Abstract: A series of iron(III)porphyrins containing NO2-substituents in the meso-ortho-phenyl or COOH-substituents in the meso-para-phenyl rings (FeP) have been used to catalyse hydrocarbon oxidation by iodosylbenzene. The FeP series were efficient and selective catalysts for alkene epoxidation and alkane hydroxylation. The most promising iron porphyrin, 5,10,15-tri(2-nitrophenyl)20-mono-(4-carboxyphenyl)porphyrin iron(III) chloride, Fe(TNMCPP)Cl, was covalently bound to aminopropylated silica (APS) through covalent binding between COOH groups in the FeP (after activation by reaction with SOCl2) and NH2 groups on the funcionalised silica resulting in the anchored catalyst SiNH(TNMCPP)FeCl. This system proved to be a highly efficient catalyst for alkene epoxidation. The same iron porphyrin was also supported on APS through electrostatic binding, resulting the heterogeneous catalyst SiNH3+(TNMCPP)FeCl. The iron centres are in different surroundings on the two supports as demonstrated by EPR, UV/VIS and oxidation reactions results. The SiNH3+(TNMCPP)FeCl is not a good catalyst for hydrocarbon oxidation, which can be attributed to the higher polarity of this support and the bis-axial coordination by the free NH2 groups of the support with the iron centre. Both effects are unfavourable for the interaction between the non-polar substrates and the catalyst.

Abstract: The rich fossil record of Crocodyliformes shows a much greater diversity in the past than today in terms of morphological disparity and occupation of niches. We conducted topology-based analyses seeking diversification shifts along the evolutionary history of the group. Our results support previous studies, indicating an initial radiation of the group following the Triassic/Jurassic mass extinction, here assumed to be related to the diversification of terrestrial protosuchians, marine thalattosuchians and semi-aquatic lineages within Neosuchia. During the Cretaceous, notosuchians embodied a second diversification event in terrestrial habitats and eusuchian lineages started diversifying before the end of the Mesozoic. Our results also support previous arguments for a minor impact of the Cretaceous/Palaeogene mass extinction on the evolutionary history of the group. This argument is not only based on the information from the fossil record, which shows basal groups surviving the mass extinction and the decline of other Mesozoic lineages before the event, but also by the diversification event encompassing only the alligatoroids in the earliest period after the extinction. Our results also indicate that, instead of a continuous process through time, Crocodyliformes diversification was patchy, with events restricted to specific subgroups in particular environments and time intervals.

Abstract: The search for novel thermostable xylanases for industrial use has intensified in recent years, and thermophilic fungi are a promising source of useful enzymes. The present work reports the heterologous expression and biochemical characterization of a novel thermostable xylanase (GH10) from the thermophilic fungus Malbranchea pulchella, the influence of glycosylation on its stability, and a potential application in sugarcane bagasse hydrolysis. Xylanase MpXyn10A was overexpressed in Aspergillus nidulans and was active against birchwood xylan, presenting an optimum activity at pH 5.8 and 80°C. MpXyn10A was 16% glycosylated and thermostable, preserving 85% activity after 24 hours at 65°C, and deglycosylation did not affect thermostability. Circular dichroism confirmed the high alpha-helical content consistent with the canonical GH10 family (β/α)8 barrel fold observed in molecular modeling. Primary structure analysis revealed the existence of eight cysteine residues which could be involved in four disulfide bonds, and this could explain the high thermostability of this enzyme even in the deglycosylated form. MpXyn10A showed promising results in biomass degradation, increasing the amount of reducing sugars in bagasse in natura and in three pretreated sugarcane bagasses. MpXyn10A was successfully secreted in Aspergillus nidulans, and a potential use for sugarcane bagasse biomass degradation was demonstrated.
