Abstract: Vaccination against foot-and-mouth disease is an effective tool for eradication and prevention of this disease. However, the presence of non-capsidal proteins (NCP) in the vaccine, produced during viral replication, has been the main problem, since their presence hamper the vigilance as its relies on the detection of antibodies against NCP to differentiate vaccinated from infected animals. Therefore, the Brazilian Ministry of Agriculture, Livestock and Supply published the Normative Instruction 50 (IN 50) that included the detection of antibodies against the NCP to evaluate the removal of these proteins. Considering the vaccine interference, this paper aimed to evaluate the frequency of reagents to NCP, analyzed by I-ELISA 3ABC/EITB system in the Laboratory of Bovine Viruses (LVB), Biological Institute (IB), Sao Paulo, SP, Brazil, from 2002 to 2012. Of the 34,705 cattle examined, it was observed that the proportion of reagents to NCP increased with age, indicating increased frequency of reagents in animals that received more vaccines, showing interference of proteins in response to vaccination. When compared before and after the publication of IN 50, there was decreased reactivity, with reduction in 2010, of nearly half compared to 2007, and even higher when compared with 2002 to 2006. This shows the effectiveness of the purification of the vaccine in response to the implementation of IN 50, although it remains some seroreactivity in cattle with multiple vaccinations. The I-ELISA 3ABC/EITB system proved to be a great tool to prevent the movement of possible carriers of the virus derived from vaccinated herds, provided that all sanitary and epidemiological context are taken in consideration.

Abstract: The effects of decreasing levels of crude protein (CP) and enzymatic supplementation on digestive traits and caecal content composition of growing rabbits were studied. In a completely randomized design 54 weaned New Zealand White rabbits were divided in four groups of 11 rabbits each and one group of 10 rabbits. The treatment groups fed on diets with 17, 16, 15 or 14% of CP supplemented with 0.05% of enzymatic complex (Vegproâ) with cellulase and protease activities. The control group fed a diet with 17% of CP without the enzymatic complex. The weigth of the full digestive tract, full and empty caecum, and caecum content were affected (P<0.05) by the CP level of the diet. Organic matter percentage, ammonia nitrogen percentage, and molar proportion of acetic and propionic acid in the caecum content were affected by CP level of the diet (P<0.05). The redution of CP level of the diet up to 14% did not show negative effects on evaluated traits.

Dieser Effekt beruht auf der Bildung und pyrolytischen Zersetzung sogenannter Buntescher Salze.

Abstract: This study aimed at evaluating the sensitivity of a multiplex PCR technique for detecting fraud by the deliberate addition of buffalo meat into the minced bovine beef. The minced bovine beef containing 0.01 %, 0.1 %, 1.0 %, 5.0 %, 10.0 %, 25.0 %, 50.0 %, 75.0 %, 90.0 %, 95.0 %, 99 %, 99.9 %, 99.99 % of minced buffalo beef were produced in triplicate, with five replicates. The minced meats exclusively of each animal species were used as control samples. Every prepared sample was analyzed by means of multiplex polymerase chain reaction. For performing the assay sensitivity analysis, the known concentrations of DNA samples (0.01 %, 0.1 %, 1.0 %, 5.0 %, 10.0 %, 25.0 %, 50.0 %, 75.0 %, 90.0 %, 95.0 %, 99.0 %, 99.9 %, 99.99 %) from Bos taurus and Bubalus bubalis species were diluted and mixed into a final volume of 10 μL; and these DNA samples were also assayed by the proposed PCR methodology. The multiplex PCR showed to be is an effective and highly sensitive technique enough for detecting increments of 10 % (2.05 ng) of bubaline DNA and of 0.1 % (0.041 ng) of bovine DNA.
