Abstract: Phytohormones are long time known as important components of signalling cascades in plant development and plant responses to various abiotic and biotic challenges. Quantifications of phytohormone levels in plants are typically carried out using GC or LC-MS/MS systems, due to their high sensitivity, specificity, and the fact that not much sample preparation is needed. However, mass spectrometer-based analyses are often affected by the particular sample type (different matrices), extraction procedure, and experimental setups, i.e. the chromatographic separation system and/or mass spectrometer analyser (Triple-quadrupole, Iontrap, TOF, Orbitrap). For these reasons, a validated method is required in order to enable comparison of data that are generated in different laboratories, under different experimental set-ups, and in different matrices. So far, many phytohormone quantification studies were done using either QTRAP or Triple-quadrupole mass spectrometers. None of them was performed under the regime of a fully-validated method. Therefore, we developed and established such validated method for quantification of stress-related phytohormones such as jasmonates, abscisic acid, salicylic acid, IAA, in the model plant Arabidopsis thaliana and the fruit crop Citrus sinensis, using an Iontrap mass spectrometer. All parameters recommended by FDA (US Food and Drug Administration) or EMEA (European Medicines Evaluation Agency) for validation of analytical methods were evaluated: sensitivity, selectivity, repeatability and reproducibility (accuracy and precision).

Abstract: Empirical phylogeographic studies have progressively sampled greater numbers of loci over time, in part motivated by theoretical papers showing that estimates of key demographic parameters improve as the number of loci increases. Recently, next-generation sequencing has been applied to questions about organismal history, with the promise of revolutionizing the field. However, no systematic assessment of how phylogeographic data sets have changed over time with respect to overall size and information content has been performed. Here, we quantify the changing nature of these genetic data sets over the past 20 years, focusing on papers published in Molecular Ecology. We found that the number of independent loci, the total number of alleles sampled and the total number of single nucleotide polymorphisms (SNPs) per data set has improved over time, with particularly dramatic increases within the past 5 years. Interestingly, uniparentally inherited organellar markers (e.g. animal mitochondrial and plant chloroplast DNA) continue to represent an important component of phylogeographic data. Single-species studies (cf. comparative studies) that focus on vertebrates (particularly fish and to some extent, birds) represent the gold standard of phylogeographic data collection. Based on the current trajectory seen in our survey data, forecast modelling indicates that the median number of SNPs per data set for studies published by the end of the year 2016 may approach ~20 000. This survey provides baseline information for understanding the evolution of phylogeographic data sets and underscores the fact that development of analytical methods for handling very large genetic data sets will be critical for facilitating growth of the field.

Abstract: A glassy carbon electrode (GCE) was modified with multi-walled carbon nanotubes (MWCNT) and silver nanoparticles (AgNPs) and applied to the simultaneous determination of hydroquinone (HQ), catechol (CC), bisphenol A (BPA) and phenol by using square-wave voltammetry. The MWCNTs were deposited on the GCE and the AgNPs were then electrodeposited onto the MWCNT/GCE by the application of 10 potential sweep cycles using an AgNP colloidal suspension. The modified GCE was characterized by using SEM, which confirmed the presence of the AgNPs. The electrochemical behavior of the material was evaluated by using cyclic voltammetry, and by electrochemical impedance spectroscopy that employed hexacyanoferrate as an electrochemical probe. The results were compared to the performance of the unmodified GCE. The modified electrode has a lower charge-transfer resistance and yields an increased signal. The peaks for HQ (0.30 V), CC (0.40 V), BPA (0.74 V) and phenol (0.83 V; all versus Ag/AgCl) are well separated under optimized conditions, which facilitates their simultaneous determination. The oxidation current increases linearly with the concentrations of HQ, CC, BPA and phenol. Detection limits are in the order of 1 μM for all 4 species, and the sensor is highly stable and reproducible. The electrode was successfully employed with the simultaneous determination of HQ, CC, BPA and phenol in spiked tap water samples.
