Abstract: Mobile communications have been undergoing a generational change every ten years or so. However, the time difference between the so-called “G’s” is also decreasing. While fifth-generation (5G) systems are becoming a commercial reality, there is already significant interest in systems beyond 5G, which we refer to as the sixth generation (6G) of wireless systems. In contrast to the already published papers on the topic, we take a top-down approach to 6G. More precisely, we present a holistic discussion of 6G systems beginning with lifestyle and societal changes driving the need for next-generation networks. This is followed by a discussion into the technical requirements needed to enable 6G applications, based on which we dissect key challenges and possibilities for practically realizable system solutions across all layers of the Open Systems Interconnection stack (i.e., from applications to the physical layer). Since many of the 6G applications will need access to an order-of-magnitude more spectrum, utilization of frequencies between 100 GHz and 1 THz becomes of paramount importance. As such, the 6G ecosystem will feature a diverse range of frequency bands, ranging from below 6 GHz up to 1 THz. We comprehensively characterize the limitations that must be overcome to realize working systems in these bands and provide a unique perspective on the physical and higher layer challenges relating to the design of next-generation core networks, new modulation and coding methods, novel multiple-access techniques, antenna arrays, wave propagation, radio frequency transceiver design, and real-time signal processing. We rigorously discuss the fundamental changes required in the core networks of the future, such as the redesign or significant reduction of the transport architecture that serves as a major source of latency for time-sensitive applications. This is in sharp contrast to the present hierarchical network architectures that are not suitable to realize many of the anticipated 6G services. While evaluating the strengths and weaknesses of key candidate 6G technologies, we differentiate what may be practically achievable over the next decade, relative to what is possible in theory. Keeping this in mind, we present concrete research challenges for each of the discussed system aspects, providing inspiration for what follows.

Abstract: Objectives This systematic review aimed to identify published observational methods assessing biomechanical exposures in occupational settings and evaluate them with reference to the needs of different users. Methods We searched scientific databases and the internet for material from 1965 to September 2008. Methods were included if they were primarily based on the systematic observation of work, the observation target was the human body, and the method was clearly described in the literature. A systematic evaluation procedure was developed to assess concurrent and predictive validity, repeatability, and aspects related to utility. At least two evaluators independently carried out this evaluation. Results We identified 30 eligible observational methods. Of these, 19 had been compared with some other method(s), varying from expert evaluation to data obtained from video recordings or through the use of technical instruments. Generally, the observations showed moderate-to-good agreement with the corresponding assessments made from video recordings; agreement was the best for large-scale body postures and work actions. Postures of wrist and hand as well as trunk rotation seemed to be more difficult to observe correctly. Intra- and inter-observer repeatability were reported for 7 and 17 methods, respectively, and were judged mostly to be moderate or good. Conclusions With training, observers can reach consistent results on clearly visible body postures and work activities. Many observational tools exist, but none evaluated in this study appeared to be generally superior. When selecting a method, users should define their needs and assess how results will influence decision-making.

Abstract: The formation of amyloid fibrils by the intrinsically disordered protein α-synuclein is a hallmark of Parkinson disease. To characterize the microscopic steps in the mechanism of aggregation of this protein we have used in vitro aggregation assays in the presence of preformed seed fibrils to determine the molecular rate constant of fibril elongation under a range of different conditions. We show that α-synuclein amyloid fibrils grow by monomer and not oligomer addition and are subject to higher-order assembly processes that decrease their capacity to grow. We also find that at neutral pH under quiescent conditions homogeneous primary nucleation and secondary processes, such as fragmentation and surface-assisted nucleation, which can lead to proliferation of the total number of aggregates, are undetectable. At pH values below 6, however, the rate of secondary nucleation increases dramatically, leading to a completely different balance between the nucleation and growth of aggregates. Thus, at mildly acidic pH values, such as those, for example, that are present in some intracellular locations, including endosomes and lysosomes, multiplication of aggregates is much faster than at normal physiological pH values, largely as a consequence of much more rapid secondary nucleation. These findings provide new insights into possible mechanisms of α-synuclein aggregation and aggregate spreading in the context of Parkinson disease.

Abstract: Objective To develop evidence-based recommendations for the diagnosis of knee osteoarthritis (OA). Methods The multidisciplinary guideline development group, representing 12 European countries, generated 10 key propositions regarding diagnosis using a Delphi consensus approach. For each recommendation, research evidence was searched systematically. Whenever possible, the sensitivity, specificity and likelihood ratio were calculated for individual diagnostic indicators and a diagnostic ladder was developed using Bayes' method. Secondary analyses were undertaken to test directly the recommendations using multiple predictive models in two populations from the UK and the Netherlands. Strength of recommendation was assessed by the EULAR visual analogue scale. Results Recommendations covered the definition of knee OA and its risk factors, subsets, typical symptoms and signs, the use of imaging and laboratory tests and differential diagnosis. Three symptoms (persistent knee pain, limited morning stiffness and reduced function) and three signs (crepitus, restricted movement and bony enlargement) appeared to be the most useful. Assuming a 12.5% background prevalence of knee OA in adults aged >= 45 years, the estimated probability of having radiographic knee OA increased with increasing number of positive features, to 99% when all six symptoms and signs were present. The performance of the recommendations in the study populations varied according to the definition of knee OA, background risk and number of tests applied. Conclusion 10 key recommendations for diagnosis of knee OA were developed using both research evidence and expert consensus. Although there is no agreed reference standard, thorough clinical assessment alone can provide a confident rule-in diagnosis.

Abstract: The Conference on Computational Natural Language Learning is accompanied every year by a shared task whose purpose is to promote natural language processing applications and evaluate them in a standard setting. In 2008 the shared task was dedicated to the joint parsing of syntactic and semantic dependencies. This shared task not only unifies the shared tasks of the previous four years under a unique dependency-based formalism, but also extends them significantly: this year's syntactic dependencies include more information such as named-entity boundaries; the semantic dependencies model roles of both verbal and nominal predicates. In this paper, we define the shared task and describe how the data sets were created. Furthermore, we report and analyze the results and describe the approaches of the participating systems.
