Abstract: Osteoarthritis (MIM 165720), characterized by degeneration of articular cartilage, is the most common form of human arthritis and a major concern for aging societies worldwide. Epidemiological and genetic studies have shown that osteoarthritis is a polygenic disease. Here, we report that the gene encoding growth differentiation factor 5 (GDF5) is associated with osteoarthritis in Asian populations. A SNP in the 5' UTR of GDF5 (+104T/C; rs143383) showed significant association (P = 1.8 x 10(-13)) with hip osteoarthritis in two independent Japanese populations. This association was replicated for knee osteoarthritis in Japanese (P = 0.0021) and Han Chinese (P = 0.00028) populations. This SNP, located in the GDF5 core promoter, exerts allelic differences on transcriptional activity in chondrogenic cells, with the susceptibility allele showing reduced activity. Our findings implicate GDF5 as a susceptibility gene for osteoarthritis and suggest that decreased GDF5 expression is involved in the pathogenesis of osteoarthritis.

Abstract: As the rapid development of automotive telematics, modern vehicles are expected to be connected through heterogeneous radio access technologies and are able to exchange massive information with their surrounding environment. By significantly expanding the network scale and conducting both real time and long term information processing, the traditional Vehicular Ad- Hoc Networks U+0028 VANETs U+0029 are evolving to the Internet of Vehicles U+0028 IoV U+0029, which promises efficient and intelligent prospect for the future transportation system. On the other hand, vehicles are not only consuming but also generating a huge amount and enormous types of data, which are referred to as Big Data. In this article, we first investigate the relationship between IoV and big data in vehicular environment, mainly on how IoV supports the transmission, storage, computing of the big data, and in return how IoV benefits from big data in terms of IoV characterization, performance evaluation and big data assisted communication protocol design. We then investigate the application of IoV big data for autonomous vehicles. Finally the emerging issues of the big data enabled IoV are discussed.

Abstract: Severe winter air pollution events, attributed to emissions from development, have increased in Beijing in recent decades. This study looks at how atmospheric conditions contribute and projects climate change will increase conditions favourable to such events. The frequency of Beijing winter severe haze episodes has increased substantially over the past decades1,2,3,4, and is commonly attributed to increased pollutant emissions from China’s rapid economic development5,6. During such episodes, levels of fine particulate matter are harmful to human health and the environment, and cause massive disruption to economic activities3,4,7,8,9,10,11,12,13,14,15,16, as occurred in January 201317,18,19,20,21. Conducive weather conditions are an important ingredient of severe haze episodes3,21, and include reduced surface winter northerlies3,21, weakened northwesterlies in the midtroposphere, and enhanced thermal stability of the lower atmosphere1,3,16,21. How such weather conditions may respond to climate change is not clear. Here we project a 50% increase in the frequency and an 80% increase in the persistence of conducive weather conditions similar to those in January 2013, in response to climate change. The frequency and persistence between the historical (1950–1999) and future (2050–2099) climate were compared in 15 models under Representative Concentration Pathway 8.5 (RCP8.5)22. The increased frequency is consistent with large-scale circulation changes, including an Arctic Oscillation upward trend23,24, weakening East Asian winter monsoon25,26, and faster warming in the lower troposphere27,28. Thus, circulation changes induced by global greenhouse gas emissions can contribute to the increased Beijing severe haze frequency.
