Abstract: This study investigated the green biosynthesis of gold (Au) and silver-gold alloy (Ag-Au) nanoparticles using cell-free extract of Bacillus safensis LAU 13 strain (GenBank accession No: KJ461434). The biosynthesized AuNPs and Ag-AuNPs were characterized using UV-Vis spectroscopy, Fourier-transform infrared spectroscopy, and transmission electron microscopy. Evaluation of the antifungal activities, degradation of malachite green, anti-coagulation of blood, and thrombolysis of human blood clot by the biosynthesized nanoparticles were investigated. The AuNPs and Ag-AuNPs had maximum absorbance at 561 and 545 nm, respectively. The FTIR peaks at 3318, 2378, 2114, 1998, 1636, 1287, 446, $421~\mathrm {cm}^{-1}$ for AuNPs; and 3310, 2345, 2203, 2033, 1636, 1273, 502, 453, 424 $\mathrm {cm}^{-1}$ for Ag-AuNPs indicated that proteins were the capping and stabilization molecules in the biosynthesized nanoparticles. The particles were fairly spherical in shape with size of 10–45 nm for AuNPs and 13–80 nm for Ag-AuNPs. Moreover, energy dispersive X-ray analysis of AuNPs revealed gold as the most prominent metal in the AuNPs solution, while silver and gold were the most prominent in the case of Ag-AuNPs. Selected area electron diffraction showed the biosynthesized nanoparticles as crystal structures with ring shape pattern. AuNPs and Ag-AuNPs displayed growth inhibitions of 66.67–90.78% against strains of Aspergillus fumigatus and A. niger at concentration of 200 $\mu \mathrm {g}/\mathrm {ml}$ , and remarkable degradation (> 90%) of malachite green after 48 h. Furthermore, the nanoparticles prevented coagulation of blood, and also completely dissolved blood clots, indicating the biomedical potential of AuNPs and Ag-AuNPs in the management of blood coagulation disorders. This is the first report of the synthesis of AuNPs and Ag-AuNPs using a strain of B. safensis for biomedical and catalytic applications.

Abstract: Osmotic dehydration of red paprika was studied using a combined sucrose (5 to 45 g/100 g) and sodium chloride (0 to 15 g/100g) solution. The effective diffusion coefficients for water and solute were determined using the slope method based on the Fickian diffusion model. The effects of concentration of sucrose, sodium chloride and their complex interaction on water and solute diffusion coefficients as well as on equilibrium moisture and solid contents were studied using central composite rotatable design of experiments. The graphical optimization showed that at optimum conditions (sucrose concentration and sodium chloride concentration were 21.86 g/100 g and 2.02 g/100 g, respectively), the following criteria were achieved: water diffusion coefficient (D cw ) ≥ 0.80 x 10 -9 m 2 /s, solid diffusion coefficient (D) ≥ 0.82 x 10 -9 m 2 /s, equilibrium moisture content (m∞) ≤ 6.85 kg/kg, and equilibrium solid content (s∞) ≤ 2.00 kg/kg.

Abstract: Rice bran contains underutilised protein materials. Sequential extraction of rice bran protein (RBP) from defatted rice bran was conducted based on the differences in their solubility. Three extraction methods were investigated. Method 1 involved the isoelectric and acetone precipitation using water, 50 g kg−1 NaCl, 0.02 mol L−1 NaOH and 70% ethanol as extracting solvents for albumin (pH 4.1), globulin (pH 4.3), glutelin (pH 4.8) and prolamin, respectively. Method 2 adopted dialysis and sequential extraction was carried out with 20 g kg−1 NaCl, 70% ethanol, 0.1 mol L−1 acetic acid and 0.1 mol L−1 NaOH solution as extracting solvents. Method 3 combined dialysis, isoelectric and acetone precipitation for the extraction. Based on the yields and data obtained from sodium dodecyl sulphate polyacrylamide gel electrophoresis, size-exclusion chromatography and differential scanning calorimetry, method 3 was chosen for the isolation and characterization of RBPs. Rice bran protein fraction (RBPF)—albumin, globulin, glutelin and prolamin were obtained in good yields. Denaturation temperature and enthalpy values of denaturation of RBPF vary. Highest phytate content was found in albumin and lowest in prolamin. The highest antioxidative and hemagglutinating activities were observed in albumin.
