Abstract: OBJETIVO: Estimar a concentracao de chumbo em colostro humano e verificar a existencia de relacao entre esse dado, consumo alimentar e fatores socioeconomicos das puerperas. METODOS: Estudo transversal com 80 puerperas de uma maternidade de Goiânia, Brasil, realizado de julho a dezembro de 2008. Aplicou-se um questionario socioeconomico e de estilo de vida, alem do questionario de frequencia de consumo alimentar. Posteriormente, foram coletadas amostras de colostro, analisadas por espectrometria de absorcao atomica por chama para quantificar o chumbo. Calculou-se a correlacao entre o teor de chumbo, as variaveis socioeconomicas e o consumo alimentar. RESULTADOS: A media da concentracao de chumbo foi 6,88 μg/L, e a mediana foi 4,65 μg/L. Os aspectos socioeconomicos nao apresentaram associacao significativa com o teor de chumbo. A ingestao de acidos graxos saturados, ovos, carnes suinas, frango e derivados (todos com valores de r = -0,26 e P = 0,020) correlacionou-se inversamente ao teor chumbo do colostro humano, ao passo que a ingestao de abobora (r = 0,26 e P = 0,019) e berinjela (r = 0,27 e P = 0,015) correlacionou-se proporcionalmente ao valor obtido. A correlacao entre abobora cozida, ovos, carnes suinas, de frango e derivados foi mantida apos ajuste na regressao logistica. CONCLUSOES: Foi detectada presenca de chumbo no colostro materno, possivelmente como resultado da exposicao dietetica das puerperas deste estudo. Alem da correlacao entre presenca de chumbo e consumo alimentar, os resultados sugerem que a ingestao de nutrientes especificos pode propiciar maior ou menor concentracao desse contaminante, sem relacao direta com fatores socioeconomicos.

Abstract: In order to achieve successful captive breeding the Podocnemis expansa, it is necessary to study their reproductive endocrinology. The purpose of this research was to evaluate and characterize plasma concentrations in gonadotrophic, gonadic, corticosterone and prolactin hormones from Giant Amazon Turtles under captive conditions. Blood samples were collected over a 15 month period. The samples were assayed by the use of radioimmunoassay, prolactin, corticosterone, LH, FSH, testosterone, 17β-estradiol and progesterone. We verified significant seasonal pattern increase in 17β-estradiol levels and decrease in progesterone levels in the course of a year, which indicates vitellogenesis. This is related to normal ovarian cycles and possibly to the functional integrity of the hypothalamus-pituitary-gonad axis of captive females. There were negative correlations between testosterone and corticosterone in the male samples, suggestive of stress (management stress) on the reproductive system. The plasma concentrations of gonadotrophic, gonadic, prolactin and corticosterone hormones may be used as a reference for further research and possible therapeutic approaches. The data collected during this research are unprecedented for this species and may serve as a reference for future research regarding the reproductive cycle of this turtle, also allowing reproductive management while in captivity. Information about these hormones must be gathered from wild populations during different periods of the year for better clarification of the reproductive physiology of this species.

Abstract: A procedure was developed for determination of 5 sedatives and 14 β-blockers in swine kidney and subsequent analysis by liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS). Three different procedures for extraction were tested, evaluated through recovery studies. The procedure using acetonitrile for extraction and cleanup with freezing at low temperature and dispersive solid phase extraction using 500 mg celite® 545 before the concentration step presented the better results. The dried samples were redissolved with methanol and analyzed using a LC-MS/MS system with electrospray ionization (ESI) operating in positive MRM mode. The recovery values for this procedure were in the 75-88% range. The robustness of the method was tested against small variations. The method was used to analyze carazolol, azaperone and azaperol in collaborative assay, obtaining results close to designed value.
