Abstract: MicroRNA-208a (mir-208a) is essential for cardiac hypertrophy and fibrosis. Endoglin, a co-receptor of transforming growth factor-β is also essential for cardiac fibrosis. Endoglin has been shown to be a target of mir-208a in the in vitro mechanical stress model. Volume overload can lead to heart failure and cardiac fibrosis. The role of mir-208a and endoglin in volume overload heart failure is well known. We sought to investigate the mechanism of regulation of mir-208a and endoglin in volume overload-induced heart failure. Aorta-caval (AV) shunt was performed in adult Sprague-Dawley rats to induce volume overload. Heart weight and heart weight/body weight ratio significantly increased in AV shunt animals. AV shunt significantly increased left ventricular end-diastolic dimension as compared to sham group. Mir-208a was significantly induced by AV shunt from 3 to 14 days. Endoglin, myosin heavy chain-β and brain natriuretic peptide were significantly induced by AV shunt from 3 to 14 days. Overexpression of mir-208a in the sham group without AV shunt significantly increased endoglin expression similar to the AV shunt group. Antagomir-208a attenuated the endoglin expression induced by AV shunt. Pretreatment with atorvastatin also attenuated the endoglin expression induced by AV shunt. AV shunt significantly increased myocardial fibrosis as compared to sham group. Overexpression of mir-208a in the sham group significantly increased myocardial fibrosis. Antagomir-208a and atorvastatin significantly attenuated the myocardial fibrosis induced by AV shunt. In conclusion, mir-208a increased endoglin expression to induce myocardial fibrosis in volume overloaded heart failure. Treatment with atorvastatin can attenuate the myocardial fibrosis induced by volume overload through inhibition of endoglin expression.

Abstract: Traditional Chinese Medicine (TCM) has been extensively used to ameliorate diseases in Asia for over thousands of years. However, owing to a lack of formal scientific validation, the absence of information regarding the mechanisms underlying TCMs restricts their application. After oral administration, TCM herbal ingredients frequently are not directly absorbed by the host, but rather enter the intestine to be transformed by gut microbiota. The gut microbiota is a microbial community living in animal intestines, and functions to maintain host homeostasis and health. Increasing evidences indicate that TCM herbs closely affect gut microbiota composition, which is associated with the conversion of herbal components into active metabolites. These may significantly affect the therapeutic activity of TCMs. Microbiota analyses, in conjunction with modern multiomics platforms, can together identify novel functional metabolites and form the basis of future TCM research.

Abstract: This paper aims to provide a comprehensive analysis of publication patterns on computational thinking (CT) over two recent periods (period I: 2006–2012; period II: 2013–2018). Based on keyword analysis, a total of 3798 (period I) and 7175 (period II) keywords were found. Derived from the content analysis, a research typology of two-period keywords was consolidated and framed according to its attributes, including background settings, domain-specific factors, and learning outcomes. Main findings show as follows: (1) Regarding the research background, students from secondary and higher education are the main participants; and computer science, mathematics, and engineering are the major subjects. (2) As the domain-specific factors, game and peer collaboration were found to be the main pedagogies, while web-based and face-to-face learning environments were almost equally referred to in CT research settings. However, compared with traditional command-based tools, Scratch, Lego, and Python were identified as the emerging visual-based programming languages. (3) Finally, the keywords related to learning outcomes were classified based on the Bloom’s framework of three learning domains. First, knowledge and mental understanding are the main goals in the cognitive domain; motivation and attitude are the main tasks in the affective domain; and social and communication skills are the central outcomes in the training of psychomotor ability. Further discussions and research directions are provided.

Abstract: Face recognition technique has been widely applied to personal identification systems due to its satisfying performance. However, its security may be a crucial issue, since many studies have shown that face recognition systems may be vulnerable in an adversarial environment, in which an adversary can camouflage as a legitimate user in order to mislead the system. Although face liveness detection methods have been proposed to distinguish real and fake faces, they are either time-consuming, costly, or sensitive to noise and illumination. This paper proposes a face liveness detection method with flash against 2D spoofing attack. Flash not only can enhance the differentiation between legitimate and illegitimate users, but it also reduces the influence of environmental factors. Two images are taken from a subject, one with flash and another without flash. Four texture and 2D structure descriptors with low computational complexity are used to capture information of the two images in our model. Advantages of our method include low installation cost of flash and no user cooperation required. A data set of 50 subjects collected under different scenarios is used in the experiments to evaluate the proposed method. The experimental results indicate that the proposed model performs better than existing liveness detection methods in different environmental scenarios. This paper confirms that the use of flash successfully improves face liveness detection in terms of accuracy, robustness, and running time.

Abstract: The identification of acid mucopolysaccharide by the liquid chromatography/tandem mass spectrometry method (LC-MS/MS) of the predominant disaccharide units of glycosaminoglycans (GAGs) (chondroitin sulfate, CS; dermatan sulfate, DS; heparan sulfate, HS) after methanolysis is validated and applicable for mucopolysaccharidosis (MPS) type determination. A total of 76 urine samples were collected and analyzed, from nine MPS I patients, 13 MPS II patients, seven MPS III patients, eight MPS VI patients, and 39 normal controls. Urinary GAG was first precipitated by the Alcian blue method followed by a treatment of 3 N HCl methanol. The protonated species of the methylated disaccharide products were detected by using a multiple reaction monitoring experiment. Internal standards, [2H6] CS, [2H6] DS and [2H6] HS, were prepared in-house by deuteriomethanolysis of CS, DS and HS. One particular disaccharide for each GAG was selected, in which the parent ion and its daughter ion after collision were m/z 426.1 → 236.2 for DS (m/z 432 → 239 for dimers derived from [2H6] CS and [2H6] DS) and m/z 384.2 → 161.9 for HS (m/z 390.4 → 162.5 for the [2H6] HS dimer). The quantities of DS and HS were determined, which varied from one MPS type to the other. The results can be used to evaluate the severity of MPS subgroups, as well as urinary GAG amelioration at follow-up after enzyme replacement therapy (ERT). The modified LC–MS/MS method for MPS type determination is specific, sensitive, validated, accurate, and applicable for simultaneous quantifications of urinary DS and HS. This method can help to make correct diagnosis of MPS patients and evaluate the effectiveness of ERT.
