Abstract: In Arabidopsis thalinana (L.) Heynh., DHDPS1 and DHDPS2 encode orthologous dihydrodipicolinate synthases (DHDPS), the first enzyme of the lysine (Lys) biosynthesis pathway. A TDNA insertion mutant of dhdps2 was previously reported to be viable and to accumulate free threonine (Thr). Analysis of additional TDNA insertion lines showedthatdhdps1anddhdps2mutantsarebothviableandthatwhereasdhdps2mutantsaccumulateThr,dhdps1plantsdo not. Thr-accumulation was complemented by heterologous expression of Escherichia coli DapA, indicating that the phenotype is due to reduced DHDPS activity in dhdps2. DHDPS1 contributes ~30% towards the total DHDPS activity in leaves of youngplants andDHDPS2contributes 70%; therefore, thethreshold of activity resulting inThr accumulation lies within this narrow range. dhdps1-dhdps2 double mutants could not be isolated, even after exogenous feeding with Lys. SegregationanalysisindicatedthatgameteslackingfunctionalDHDPSgenesaredefective,asareembryos.Plantscarrying only a single DHDPS2 gene do not accumulate Thr, but they show a gametophytic defect that is partially rescued by Lys application. Despite the accumulation of Thr, dhdps2 seedlings are no more sensitive than wild-type plants to growth inhibition by Lys or the Lys precursor diaminopimelate. They also are not rescued by methionine at growth-inhibitory Lys concentrations. Exogenous application of Lys and methionine to dhdps2 mutants did not reduce the accumulation of Thr.

Abstract: Identifying potential patterns in pirarucu ( Arapaima gigas ) composition, as a function of the different fish muscle zones (dorse, venter, ventrecha and tail), was the main objective of this research. For such, the different pirarucu muscle zones were evaluated, in order to obtain proximate composition and minerals content. It was also determined amino acids and fatty acids contents in fish muscle. The dorsal, ventral and tail muscle zones presented similar moisture (76.5-78.2%), protein (17.8-18.9%), total lipids (1.0-1.5%) and ash (0.9-1.2%) contents. On the other hand, the ventrecha zone presented 25.8% of protein and the major total lipids content (17.1%). The main minerals found in fish muscle were K (183.5-288.6 mg/100 g muscle) and Na (65.1-175.5 mg/100 g). Glutamic acid (3027.6 mg/100 g muscle) was the main amino acid found in fish muscle, which presented 48% of essential amino acids in the protein fraction. Lipids content showed 57.3% of unsaturated fatty acids and 42.7% of saturated fatty acids.

Abstract: The objective of this study was to describe the epidemiological situation of bovine tuberculosis in Mato Grosso do Sul. The State was divided into three regions, Pantanal, Planalto Norte, and Planalto Sul. In each region, properties were randomly chosen and a pre-established number of animals were also randomly selected; these animals were submitted to the intradermal comparative cervical tuberculin diagnostic test. In total, 17,121 animals from 938 properties were tested. In cases of inconclusive results, the animals were retested with the same diagnostic procedure within a minimum interval of 60 days. An epidemiological questionnaire was administered in the properties sampled to investigate possible risk factors for the disease. In the State, the prevalence of infected herds was 1.3% (95% confidence interval [CI]: 0.72–2.37%) and that of infected animals was 0.035% (95% CI: 0.017–0.069%). Point estimates indicated a higher concentration of infected herds (2.61%; 95% CI: 1.31–5.15%) and infected animals (0.132%; 95% CI: 0.055–0.315%) in the Planalto Sul region, which has a predominance of dairy properties. The prevalence of herds infected with bovine tuberculosis was found to be associated with the production of milk, with technification in the mode of production represented by the use of mechanical milking. Thus, it is recommended that the State adopt strategies for the eradication of bovine tuberculosis, with the structuring of the surveillance system for detection and mitigation of infected herds, incorporating risk-based surveillance elements.
