Abstract: We studied the effect of compounds that uncouple oxidative phosphorylation on membrane function in Streptoccocus faecalis, an organism which relies upon glycolysis for the generation of metabolic energy. At low concentrations (ranging from 10−7 to 10−4m), tetrachlorosalicylanilide, tetramethyldipicrylamine, carbonylcyanide m-chlorophenylhydrazone, pentachlorophenol, and dicoumarol strongly inhibited energy-dependent transport of rubidium, phosphate, and certain amino acids. However, these compounds had little effect on the generation of adenosine triphosphate via glycolysis or on its utilization for the synthesis of macromolecules. They also did not seriously inhibit uptake of those monosaccharides and amino acids which do not require concurrent metabolism. It is proposed that the uncouplers interfere with the utilization of metabolic energy for membrane transport. The uncouplers accelerated the translocation of protons across the cytoplasmic membrane. It appears that a proton-impermeable membrane is required for transport, perhaps, because a proton gradient is involved in the coupling of metabolic energy to the translocation of substrates across the membrane.

Abstract: At a concentration of 10(-6)m, nigericin and monactin inhibited growth of Streptococcus faecalis, and the inhibition was reversed by addition of excess K(+). In the presence of certain antibiotics, the cells exhibited increased permeability to certain cations; internal Rb(+) was rapidly lost by exchange with external H(+), K(+) Rb(+), and, more slowly, with Na(+) and Li(+). No effect was observed on the penetration of other small molecules. Cation exchanges induced by nigericin and monactin were metabolically passive and apparently did not involve the energy-dependent K(+) pump. When the cells were washed, the cytoplasmic membrane recovered its original impermeability to cations. By use of monactin, we prepared cells whose K(+) content had been completely replaced by other cations, and the metabolic characteristics of K(+)-depleted cells were studied. Cells containing only Na(+) glycolyzed almost as well as did normal ones and, under proper conditions, could accumulate amino acids and orthophosphate. These cells also incorporated (14)C-uracil into ribonucleic acid but incorporation of (14)C-leucine into protein was strictly dependent upon the addition of K(+). When K(+) or Rb(+) was added to sodium-loaded cells undergoing glycolysis, these ions were accumulated by stoichiometric exchange for Na(+). From concurrent measurements of the rate of glycolysis, it was calculated that one mole-pair of cations was exchanged for each mole of adenosine triphosphate produced.

Abstract: A NEW concept of regulation of sodium excretion by the mammalian kidney has evolved since the work of DeWardener et al.1,2. During extracellular fluid volume expansion, a factor or factors other than changes in glomerular nitration rate and mineralocorticoid hormone activity influence sodium diuresis. The same factor(s) appear to contribute to the natriuresis per nephron initiated by nephron reduction and observed in advancing renal disease3. In both extracellular fluid volume expansion and nephron reduction there is evidence4,5 that a humoral substance inhibits sodium reabsorption in single proximal tubules of rats. At least a part of the natriuretic activity may be humorally mediated4 and Rector et al.5,6 have termed the active agent the “natriuretic hormone”. The agent has also been called the “third factor” (glomerular filtration rate representing the first factor and mineralocorticoid hormone activity the second factor).

Abstract: A nonprotective hyperimmune state characterized by high serum titers specific for cryptococcal capsular substance can be stimulated in mice by injections of soluble cryptococcal polysaccharide coupled to bovine gamma globulin. When the nonprotective quality of the antibody became apparent, we sought to learn whether the humoral anticryptococcal polysaccharide was reactive in vivo for cryptococcal capsules after an experimental infection. For this purpose homogenates of organs from infected mice were examined by immunofluorescence techniques for detecting mouse immunoglobulins on the capsules of tissue fungi. Abstract: Electrocardiographic studies before and after treatment were performed in 30 children with lead poisoning. It was found that abnormal electrocardiograms were present in 70% of the patients on admission. After completion of therapy, the electrocardiographic findings remained abnormal in only four (13%) of the patients. The abnormalities described most likely represent myocardial damage. It is proposed that a cardiovascular evaluation should be made as a routine procedure in every child with lead poisoning.

Abstract: Concanavalin A injected into the skin of mice induced the formation of hemorrhagic Arthus-like lesions. No lesions resulted if concanavalin A was adsorbed with insoluble a2 - macroglobulin or if mice were first treated with nitrogen mustard. The ability of concanavalin A, phytohemagglutinin, or pokeweed mitogen to induce skin lesions seemed to parallel their ability to precipitate serum proteins.

Abstract: The propensity of diazoxide to induce hyperglycemia has been evident since Dollery and associates' initially recorded this, then undesirable, side-effect. Although administration of diazoxide reduces or blocks the insulin response to glucose in a wide range of hypoglycemic states, the observation that hyperglycemia could be effected without reducing insulin level^,^,^ and that hyperglycemia can be evoked in pancreatectomized animal^,^ suggests an additional, noninsulinmediated, hyperglycemic mechanism. The studies reported here were directed toward the demonstration and better definition of such an effect of diazoxide.

Abstract: This case report concerns the fourth recorded instance of renal-cell carcinoma occurring in two members of the same immediate family—a sister and brother in the geriatric age group. In the sister's case, nephrectomy was performed and the patient is alive and clinically well three years later even though operation revealed extensive involvement of the hilar structures of the kidney. The brother is being managed with chemotherapy and radiation because of widespread carcinoma.

Abstract: Of six antibiotics investigated, streptovaricin C had the most marked chemotherapeutic effect on Mycobacterium kansasii infections in mice. By the intraperitoneal route this antibiotic caused elimination of the pathogens from all organs. Kanamycin eliminated the pathogens from the lungs of all animals and from the spleens and livers of most of them. Bluensomycin also removed the pathogens from the lungs of all animals, and spectinomycin and lincomycin, from the lungs of the majority of the animals. The three latter antibiotics lowered the bacterial counts in liver and spleen. Streptovaricin C also decreased the bacterial counts in brain, spleen, and liver of mice inoculated intracerebrally with M. kansasii. In one experiment it completely eliminated this pathogen from the spleen and almost completely from the liver. The effect of streptovaricin C on the cerebral infection was more marked than that of streptovaricin complex. Respiratory and cerebral infections of mice with M. avium, serotypes I and II, were limited by streptovaricin C, and marked decreases of the bacterial counts in brain, lungs, spleen, and liver were observed.
