuidado da saude centrado na familia. Conclui-se que esses instrumentos podem ser adotados pela equipe de saude da familia como uma forma de abordar a familia, de modo que esta se sinta participante do processo de coleta de informacoes, bem como possibilita conhecer a familia no que tange a estrutura familiar, cultura, ciclo de vida, relacoes e inter-relacoes.
