TL;DR: The effects of methyl jasmonate (MeJA), an elicitor of plant defense mechanisms, on the biosynthesis of diosgenin, a steroidal saponin, were investigated in six fenugreek varieties and may be considered a promising elicitor for diOSgenin production by fenUGreek plants.
Abstract: The effects of methyl jasmonate (MeJA), an elicitor of plant defense mechanisms, on the biosynthesis of diosgenin, a steroidal saponin, were investigated in six fenugreek (Trigonella foenum-graecum) varieties (Gujarat Methi-2, Kasuri-1, Kasuri-2, Pusa Early Branching, Rajasthan Methi and Maharashtra Methi-5). Treatment with 0.01% MeJA increased diosgenin levels, in 12 days old seedlings, from 0.5%-0.9% to 1.1%-1.8%. In addition, MeJA upregulated the expression of two pivotal genes of the mevalonate pathway, the metabolic route leading to diosgenin: 3-hydroxy-3-methylglutaryl-CoA reductase (HMG) and sterol-3-β-glucosyl transferase (STRL). In particular, MeJA increased the expression of HMG and STRL genes by 3.2- and 22.2-fold, respectively, in the Gujarat Methi-2 variety, and by 25.4- and 28.4-fold, respectively, in the Kasuri-2 variety. Therefore, MeJA may be considered a promising elicitor for diosgenin production by fenugreek plants.

TL;DR: The present study concludes that AQ is not influenced by gender, stream of education and family factors, rather it was seen to be influenced by the type of schools.
Abstract: The present research was conducted to understand the Adversity Quotient (AQ) of students who are going to enter into higher education. Higher secondary is the stage from where they move from school to higher education. So, the present research was conducted to study the AQ of 11th standard, English medium school students in Gandhinagar city, Gujarat with reference to various variables. The study was conducted on a sample of 461 boys and girls of 11 th standard school students from Gujarat State Board of Education (GSEB) and Central Board of Secondary Education (CBSE) schools. A self-constructed AQ scale was used to collect data from the students. The collected data was analyzed using statistical techniques like mean, SD and C.R values. Result revealed that there was no significant difference in the mean scores of AQ on the basis of gender, stream of education i.e. Commerce, Science and Arts, and various family variables like nature of the family, size of the family, qualification of parents, parents‟ working status and parents‟ occupation. However, a significant difference was found in the mean scores of AQ of students on the basis of board of School i.e. GSEB and CBSE. The present study concludes that AQ is not influenced by gender, stream of education and family factors, rather it was seen to be influenced by the type of schools. Investigator inquired and understood the AQ profile of higher secondary school students as it is the gate way to higher Education. By understanding the AQ levels of students, the faculty and staffs can be better equipped to support students as they navigate the stressors of college life.

TL;DR: D-optimal design was used to optimize the amount of components in liquid self nanoemulsifying drug delivery system and the optimized S-SNEDDS was found to be stable and its shelf life was finding to be 2.2 years.
Abstract: The solid-self nanoemulsifying drug delivery system (S-SNEDDS) of Amiodarone hydrochloride (AH) was prepared and evaluated. AH exhibits poor aqueous solubility (0.3-0.5 mg/ml) and therefore variable oral bioavailability. Capmul MCM, Cremophor RH-40 and Propylene glycol were identified as oil, surfactant and co-surfactant for preparing L-SNEDDS. D-optimal design was used to optimize the amount of components in liquid self nanoemulsifying drug delivery system (L-SNEDDS). Optimized AH-L-SNEDDS having 15.8 nm globule size and 99.5 %transmittance was then adsorbed on Neusilin US2 to form solid self nanoemulsifying drug delivery system (AH-SSNEDDS). AH loaded L-SNEDDS and S-SNEDDS were characterized for various physicochemical properties and solid state properties. In vitro dissolution, ex vivo drug release study and In vivo study were performed for pure AH, AH-LSNEDDS and AH-S-SNEDDS. Both AH loaded L-SNEDDS and S-SNEDDS showed more than 95% drug release in 20 min during drug release studies. In vivo study revealed that release of AH from S-SNEDDS was 2.26 times and LSNEDDS was 1.83 times higher than that from suspension when given to rabbits (p < 0.01). The optimized S-SNEDDS was found to be stable and its shelf life was found to be 2.2 years. S-SNEDDS could serve as a potential drug delivery system for AH.

TL;DR: In this article, a selective and robust method for determination of Cape in dried blood spots (DBS) by liquid chromatography-tandem mass spectrometry (LC-MS/MS) was described.
Abstract: Objective: Capecitabine (Cape), the first oral prodrug which belongs to the group of fluoro pyrimidines is the most frequently prescribed anticancer drug for the treatment of metastatic breast and colorectal cancers. The article describes a selective and robust method for determination of Cape in dried blood spots (DBS) by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Methods: Cape fortified DBS was punched and extracted with ethyl acetate using capecitabine-d11 as the internal standard (IS). Chromatographic separation of Cape and IS from endogenous matrix was performed on Phenomenex Gemini C18 (150 × 4.6 mm, 5mm) column under isocratic condition using acetonitrile: 2 mmol ammonium formate (pH 3.0, adjusted with 0.1 % formic acid) (80:20, v/v ) as the mobile phase. Detection and quantification were carried on a triple quadrupole mass spectrometer, using electro spray ionization technique in the positive ionization mode. Results: The method was established over a concentration range of 10-10000 ng/ml. Accuracy, precision, selectivity, recovery, matrix effect and stability of the analyte were also estimated and the results were within the acceptance criteria. Further, precise results were obtained using an optimum spot volume of 10 µl with good spot homogeneity. Blood samples with hematocrit values varying from 24 % to 45 % gave acceptable results with good accuracy and precision. Conclusion: The efficiency of dried blood spot sample preparation, short analysis time and high selectivity permits estimation of Cape in a small blood volume. The validation results suggest that the method is precise, accurate, and reproducible and can be useful in therapeutic drug monitoring of Cape.

Abstract: A simple, precise, and rapid stable isotope dilution liquid chromatography-tandem mass spectrometry method has been developed and validated for the quantification of rilpivirine, a non-nucleoside reverse transcriptase inhibitor in human plasma. Rilpivirine and its deuterated analogue, rilpivirine-d6, used as an internal standard (IS) were quantitatively extracted by liquid-liquid extraction with methyl-tert-butyl ether and diethyl ether solvent mixture from 50 μL plasma. The chromatography was achieved on Gemini C18 (150 × 4.6 mm, 5 µm) analytical column in a run time of 2.2 min. The precursor product ion transitions for rilpivirine (m/z 367.1 128.0) and IS (m/z 373.2 134.2) were monitored on a triple quadrupole mass spectrometer in the positive ionization mode. The linearity of the method was established in the concentration range of 0.5–200 ng/mL. The mean extraction recovery for rilpivirine (94.9%) and IS (99.9%) from spiked plasma samples was consistent and reproducible. The IS-normalized matrix factors for rilpivirine ranged from 0.98 to 1.02 across three quality controls. Bench top, freeze-thaw, wet extract, and long-term stability of rilpivirine was examined in spiked plasma samples. The application of the method was demonstrated by a bioequivalence study with 25 mg rilpivirine tablet formulation in 40 healthy subjects. The assay reproducibility was shown by reanalysis of 200 study samples and the % change in the concentration of repeat values from the original values was within ±15%. TL;DR: This paper analyzes the present GUI Grid - Alchemi with its components and challenges and proposes a toolkit-Scady which solves the challenges of AlChemi and provides higher performance.
Abstract: Grid computing has resulted in faster execution of applications. Day by day the requirement for execution speed of applications is increasing. To achieve this, suitable middleware is needed and to be configured requirement specific. An application can be executed faster only when there is no node failure or network failure. Moreover, at run time, if a node fails, there should be some alternative arrangement to reconfigure other node to complete the task which was supposed to be done by the failed node. Thus, a middleware is needed that can be configured according to the requirements of the application. Such a middleware would help in increasing the performance of the application. In most of the middleware available presently, dynamic configuration is not facilitated. This has become one of the reasons for the failure of adoption or implementation of Grid in many organizations. This paper analyzes the present GUI Grid - Alchemi with its components and challenges. It also proposes a toolkit-Scady which solves the challenges of Alchemi and provides higher performance. Two experiments are done and their result analysis shows the performance of the proposed toolkit.

TL;DR: The ability of ensemble techniques to improve the accuracy of basic J48 algorithm and Naïve Bayes algorithm is focused on.
Abstract: This Classification using ensemble generally combines multiple classifiers that results in the improvement in the accuracy of the classification. Experimenting with the same dataset using the single classifier provides lesser accuracy than ensemble techniques. Many researches have been carried out using the technique of combining the predictions of multiple classifiers to generate a single classifier. The produced classifiers provide more accurate results than any individual classifier. This paper focuses on the ability of ensemble techniques to improve the accuracy of basic J48 algorithm. Ensemble techniques like Bagging and Boosting improved the efficiency of the J48 classifier. Experiments have been carried out on many datasets taken from UCI repository to investigate the effects of ensemble techniques on J48 and Naïve Bayes algorithm. WEKA tool is used to measure the effectiveness of a classifier model.
