Abstract: When the bursa of Fabricius was isolated from environmental stimuli across the bursal epithelium by clipping the bursal duct on the 19th day of incubation, marked retardation was observed both in the bursal development and in the maturation of the immune reactivity. Neither necrotic changes nor involution in the bursa was, however, observed at any stage examined after clipping the bursal duct. Lymphopoiesis in the bursa and the immune reactivity of these chickens gradually increased with age, although they were constantly lower than those of controls. These results suggest that environmental stimuli across the bursal epithelium play an important role in teh amplification mechanisms of B-cell differentiation in the bursa.

Abstract: Many studies have been reported on the reaction of formaldehyde (FA) with amino acids or proteins, and FA is assumed to react with the alpha-amino group as well as some of the side chain groups. In most of these investigations a large excess of FA relative to amino acids or proteins was employed. In the present study, however, we carried out the reaction with a smaller excess of FA in order to clarify the reactivity, firstly with the alpha-amino groups, and secondly with specific side chain groups. No evidence for so-called Schiff base (-N = CH2) formation was obtained in the reaction with the alpha-amino groups, but the formation of an acid-labile N-hydroxymethyl compound as a major product was suggested by NMR and IR at greater than pH 9.2. There was no indication, however, of the presence of such a product below pH 9.2, and the amount of N-hydroxymethyl product increased in parallel with the reaction pH. The higher the reaction pH (greater than or equal to 9.2), the greater the consumption of FA, up to 2 mol/mol Ala. In addition, the larger the excess of FA, the smaller the amount of free amino group remaining (at pH 9.7). In the assignment of IR spectra, discrete absorption bands of the alpha-carboxyl group of Ala were observed, which reflected ionization states of the alpha-amino group, and these were utilized for analysis of the reaction mechanism. Furthermore, among amino acids with side chain groups, His, Trp, and Arg showed high reactivity and Asn showed moderate reactivity. The products were relatively stable and were purified and subjected to instrumental analyses. Sixteen other amino acids including Tyr and Lys did not yield stable products. The products from Arg were unique because of the non-involvement of the amino group, and were reversibly converted to the original Arg upon acid hydrolysis. The products from His, Trp and Asn all involved amino or amide nitrogen forming cyclic ring structures with methylene derived from FA. The chemical structures of these products were determined on the basis of elemental analyses, MS and NMR.

Abstract: A new sensitive, rapid and simple method for lysozyme assay is described which is based on either fluorescence polarization or fluorescence intensity using fluorescein-labeled peptidoglycan as a substrate. The peptidoglycan was obtained from Micrococcus lysodeikticus after extensive digestion with Pronase and washing with Triton X-100 followed by various solvents. Subsequently, it was labeled with fluorescein isothiocyanate (FITC) at the amino group of the peptide. When the FITC-labeled substrate was subjected to lysozyme digestion, an increase of fluorescence intensity or a decrease of fluorescence polarization value (P value) was apparent in five minutes at a lysozyme concentrations as low as 0.1 or 0.01 micrograms/ml, respectively. The effect of other hydrolytic enzymes including alpha-mannosidase, proteases and RNase on the P value was found to be negligible. The measured values represented the specificity and dose of lysozyme added. Apparent Vmax and Km values for two different lysozymes, chicken egg white and human, could be determined by this method.

Abstract: The biological properties of Epstein-Barr virus (EBV) from a Burkitt lymphoma cell line, Jijoye, were examined. the synthesis of virus capsid antigen (VCA) and early antigen (EA) in Jijoye cells was markedly enhanced by shift-down of the temperature of incubation from 37 degrees C to 33 degrees C. Cultures of Jijoye cells at 33 degrees C released a large amount of transforming EBV (10(5.2) of 50% transforming doses/ml) into the culture fluid. However, no infectious virus was produced in all cultures at 37 degrees C during the course of this study. The EBV (Jijoye EBV) from Jijoye line was found to possess only transforming activity, but not EA-inducing activity. Jijoye EBV lacks adsorbing capacity to Jijoye cells, in contrast to P3HR-I EBV which can adsorb to Jijoye cells. The Jijoye cells were highly susceptible to superinfection with P3HR-I EBV as demonstrated by the induction of EA, VCA and infectious EBV. The EBV induced by the P3HR-I EBV superinfection of Jijoye cells has also transforming activity but neither EA-inducing activity nor adsorbing capacity to Jijoye cells.

Abstract: The alteration in activities of multiple protein kinases has been studied in the endometrium of a rabbit treated with estrogen and progesterone. The administration of estrogen or progesterone to the castrated rabbit resulted in a remarkable increase of total activity in the cytosol fraction of the endometrium. The administration of estrogen caused an increase of type I adenosine-3',5'-monophosphate-dependent (cAMP-dependent) protein kinase and a slight decrease of type II cAMP-dependent protein kinase. In contrast, the treatment with progesterone after priming administration of estrogen brought about an increase of type II cAMP-dependent protein kinase and a decrease of type I cAMP-dependent protein kinase. Therefore, the activity ratio of type II to type I decreased by estrogen and increased by progesterone. The simultaneous administration of cycloheximide abolished the stimulatory effect of respective hormones on the level of each protein kinase. The activity profile of protein kinases on DEAE-cellulose column after ovulation caused by the administration of human chorionic gonadotropin to a non-castrated rabbit was similar to that of the rabbit treated with progesterone. The results presented demonstrate the specific regulation by the steroid hormones of de novo synthesis of protein kinases in the target organ.

The antibiotic has the molecular formula C27H26O9 and is the polycyclic aromatic hydrocarbon group of antibiotics. Toromycin is active against gram-positive bacteria, mycobacteria, mycoplasma, DNA viruses and some bacteriophages.

Abstract: Summary The urinary extracts from patients with aplastic anaemia and from healthy donors were investigated in vivo and in vitro for their ability to stimulate megakaryopoiesis and platelet production. There was a significantly higher concentration of thrombopoiesis stimulating factor (TSF) and megakaryocyte colony stimulating factor (MEG-CSF) in the urine from patients with aplastic anaemia than in that from healthy donors. Neuraminidase treatment did not affect the thrombopoietic activity of TSF, whereas coexisting erythropoictin (EPO) in the extract lost its activity in vivo. These findings suggest that TSF and/or MEG-CSF seems to be different from EPO and that the urine from aplastic anaemia patients would be a good source of TSF and MEG-CSF for purification and characterization.

Abstract: A fatal case of puffer poisoning in an 80-year-old man is reported. By using a bioassay with mice and extracts of crude poisons, the toxicities of the autopsy materials, fresh puffer ovaries, and the remaining cooked ovaries were determined and expressed in mouse units per gram (M.U.). Some medicolegal aspects of puffer poisoning are also discussed. Puffer poisoning is a primary cause of fatal food poisoning in Japan even though its frequency is not high.

Abstract: In the newborn rat liver, glycogenosomes appeared at about 6 hours after birth and gradually increased in number, despite high activity of lysosomal acid alpha-glucosidase. The glycogenosomes then disappeared completely with 2 days after birth. Glycogen extracted from fetal rat liver differed in molecular structure from that of adult rat liver. In measurements of optical rotatory dispersion (ORD) and circular dichroism (CD) absorption spectra, fetal-type glycogen showed the Cotton effect whereas the adult rat liver glycogen did not. The degree of branching and S-values of the two glycogens were also different. With the disappearance of glycogenosomes, fetal-type glycogen disappeared completely, and adult-type glycogen then appeared strongly. This was demonstrated in the intracytoplasmic matrix by electron microscopy. The present experiments indicate that glycogenosomes may be formed even under conditions of high acid alpha-glucosidase, and that autophagy of glycogen macromolecules in the lysosomal system is closely related to different structural features of fetal-type glycogen in the neonatal period.

Abstract: Autoantibody-producing B cell lines were established from peripheral blood lymphocytes of patients with systemic lupus erythematosus. Peripheral blood lymphocytes obtained from five of seven patients were successfully transformed by Epstein-Barr virus. Two of four established B lymphoblastoid cell lines examined in this study produced anti-nuclear factor antibodies and one of them produced anti-single-stranded DNA and anti-double-stranded DNA antibodies. These results indicate that B cell clones committed to self antigens are transformed by Epstein-Barr virus and continue to produce autoantibodies. In order to establish a monoclonal autoantibody-producing B cell line, the cells were cloned by a limiting dilution method. The data suggest that it is possible to establish a monoclonal autoantibody-producing B cell line by the combination of transformation of B cells by Epstein-Barr virus and extensive cloning.

Abstract: Treatment of 6-(N-alkylanilino)-2-phenylpyrimidin-4 (3H)-ones, which were prepared by the condensation of 6-chloro-2-phenylpyrimidin-4 (3H)-one with N-alkylanilines, with the Vilsmeier reagent (dimethylformamide-phosphoryl chloride) gave the corresponding 10-alkyl-2-deoxo-2-phenyl-5-deazaflavins (III) in excellent yields. Heating of 6-chloro-5-formyl-3-phenyluracil with N-alkylanilines in dimethylformamide gave the corresponding 10-alkyl-3-phenyl-5-deazaflavins (VI) in a single step. The abilities of the 5-deazaflavins (III) and (VI) thus obtained to oxidize benzyl alcohol and benzylamine were compared, and some automatic recycling of the oxidation reactions was observed. The reaction of compounds III with benzylamine exceptionally gave the adducts, 5-benzylamino-2-deoxo-2-phenyl-5-deazaflavins.

Abstract: Two Japanese males developed spontaneous gangrene of the scrotum and penis. Antibiotics were given and surgical treatments were performed successfully. The latter consisted of debridement of the wound, local flaps and free skin grafts. We failed to obtain significant bacterial cultures. Histology revealed thrombosed and thickened small vessels in the reticular dermis near the border of the gangrenous scrotal tissue.
