Abstract: Micro-Raman spectra of formalin-fixed oral squamous normal and carcinoma tissues, stored at room temperature for 2 months, have been recorded. Spectra were recorded both in the epithelial and subepithelial regions of the tissues. No noticeable spectral contamination due to formalin was observed. Very significant differences between spectra of normal epithelial and malignant epithelial samples were found. No such differences in spectra of subepithelial malignant and subepithelial normal samples could be observed. This study shows that spectra from the epithelial region changes drastically because of malignancy-induced biochemical changes in this region. Major differences between normal and malignant spectra seem to arise from the protein composition, conformational/structural changes, and possible increase in protein content in malignant epithelia. The differences between normal epithelial and subepithelial spectra, as expected, arise mainly from the collagen in subepithelial tissue. Principal component analysis of the combined sets of spectra-epithelial and subepithelial, normal and malignant- showed that very good discrimination can be achieved by Raman microspectroscopy. This study thus validates the suitability of formalin-fixed tissues for optical pathology in oral malignancy.

Abstract: A number of ring substituted analogues of curcumin were synthesized. Their antioxidant properties were studied using three models, inhibition of lipid peroxidation, scavenging of 1,1′-diphenyl-2-picrylhydrazyl (DPPH) and 2,2′-azinobis(3-ethyl-benzthiazoline-6-sulphonate radical (ABTS+). In all the models, the phenolic analogues were more active than the non-phenolic analogues, some of which were inactive. The highest antioxidant activity was obtained when the phenolic group was sterically hindered by the introduction of two methyl groups at the ortho position. This and several other compounds were more active than the standard antioxidants α-tocopherol and trolox. This study has demonstrated that the phenolic group is important for the antioxidant activity of curcumin and that the structural features that enhance the antioxidant properties of phenols are optimized in curcumin to a significant extent.

Abstract: With increasing evidence for faulty paternal contribution to reproduction, there has been a steady increase in studies highlighting an association between sperm DNA damage, failed/delayed fertilisation and aberrant embryo development. Owing to prevailing ambiguity, the aims of the study were to analyse the genetic integrity of the male gamete and then to understand its association with age, standard semen parameters, lifestyle and occupational factors. The study included 504 subjects, attending university infertility clinic for fertility evaluation and treatment. Semen characteristics were analysed by standard criteria; terminal deoxynucelotidyl transferase-mediated nick end-labelling assay was employed for DNA damage assessment. The average incidence of sperm DNA damage in patients with normozoospermic semen parameters was <10%. Patients with oligozoospermia, severe oligozoospermia, oligoasthenoteratospermia, asthenoteratozoospermia and necrozoospermia had significantly higher level of sperm DNA damage (P < 0.001). Patients above 40 years of age had significantly high levels of DNA damage (P < 0.001) compared with their counterparts. Patients with varicocele and a history of alcohol consumption had higher incidence of spermatozoa with DNA damage (P < 0.01). Poor sperm characteristics in the ejaculate are associated with increased sperm DNA damage. Age-related increase in sperm DNA damage and association of the same with varicocele and alcohol consumption are also demonstrated.

Abstract: An environmentally sound approach towards the green synthesis of zinc oxide nanostructures has been achieved with an aqueous extract of Calliandra haematocephala leaves. The nanoparticles were characterized using various analytical techniques to substantiate the structural details. An absorption band at 358 nm corresponds to the formation of zinc oxide nanoparticles. Scanning electron microscopy revealed the nanoflower morphology of the nanoparticles. Energy dispersive spectral analysis portrayed the strong presence of zinc and oxygen, while X-ray diffraction showed the nanoparticles to conform to hexagonally-formed wurtzite structure. The crystallite size of the nanoflowers was estimated to be 19.45 nm. Vibrational frequencies, typical of zinc‑oxygen and other functional groups, were revealed using Fourier transform infrared spectroscopy. BET analysis revealed that the pores were of mesoporous nature with an estimated specific surface area of 9.18 m2/g. The photocatalytic nature of the nanoparticles was established by the degradation of methylene blue (MB) dye, under solar radiation. Up to 88% degradation was achieved in a duration of 270 min. Kinetic data from the studies proved that the reaction was compliant with first-order model, with rate constant as 0.01 min−1. The study illustrated the synthesis of zinc oxide nanoparticles using a novel source, viz., the leaves of C. haematocephala.
