Abstract: Clonal chromosome aberrations were detected in 8 short-term cultured malignant peripheral nerve sheath tumors (MPNST). Seven had a near-triploid chromosome number and I was in the hyperhaploid-hypodiploid range. No recurrent structural rearrangements were found; the bands most frequently involved (3 tumors) were 7p11, 12p13 and 14q11. The most common numerical changes were loss of a sex chromosome (all tumors) and loss of at least 1 copy of chromosomes 8, 16 and 22 (4 tumors). Pooling our data with those on the 20 previously published MPNST with abnormal karyotypes, we found that the chromosome number has often been in the triploid range (12 tumors), with stem line variation between 34 and 270. All chromosome arms, except 22p and the Y chromosome, were involved in recombinations. The most frequently rearranged bands were 7p22 (6 tumors) and 1p21, 7p11 and 14q11 (5 tumors each). Most numerical and unbalanced structural aberrations have led to loss of genetic material, in particular from Xq26-qter (13 tumors); 11q22-qter and 13p (12 tumors); 9p22-pter, 11p13-pter, 17p and 17q11-21 (11 tumors); 1p22-32 and 1p34-pter (10 tumors) and 6q25-qter and chromosome 16 (9 tumors). © 1995 Wiley-Liss, Inc.

Abstract: The recA gene has been used as a target in screening for the presence of acinetobacters on the genospecies level and differentiation of relevant acinetobacter species from one another by PCR. Primers deduced from known recA gene sequences of Acinetobacter calcoaceticus and Neisseria gonorrhoeae allowed the amplification of DNAs from all Acinetobacter genospecies. The size of the amplified DNA fragment from all genospecies tested was approximately 435–500 bp relative to DNA size markers. The amplified products were examined further by restriction fragment length polymorphism (RFLP) analysis. Restriction analysis with only two enzymes, MboI and HinfI, enabled us to identify all known genospecies. Since this method uses conserved recA gene sequences for primers, it is expected to be applicable for the identification of most bacterial species.

Abstract: Differences in drug-melanin interactions were determined for 13 phenothiazine neuroleptics and 2 dibenzazepine thymoleptics by means of high-performace liquid chromatography. The chromatographic column was packed with a stationary phase obtained by chemical immobilization of synthetic L-dopa melanin on silica particles. For six phenothiazines the melanin-binding parameters were also determined by an ultrafiltration method. Correlation between measures of drug-melanin interaction determined chromatographically and by the standard slow-equilibrium method was significant, however moderate. The chromatographic method of assessing interactions between drugs and melanin permitted reliable and quantitatively comparable data for representative series of solutes to be readily obtained. Such data were subjected to the analysis of quantitative structure-retention relationships (QSRR). It was found that retention of the agents on the immobilized melanin column could be described by two-parameter regression equations comprising the energy of the lowest unoccupied molecular orbital and either the water-accessible surface area of a drug molecule or its hydrophobicity parameter, determined chromatographically on an immobilized artificial membrane column. The QSRR equation derived allows for the estimation of melanin binding based on the structure of a compound candidate, and thus rationalizes predictions of potential toxicity of drugs or drug candidates.

Abstract: Previous studies have shown that symptom complex in patients with mitral valve prolapse syndrome (MVPS) may result from various forms of autonomic dysfunction. However, the interaction between adrenergic activity and renin release has not been studied. The purpose of this investigation was to assess changes in plasma renin activity (PRA) and plasma aldosterone in response to prolonged orthostatic stimulation in 19 young men with MVPS when compared to age- and sex-matched 20 control subjects. Heart rate was measured, and blood samples for PRA and aldosterone were obtained in the supine position and 90 min after assuming an upright position and slow walk. With posture the heart rate increase was greater in patients with MVPS in comparison with control subjects (19 +/- 9 vs. 12 +/- 8 beats per min, p < 0.05). Also an increase of PRA during upright activity was significantly greater in patients with MVPS (control 1.0 +/- 0.5 ng angiotensin I/mlxh, MVPS 1.8 +/- 1.7 ng angiotensin I/mlxh; p < 0.05). A significant correlation (r = 0.52, p < 0.05) was found between a rise of heart rate and PRA in patients with MVPS, but not in the control subjects. During prolonged upright activity aldosterone increased significantly in control subjects (at rest 74 +/- 41 pg/ml, with posture 98 +/- 68 pg/ml, p < 0.05) but did not change in patients with MVPS (at rest 64 +/- 29 pg/ml, with posture 67 +/- 26 pg/ml, NS).(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract: Quantitative structure-(electrophoretic) retention relationships (QSRR) were determined for metal cations using ionic radius, atomic mass and ionization energy as structural parameters. The QSRR show that atomic mass, Am, can be treated as a descriptor of differences of electrophoretic retardation of inorganic cations. Am is a measure of the effective surface area of the solutes. The ionic radius of metals, Ir, accounts for the effective charge of a solute. The ionization energy, Ei, provides a correction to Ir with regard to solute charge. High predictive potency of QSRR and direct access to the required descriptors allow for the application of the approach in identification of individual components of mixtures of metals. In case of the capillary electrophoretic parameters of organic solutes molecular mass is a most significant retention descriptor identified. It can be considered a first approximation of the electrophoretic retardation factor. Further progress in QSRR in capillary electrophoresis of drugs requires reliable descriptors of effective charge of organic solutes.

Abstract: It was shown that atrial natriuretic factor (ANF)-induced increase in glomerular filtration rate (GFR) is blunted in low sodium diet. We investigated whether saralasin, as an angiotensin II receptor antagonist, or verapamil, as a calcium entry blocker, or theophylline and zaprinast, as inhibitors of cGMP-phosphodiesterase activity, could restore the effect of ANF on GFR increase in low sodium diet rats. ANF alone (5 micrograms/kg bolus then 0.5 micrograms/min/kg BW maintenance) increased diuresis and natriuresis to the same extend in low and normal sodium diet rats but had no GFR-increasing effect in low sodium diet rats. Infusion of ANF in the presence of verapamil, saralasin, theophylline or zaprinast induced a significant increase in GFR and filtration fraction (FF). Administration of verapamil or saralasin alone did not alter GFR and FF whereas theophylline or zaprinast alone resulted in moderate but significant increases in both parameters. The increase of nephrogenous cGMP excretion in response to ANF infusion in low sodium diet rats was markedly lower as compared to normal sodium diet rats (243.4 +/- 43.3 vs. 444.0 +/- 35.6 pmol/min, respectively, p < 0.01). Administration of a selective inhibitor of cGMP-phosphodiesterase activity (zaprinast) abolished the differences in ANF-stimulated nephrogenous cGMP excretion in low and normal sodium diet rats. Basal and peak ANF (0.5 microM)-stimulated cGMP formation by isolated glomeruli was significantly lower in low than normal sodium diet rats (1.4 +/- 0.1 vs. 2.9 +/- 0.2 and 7.1 +/- 0.5 vs. 12.5 +/- 1.1 pmol/mg protein, for basal and ANF-stimulated cGMP formation, respectively; both p < 0.05). Zaprinast both alone and in combination with ANF, potentiated cGMP formation by glomeruli isolated from both groups of rats. In the presence of zaprinast, there were no differences in both basal and peak ANF-stimulated cGMP formation by glomeruli isolated from low and normal sodium diet rats. cGMP-phosphodiesterase activity was the same in the medulla of both groups of rats but markedly higher in the renal cortex of low sodium diet rats as compared to normal sodium diet rats (82.6 +/- 6.0 vs. 59.8 +/- 4.3, respectively; p < 0.05). These data suggest that the lack of GFR-increasing response to ANF in low sodium diet rats is primarily due to the increase of cGMP hydrolysis in glomeruli.

Abstract: In this article the results of molecular marker p53 examinations were presented in relation to the following established breast cancer prognostic factors: age, histologic type, histologic grade, lymph node involvement, tumor size as well as estrogen a progesterone receptor status. Twenty one percent of these primary breast cancer specimens exhibited the overexpression of p53 protein. Significant associations were found between p53 overexpression and younger age, high histologic grade and low content of estrogen and progesterone receptors. Identification of p53-positive breast carcinomas potentially represents a clinically useful indicator of breast cancer aggressiveness.

Abstract: Crystals consisting of two distinct chemical entities, tautomers of each other, in exact 1∶1 ratio, have been obtained and their structure determined by X-ray analysis. The crystals of C9H11N3·C9H11N3 are monoclinic,P21/c,a=15.674(3),b=17.085(3),c=13.758(3)A, β=90.78(2)°,Z=8. There are two hydroxylamine and two aminonitrone molecules in the asymmetric unit. Hydrogen bonds connect those molecules into chiral layers. Layers of opposite chirality alternate andthe crystal is centrosymmetric as a whole. Within those layers chains of tautomers joined by very strong O−H... O and strong N−H... N bonds can be recognized. Proton transfer along those chains with simultaneous rearrangement of π-bonds within the molecules would result in interconversion of tautomers and would affect chirality of the layer.

Abstract: The secondary structure of human brain cytosolic and mitochondrial 'malic' enzymes purified to homogeneity has been investigated by Fourier-transform IR spectroscopy. The absorbance IR spectra of these two isoenzymes were slightly different, but calculated secondary-structure compositions were essentially similar (38% alpha-helix, 38-39% beta-sheet, 14% beta-turn and 9-10% random structure). These proportions were not affected by succinate, a positive effector of mitochondrial 'malic' enzyme activity. IR spectra indicate that the tertiary structures of human brain cytosolic and mitochondrial 'malic' enzymes are slightly different, and addition of succinate does not cause conformational changes to the tertiary structure of the mitochondrial enzyme. Thermal-denaturation patterns of the cytosolic and mitochondrial enzymes, obtained from spectra recorded at different temperatures in the absence or presence of Mg2+, suggest that the tertiary structure of both isoenzymes is stabilized by bivalent cations and that the cytosolic enzyme possesses a more compact tertiary structure.

Abstract: The possible association between cigarette smoking and the risk of benign breast disease (BBD) was assessed in a case-control study conducted in Gdansk, Poland, between 1990 and 1994. The study compared 160 women with newly diagnosed BBD admitted to the Gdansk Cancer Outpatients Clinic and 160 controls, women from outpatients clinics at the Medical University of Gdansk. There was no convincing evidence of an association, either positive or negative, between various indicators of smoking habit (smoking status, number of cigarettes smoked per day, duration of smoking) and the risk of BBD. Slightly lower relative risks (RRs) of BBD in ex-smokers of 10 or more cigarettes per day (RR = 0.9; 95% confidence interval, CI: 0.4-2.2), and with duration of smoking > or = 20 years (RR = 0.8; 95% CI: 0.1-3.4), were also observed in current smokers (RR = 0.8; 95% CI: 0.4-1.5), and (RR = 0.8; 95% CI: 0.1-3.4), but these findings were not statistically significant.

Abstract: The thermal decomposition of antituberculous drugs-ethambutol (base and dihydrochloride), isoniazid, ethionamid and pyrazinamid has been studied by DTA, TG and DTG techniques. General remarks have been made on their thermal destruction. The effect of sample size over the range 20–200 mg and heating rate over the range 3–15 deg·min−1 on the thermal degradation has also been investigated. Moreover, based on the Kissinger’s equation, the values of the kinetic parameters were determined.

Abstract: Natural killer (NK) cytotoxic activity during the growth of melanotic (Ma) and amelanotic (Ab) variants of Bomirski hamster melanoma, in the blood and the spleen, was examined. The melanoma variants differed in their growth rate and metastatic pattern. Ability to form conjugates by effectors with targets and cytotoxic effects were compared. As targets K562 cells and tumor cells were used. It was found that during the growth of Ma melanoma activity of NK cells was not changing but the sensitivity of tumor cells was decreasing. However, during growth of Ab melanoma both NK activity and NK sensitivity were reduced.

Abstract: Leiomyomas are common, benign tumors which may occur at any location containing smooth muscle cells (1). Most of these tumors originate in the female genital tracts, and in particular, in the uterus. Leiomyomas are relatively avascular and tightly compacted tumors of smooth muscle cells and fibroblasts. Usually they are surrounded by a pseudocapsule of alveolar tissue and have two relatively large nourishing vessels. The majority of Leiomyomas are of normal karyotype, but their cytogenic profile is strinkingly similar to lipomas (2).
