Abstract: When C2C12 pluripotent mesenchymal precursor cells are treated with transforming growth factor beta1 (TGF-beta1), terminal differentiation into myotubes is blocked. Treatment with bone morphogenetic protein 2 (BMP-2) not only blocks myogenic differentiation of C2C12 cells but also induces osteoblast differentiation. The molecular mechanisms governing the ability of TGF-beta1 and BMP-2 to both induce ligand-specific responses and inhibit myogenic differentiation are not known. We identified Runx2/PEBP2alphaA/Cbfa1, a global regulator of osteogenesis, as a major TGF-beta1-responsive element binding protein induced by TGF-beta1 and BMP-2 in C2C12 cells. Consistent with the observation that Runx2 can be induced by either TGF-beta1 or BMP-2, the exogenous expression of Runx2 mediated some of the effects of TGF-beta1 and BMP-2 but not osteoblast-specific gene expression. Runx2 mimicked common effects of TGF-beta1 and BMP-2 by inducing expression of matrix gene products (for example, collagen and fibronectin), suppressing MyoD expression, and inhibiting myotube formation of C2C12 cells. For osteoblast differentiation, an additional effector, BMP-specific Smad protein, was required. Our results indicate that Runx2 is a major target gene shared by TGF-beta and BMP signaling pathways and that the coordinated action of Runx2 and BMP-activated Smads leads to the induction of osteoblast-specific gene expression in C2C12 cells.

Abstract: Twenty-one naturally occurring flavonoids were tested for inhibitory activities against alpha-glucosidase (EC 3.2.1.20) and alpha-amylase (EC 3.2.1.1). Luteolin, amentoflavone, luteolin 7-O-glucoside, and daidzein were the strongest inhibitors among the compounds tested. Luteolin inhibited alpha-glucosidase by 36% at the concentration of 0.5 mg/ml and was stronger than acarbose, the most widely prescribed drug, in inhibitory potency, suggesting that it has the possibility to effectively suppress postprandial hyperglycemia in patients with non-insulin dependent diabetes mellitus. Luteolin also inhibited alpha-amylase effectively although it was less potent than acarbose. The clinical value of luteolin needs to be further evaluated.

Abstract: Microwave radiation in Escherichia coli and Bacillus subtilis cell suspensions resulted in a dramatic reduction of the viable counts as well as increases in the amounts of DNA and protein released from the cells according to the increase of the final temperature of the cell suspensions. However, no significant reduction of cell density was observed in either cell suspension. It is believed that this is due to the fact that most of the bacterial cells inactivated by microwave radiation remained unlysed. Scanning electron microscopy of the microwave-heated cells revealed severe damage on the surface of most E. coli cells, yet there was no significant change observed in the B. subtilis cells. Microwave-injured E. coli cells were easily lysed in the presence of sodium dodecyl sulfate (SDS), yet B. subtilis cells were resistant to SDS.

Abstract: Using a longitudinal sample of eighth graders who lived with both biological parents in 1988, we estimate the increased risk of dropping out among students whose two-parent families experienced disruption over the subsequent 4 years. We differentiate the impact on school dropout of initial family income before family disruption from income change after the marriage ends. This approach helps to disentangle two interpretations of the relation between income and family structure. Our results suggest that both measures of income are significant contributors of the risk of dropping out for children whose families became mother-only families. When income loss is taken into account, along with initial income and other family, demographic, and prior school achievement factors, there is no evidence of increased risk of dropout among children who began to live with a single mother during the 1988-1992 period.

Abstract: UBVRI and Hα CCD photometry has been performed in the southern region of NGC 2264 around the Cone Nebula. A nearly complete list of pre–main-sequence (PMS) members in the cluster has been made from the compilation of Hα emission stars identified in this study plus those selected in previous investigations, together with stars with X-ray emission. Using the H-R diagram, we tested a set of four PMS evolution models based on the PMS age and age spread estimate for the cluster and the mass-age relationship among individual stars. The resultant initial mass function (IMF) is in good agreement with the field star IMF of the solar neighborhood, with an IMF slope Γ = -1.7 in the mass range 0.3 ≤ log m ≤ 0.8.

Abstract: Melorheostosis is a rare bone disease characterized by linear hyperostosis and associated soft tissue abnormalities. The skin overlying the involved bone lesion is often tense, shiny, erythematous, and scleodermatous. In order to look for genes differentially expressed between the normal and involved skin, we cultured skin fibroblasts from the skin lesions of several afflicted patients, and identified differentially expressed genes by reverse dot-blot hybridization. We found that the genes human TGF-beta-induced gene product (betaig-h3), osteoblast-specific factor 2, osteonectin, fibronectin, and type I collagen were all downregulated in the affected skin fibroblasts, with betaig-h3 the most significantly affected. The expression of betaig-h3 was induced by TGF-beta in both affected and normal fibroblasts. In an effort to determine the mechanism of bone and skin abnormalities in melorheostosis, we made recombinant betaig-h3. Both immobilized and soluble recombinant betaig-h3 proteins with or without an RGD motif inhibited bone nodule formation of osteoblasts in vitro. Taken together, our results suggest that altered expression of several adhesion proteins may contribute to the development of hyperostosis and concomitant soft tissue abnormalities of melorheostosis, with betaig-h3 in particular playing an important role in osteogenesis.

The entire run I data sample of 110 pb{sup -1} of proton-antiproton collisions at {radical}(s)=1.8 TeV is used to identify a signal sample of {approx}400 B{yields}J/{psi} K{sub S}{sup 0} events, where J/{psi}{yields}{mu}{sup +}{mu}{sup -} and K{sub S}{sup 0}{yields}{pi}{sup +}{pi}{sup -}. The flavor of the neutral B meson is identified at the time of production by combining information from three tagging algorithms: a same-side tag, a jet-charge tag, and a soft-lepton tag. A maximum likelihood fitting method is used to determine sin 2{beta}=0.79{sub -0.44}{sup +0.41}(stat+syst). This value of sin 2{beta} is consistent with the standard model prediction, based upon existing measurements, of a large positive CP-violating asymmetry in this decay mode. Abstract: Critical properties for carbon dioxide + methanol, + ethanol, + 1-propanol, and + 1-butanol were measured using a variable-volume view cell apparatus. Binary mixtures with carbon dioxide mole fractions of 0.976 to 0.637 have been investigated up to 430 K and 175 bar. The dew point and bubble point of the mixtures were measured, particularly in the vicinity of the critical point, in narrow increments in the P−T projection, and the critical point was estimated using two adjacent dew and bubble points. The experimental results were compared with the literature values.
