Abstract: The cellulases produced under pH controlled fermentation conditions with 5% Solka Floc and cornsteep liquor as substrates by Trichoderma reesei wild type QM6a and two mutants, Rut-C30 and RL-P37, have been separated by isoelectric focusing in polyacrylamide gels. The total complement of secreted proteins of the two mutants was distinct from the parent. However, the number and isoelectric points of the various enzymes in the cellulase complex were unchanged in the mutants. All secreted proteins stained with Schiff's reagent which indicated they were glycoproteins. One mutant, Rut-C30, exhibited a dramatic shift in the CBH I proteins during the course of the fermentation. RL-P37 showed a two-fold increase in the specific activity of both the total cellulase complex and endoglucanase. In addition a productivity on the order of 100 IU/l/h was achieved. Co-produced with the cellulases were at least two acid proteases with differential activity towards azocoll and azocasein.

Abstract: Although supervision is recognized as a significant tenant of professional growth for counseling and psychotherapy students, the variability of the effectiveness, or ineffectiveness, of supervision has come under scrutiny in recent times. Our sample of 128 participants shed light on the most effective (e.g., encouraged autonomy, strengthened the supervisory relationship, and facilitated open discussion) and most ineffective (e.g., depreciated supervision, performed ineffective client conceptualization and treatment, and weakened the supervisory relationship) supervisor skills, techniques, and behaviors. Moreover, effective and ineffective behaviors, along with best and worst supervisors, were significantly differentiated based on the supervisory working alliance, supervisor style, supervisor self-disclosure, supervisee nondisclosure, and supervisee evaluation. Implications for supervision competencies and supervisor accountability are discussed.

Abstract: A random sample of primary grade teachers from across the United States was surveyed about their instructional practices in handwriting. Nine out of every ten teachers indicated that they taught handwriting, averaging 70 minutes of instruction per week. Only 12% of teachers, however, indicated that the education courses taken in college adequately prepared them to teach handwriting. Despite this lack of formal preparation, the majority of teachers used a variety of recommended instructional practices for teaching handwriting. The application of such practices, though, was applied unevenly, raising concerns about the quality of handwriting instruction for all children.

Abstract: The direct synthesis of hydrogen peroxide from H2 and O2 using a range of Au, Pd and Au–Pd metal nanoparticles supported on iron oxide is described and discussed, and in particular the microstructure of the catalysts are investigated using a detailed electron microscopy study. Iron oxide was selected as a support because Au/Fe2O3 catalysts are known to be very active for low temperature CO oxidation. Hydrogen peroxide synthesis was investigated at low temperatures (2 °C) and short reaction (residence) time, and the addition of Pd to the Au catalyst was found to increase the rate of hydrogen peroxide synthesis as well as the concentration of hydrogen peroxide formed. Indeed the rates of hydrogen peroxide synthesis are higher for the Au–Pd alloy catalysts as compared to the Au or Pd only catalysts. These catalyst materials were also investigated for CO oxidation at 25 °C and all were found to be almost inactive. In contrast, Au-based catalysts that are very effective for low temperature CO oxidation were found to be totally inactive for H2 oxidation to H2O2. This suggests an inverse correlation between catalysts that are active for either CO or H2 activation. The microstructure of the Au–Pd/Fe2O3 catalysts was studied using scanning transmission electron microscopy and the metal alloy nanoparticles were found to have a core–shell morphology with Pd concentrated on the catalyst surface.
