Abstract: The cell membrane is a highly selective barrier. This limits the cellular uptake of molecules including DNA, oligonucleotides, peptides and proteins used as therapeutic agents. Different approaches have been employed to increase the membrane permeability and intracellular delivery of these therapeutic molecules. One such approach is the use of Cell Penetrating Peptides (CPPs). CPPs represent a new and innovative concept, which bypasses the problem of bioavailability of drugs. The success of CPPs lies in their ability to unlock intracellular and even intranuclear targets for the delivery of agents ranging from peptides to antibodies and drug-loaded nanoparticles. This review highlights the development of cell penetrating peptides for cell-specific delivery strategies involving biomolecules that can be triggered spatially and temporally within a cell transport pathway by change in physiological conditions. The review also discusses conjugations of therapeutic agents to CPPs for enhanced intracellular delivery and bioavailability that are at the clinical stage of development.

Abstract: Climate is a key input of rain-fed agriculture. Climate variability and change has been the most important determinant of crop yields in Kenya and other parts of the world. However, there has been not much research on local understanding of the effect of climate variability on maize yields in Arid and Semi arid Lands (ASALs) of lower eastern Kenya counties. The effect of three climatic parameters on maize yields on different temporal and spatial scales was evaluated in order to provide basis for maize crop monitoring and modeling. This paper argues that maize yields were declining at high levels in Machakos County followed by Kitui, Mwingi, and Makueni Counties. The maize yields Z-values and thus the effect of climate was predominately negative in the period 1994–2008 in all the counties. Rainfall trend analysis revealed that four of the six weather stations were declining up to 3 mm pa. Evidently there was upward warming of annual and seasonal temperatures at rate of 0.03 °C pa. The study has confirmed that the arid and semi arid counties suffer from significant climate variability which has huge implications on maize yields and food security of lower eastern Kenya. Thus, to counter the adverse effects of climate change, it is necessary to climate-proof agricultural crops through adaptation strategies such as developing maize varieties that tolerate water stress and mature early, practice early planting, increase the awareness of climate change and its impacts on agriculture, and develop appropriate mitigation measures. These findings are crucial in planning appropriate adaptation mechanisms in support of enhancing resilience of maize production and food security.

Abstract: This study investigates the spatial-temporal trends and variability of rainfall within East and South Africa (ESA) region. The newly available Climate Hazards group Infrared Precipitation with Stations (CHIRPS-v2) gridded data spanning 37 years (1981 to 2017) was validated against gauge observations (N = 4243) and utilised to map zones experiencing significant monotonic rainfall trends. Standardised annual rainfall anomalies revealed the spatial-temporal distribution of below and above normal rains that are associated with droughts and floods respectively. Results showed that CHIRPS-v2 data had a satisfactory skill to estimate monthly rainfall with Kling-Gupta efficiency (KGE = 0.68 and a high temporal agreement (r = 0.73) while also preserving total amount (β = 0.99) and variability (γ = 0.8). Two contiguous zones with significant increase in annual rainfall (3–15 mm year−1) occurred in Southwest Zambia and in Northern Lake Victoria Basin between Kenya and Uganda. The most significant decrease in annual rainfall (− 20 mm year−1) was recorded at Mount Kilimanjaro in Tanzania. Other significant decreases in annual rainfall ranging between − 4 and − 10 mm year−1 were observed in Southwest Tanzania, Central-South Kenya, Central Uganda and Western Rwanda. CHIRPS-v2 rainfall product provides reliable high spatial resolution information on amount of rainfall that can complement sparse rain gauge network in rain-fed agricultural systems in ESA region. The observed spatial-temporal trends and variability in rainfall are important basis for guiding targeting of appropriate adaptive measures across multiple sectors.

Abstract: The detection of antibiotics in water systems has instigated great environmental concern due to the toxicological effects associated with these compounds. Their discharge into the environment results from the ubiquity of use in medical, veterinary, and agricultural practices. Some of the effects of antibiotics include development of antibiotic-resistant bacteria, making it difficult to treat diseases, variation in natural microbial communities, and enzyme activities. In this study, the first comprehensive survey of some frequently used antibiotics namely ampicillin (AMP), amoxicillin (AMX), sulfamethoxazole (SMX), chloramphenicol (CAP), and ciprofloxacin (CPF) within Lake Victoria Basin of Kenya is presented. Sludge and wastewater samples were collected from wastewater treatment plants (WWTPs) and hospital lagoons within the study area. Samples were extracted and cleaned by solid-phase extraction, and analysis was carried out using high-performance liquid chromatography (HPLC). All wastewater samples and sludge collected contained quantifiable levels of the selected antibiotics. The highest concentrations were recorded for AMP with WWTPs and hospitals having 0.36 ± 0.04 and 0.79 ± 0.07 μg/L, respectively. In sludge samples, SMX recorded the highest concentrations of 276 ± 12 ng/g. The high levels in sludge indicate the preferential partition of antibiotics onto solid phase, posing great danger to consumers of crops grown in biosolid-amended soils. The daily discharge loads of antibiotics from nine WWTPs ranged between 80.75 and 3044.9 mg day−1 with a total discharge of 6395.85 mg day−1, signifying a high potential of water resource pollution within the region. This report will aid in the assessment of the risks posed by antibiotics released into the environment.
