Abstract: The sera of mice and rats which are operationally tolerant to allografts of skin commonly contain factor(s) which specifically inhibit (or block) the cytotoxic effect of lymphocytes sensitized to the histocompatibility antigens of the strain of the graft donor (1, 2). This finding, and the observation that the serum-blocking factors are usually promptly lost in animals in which tolerance has been intentionally terminated by the inoculation of syngeneic lymphoid cells, have suggested that these factors may be of primary importance in the maintenance of the tolerant state (3). Relatively little is known about the nature of serum-blocking factors and their mechanism of action. A recent study has shown that blocking activity can be selectively removed from tolerant serum by absorption with allogeneic cells and subsequently eluted from the absorbing cells at low pH.
