Abstract: DNA analysis is frequently used to acquire information from biological material to aid enquiries associated with criminal offences, disaster victim identification and missing persons investigations. As the relevance and value of DNA profiling to forensic investigations has increased, so too has the desire to generate this information from smaller amounts of DNA. Trace DNA samples may be defined as any sample which falls below recommended thresholds at any stage of the analysis, from sample detection through to profile interpretation, and can not be defined by a precise picogram amount. Here we review aspects associated with the collection, DNA extraction, amplification, profiling and interpretation of trace DNA samples. Contamination and transfer issues are also briefly discussed within the context of trace DNA analysis. Whilst several methodological changes have facilitated profiling from trace samples in recent years it is also clear that many opportunities exist for further improvements.

Abstract: Primary progressive aphasia comprises a heterogeneous group of neurodegenerative conditions with diverse clinical profiles and underlying pathological substrates. A major development has been the publication of the recent International Consensus Criteria for the three major variants namely: semantic, non-fluent/agrammatic and logopenic. The logopenic variant is assumed to represent an atypical presentation of Alzheimer pathology although evidence for this is, at present, limited. The semantic and non-fluent/agrammatic variants are largely associated with frontotemporal lobar degeneration with TDP-43 and tau pathology, respectively. The applicability of the International Consensus Criteria to an unselected clinical sample is unknown and no agreed clinical evaluation scale on which to derive the diagnosis exists. We assessed 47 consecutive cases of primary progressive aphasic seen over a 3-year period in a specialist centre, using a newly developed progressive aphasia language scale. A subgroup of 30 cases underwent (11)C-labelled Pittsburgh Compound B positron emission tomography imaging, a putative biomarker of Alzheimer's disease that detects β-amyloid accumulation, and they were compared with an age-matched group (n = 10) with typical, predominately amnestic Alzheimer's disease. The application of an algorithm based on four key speech and language variables (motor speech disorders, agrammatism, single-word comprehension and sentence repetition) classified 45 of 47 (96%) of patients and showed high concordance with the gold standard expert clinical diagnosis based on the International Consensus Criteria. The level of neocortical β-amyloid burden varied considerably across aphasic variants. Of 13 logopenic patients, 12 (92%) had positive β-amyloid uptake. In contrast, one of nine (11%) semantic variant and two of eight (25%) non-fluent/agrammatic cases were positive. The distribution of β-amyloid across cortical regions of interest was identical in cases with the logopenic variant to that of patients with typical Alzheimer's disease although the total load was lower in the aphasic cases. Impairments of sentence repetition and sentence comprehension were positively correlated with neocortical burden of β-amyloid, whereas impaired single-word comprehension showed a negative correlation. The International Consensus Criteria can be applied to the majority of cases with primary progressive aphasic using a simple speech and language assessment scale based upon four key variables. β-amyloid imaging confirms the higher rate of Alzheimer pathology in the logopenic variant and, in turn, the low rates in the other two variants. The study offers insight into the biological basis of clinical manifestations of Alzheimer's disease, which appear topographically independent of β-amyloid load.

Abstract: The future of centimetre and metre-wave astronomy lies with the Square Kilometre Array (SKA), a telescope under development by a consortium of 17 countries that will be 50 times more sensitive than any existing radio facility. Most of the key science for the SKA will be addressed through large-area imaging of the Universe at frequencies from a few hundred MHz to a few GHz. The Australian SKA Pathfinder (ASKAP) is a technology demonstrator aimed in the mid-frequency range, and achieves instantaneous wide-area imaging through the development and deployment of phased-array feed systems on parabolic reflectors. The large field-of-view makes ASKAP an unprecedented synoptic telescope that will make substantial advances in SKA key science. ASKAP will be located at the Murchison Radio Observatory in inland Western Australia, one of the most radio-quiet locations on the Earth and one of two sites selected by the international community as a potential location for the SKA. In this paper, we outline an ambitious science program for ASKAP, examining key science such as understanding the evolution, formation and population of galaxies including our own, understanding the magnetic Universe, revealing the transient radio sky and searching for gravitational waves.

Abstract: Reports on the effect of plant residues on soil pH have been contradictory. The conflicting accounts have been suggested to result from differences in compositions and types of plant residues and characteristics of soils. This incubation study examined the effect of plant residues differing in concentrations of N (3–49 g kg−1) and of alkalinity (excess cations) (220–1560 mmol kg−1) on pH change of three soils differing in initial pH (3.9–5.1 in 0.01 M CaCl2). The addition of plant residues at a rate of 15 g kg−1 soil weight increased the pH of all soils by up to 3.4 units and the pH reached the maximum at day 42 after incubation for Wodjil (initial pH 3.87) and Bodallin (pH 4.54) soils and day 14 for Lancelin soil (pH 5.1). The amount of pH buffering was decreased by residue addition in Wodjil soil, increased in Bodallin soil and remained unchanged in Lancelin soil, which closely related to changes of soil pH. Residue addition increased NH 4 + concentration and the increase in NH 4 + concentration generally correlated positively with the concentration of residue N. The NH 4 + concentration increased with time, reached the peak at Days 42–105 for Wodjil soil, Days 14–105 for Bodallin soil and Days 14–42 for Lancelin soil, and then decreased only in Lancelin soil. The concentration of NO 3 − was kept minimal in Wodjil and Bodallin soils. In Lancelin soil, NO 3 − concentrations increased with incubation time from days 14–28. Irrespective of plant residue and incubation time, the amounts of alkalinity produced due to residue addition correlated highly with the sum of the alkalinity added as plant residues (excess cations) and those resulting from mineralization of residue N, with the slope of regression lines decreasing with increase of the initial soil pH. Direct shaking of soil with the residues at the same rate of alkalinity (excess cations) under sterile conditions increased the pH of the Wodjil soil but decreased it in the Lancelin soil. It is suggested that the decarboxylation of organic anions (as indicated by excess cations) of added plant residues and ammonification of the residue N causes soil pH increase whereas nitrification of mineralised residue nitrogen causes soil pH decrease, and that the association/dissociation of organic compounds also plays a role in soil pH change, depending initial pH of the soil. The overall effect on soil pH after addition of plant residues would therefore depend on the extent of each of these processes under given conditions.

Abstract: Cell quotas of microcystin (QMCYST; femtomoles of MCYST per cell), protein, and chlorophyll a (Chl a), cell dry weight, and cell volume were measured over a range of growth rates in N-limited chemostat cultures of the toxic cyanobacterium Microcystis aeruginosa MASH 01-A19. There was a positive linear relationship between QMCYST and specific growth rate (m), from which we propose a generalized model that enables QMCYST at any nutrient-limited growth rate to be predicted based on a single batch culture experiment. The model predicts QMCYST from m, mmax (maximum specific growth rate), QMCYSTmax (maximum cell quota), and QMCYSTmin (minimum cell quota). Under the conditions examined in this study, we predict a QMCYSTmax of 0.129 fmol cell 21 at mmax and a QMCYSTmin of 0.050 fmol cell 21 at m5 0. Net MCYST production rate (RMCYST) asymptotes to zero at m5 0 and reaches a maximum of 0.155 fmol cell 21 day 21 at mmax. MCYST/dry weight ratio (milligrams per gram [dry weight]) increased linearly with m, whereas the MCYST/protein ratio reached a maximum at intermediate m. In contrast, the MCYST/Chl a ratio remained constant. Cell volume correlated negatively with m, leading to an increase in intracellular MCYST concentration at high m. Taken together, our results show that fast-growing cells of N-limited M. aeruginosa are smaller, are of lower mass, and have a higher intracellular MCYST quota and concentration than slow-growing cells. The data also highlight the importance of determining cell MCYST quotas, as potentially confusing interpretations can arise from determining MCYST content as a ratio to other cell components.
