Abstract: A literature curated dataset containing 24 distinct metal oxide (MexOy) nanoparticles (NPs), including 15 physicochemical, structural and assay-related descriptors, was enriched with 62 atomistic computational descriptors and exploited to produce a robust and validated in silico model for prediction of NP cytotoxicity. The model can be used to predict the cytotoxicity (cell viability) of MexOy NPs based on the colorimetric lactate dehydrogenase (LDH) assay and the luminometric adenosine triphosphate (ATP) assay, both of which quantify irreversible cell membrane damage. Out of the 77 total descriptors used, 7 were identified as being significant for induction of cytotoxicity by MexOy NPs. These were NP core size, hydrodynamic size, assay type, exposure dose, the energy of the MexOy conduction band (EC), the coordination number of the metal atoms on the NP surface (Avg. C.N. Me atoms surface) and the average force vector surface normal component of all metal atoms (v⟂ Me atoms surface). The significance and effect of these descriptors is discussed to demonstrate their direct correlation with cytotoxicity. The produced model has been made publicly available by the Horizon 2020 (H2020) NanoSolveIT project and will be added to the project's Integrated Approach to Testing and Assessment (IATA).

Abstract: Aging-associated neurodegenerative diseases, which are characterized by progressive neuronal death and synapses loss in human brain, are rapidly growing affecting millions of people globally. Alzheimer's is the most common neurodegenerative disease and it can be caused by genetic and environmental risk factors. This review describes the amyloid-β and Tau hypotheses leading to amyloid plaques and neurofibrillary tangles, respectively which are the predominant pathways for the development of anti-Alzheimer's small molecule inhibitors. The function and structure of the druggable targets of these two pathways including β-secretase, γ-secretase, and Tau are discussed in this review article. Computer-Aided Drug Design including computational structure-based design and ligand-based design have been employed successfully to develop inhibitors for biomolecular targets involved in Alzheimer's. The application of computational molecular modeling for the discovery of small molecule inhibitors and modulators for β-secretase and γ-secretase is summarized. Examples of computational approaches employed for the development of anti-amyloid aggregation and anti-Tau phosphorylation, proteolysis and aggregation inhibitors are also reported.

Abstract: A series of nineteen amino acid analogues of amantadine (Amt) and rimantadine (Rim) were synthesized and their antiviral activity was evaluated against influenza virus A (H3N2). Among these analogues, the conjugation of rimantadine with glycine illustrated high antiviral activity combined with low cytotoxicity. Moreover, this compound presented a profoundly high stability after in vitro incubation in human plasma for 24 h. Its thermal stability was established using differential and gravimetric thermal analysis. The crystal structure of glycyl-rimantadine revealed that it crystallizes in the orthorhombic Pbca space group. The structure–activity relationship for this class of compounds was established, with CoMFA (Comparative Molecular Field Analysis) 3D-Quantitative Structure Activity Relationships (3D-QSAR) studies predicting the activities of synthetic molecules. In addition, molecular docking studies were conducted, revealing the structural requirements for the activity of the synthetic molecules.

Abstract: Introduction: Parkinson's Disease (PD) is a neurodegenerative central nervous system (CNS) disorder characterized by dopaminergic neuron degeneration with consequent reduction in striatal dopamine (DA) levels that leads to motor symptoms. Catechol-O-methyltransferase (COMT, E.C 2.1.1.6) inactivates dopamine and other substrates bearing catechol through the methylation of a hydroxyl group. COMT inhibition can block metabolism of catecholamines including DA. Since the increase in DA bioavailability is dependent on the inhibition of DA metabolism at the periphery, the development of COMT inhibitors as adjuvants to levodopa/aromatic amino acid decarboxylase (AADC) inhibitor treatment improves the clinical benefits of PD symptomatic treatment significantly.Areas covered: This review focuses on the contribution of computational studies to develop novel COMT inhibitors as therapeutics of Parkinson's disease with substantially improved efficacy.Expert opinion: The increasing use of in silico methods and the development of new chemoinformatic tools in combination with the knowledge gained from the development of different inhibitors studied both in silico, in vitro and in vivo, could help solve a number of issues related to the shortcomings of currently marketed treatments. They can also aid to open new avenues for centrally acting COMT inhibitors, and perhaps irreversible inhibitors, to be tested for PD and other neurological diseases.

Abstract: This paper examines the media representations of scandalous parliamentary talk on same-sex child fostering in the discourses of representatives of the radical-right Golden Dawn party in Greece, but also by an MP of the conservative ANEL party of the SYRIZA-ANEL joint government at the time. Through discourse- and conversation analysis of online articles and a broadcast interview, it is shown that the media framing of populist statements is negotiated. Moreover, the interview enacts a subtly achieved interactional synergy between the interviewer and the politician, thus failing to address the issues through substantive public dialogue. It is argued that the process of (re)mediating racist or homophobic talk has the potential to serve as a publicity tool creating increased visibility for right-wing populist politicians, their core ideologies and policy platforms. This creates a challenge for practitioners of journalism who must balance disparate concerns in reporting on scandalous talk.

Abstract: The actual unavailability of cutting-edge antibiotics and healthcare in general for large parts of an ever more susceptible global population combines with the fast emergence of new and/or evolving pathogens to challenge the viability of the concept of healthcare, especially in environments of inadequate or failing infrastructures. The dissemination of pathogens within or on their hosts changes the geoepidemiology and presents new diagnostic challenges, aggravated by biosecurity concerns about spontaneous or bioengineered threats, which might escalate to Global Catastrophic BioRisks. A possible solution might be shifting priorities, such as considering healthcare as a state objective, along with biosecurity, instead of a procurable commercial amenity. Instrumental is the use of improved, low logistical footprint, detection platforms, used mainly on serology and DNA analysis so as to scan the expanded scope of human, animal, and plant pathogens, which have long started transitioning among hosts. Innovative combinations of novel and existing antimicrobial approaches might provide sustainable and applicable interventional solutions, more effective than current ones.

Abstract: We suppose that a vehicle visits N ordered customers in order to collect from them two similar but not identical materials. The actual quantity and the actual type of material that each customer possesses become known only when the vehicle arrives at the customer’s location. It is assumed that the vehicle has two compartments. We name these compartments, Compartment 1 and Compartment 2. It is assumed that Compartment 1 is suitable for loading Material 1 and Compartment 2 is suitable for loading Material 2. However it is permitted to load items of Material 1 into Compartment 2 and items of Material 2 into Compartment 1. These actions cause extra costs that are due to extra labor. It is permissible for the vehicle to interrupt its route and go to the depot to unload the items of both materials. The costs for travelling from each customer to the next one and the costs for travelling from each customer to the depot are known. The objective is to find the routing strategy that minimizes the total expected cost among all possible strategies for servicing all customers. A dynamic programming algorithm is designed for the determination of the routing strategy that minimizes the total expected cost among all possible strategies. The structure of optimal routing strategy is characterized by a set of critical numbers for each customer.

We prove on assumption of the Generalized Riemann Hypothesis that each sufficiently large odd integer $N_0$ can be represented in the form $$N_0=p_1+p_2+p_3\:,$$ where the $p_i$ are of the form $p_i=[n_i^{c_0}]$, $n_i\in\mathbb{N}$, for $i=1,2$ and the decimal expansion of $p_3$ does not contain the digit $a_0$. The proof merges methods of J. Maynard from his paper on the infinitude of primes with restricted digits, results of A. Balog and J. Friedlander on Piatetski-Shapiro primes and the Hardy-Littlewood circle method in two variables. This is the first result on the ternary Goldbach problem with primes of mixed type which involves primes with missing digits.

The proposed approach can be implemented in a hybrid way using numerical and symbolical arithmetic. Numerical examples illustrating the proposed sharing schemes are also given.
