Abstract: Oxygen isotope records of five stalagmites from Hulu Cave near Nanjing bear a remarkable resemblance to oxygen isotope records from Greenland ice cores, suggesting that East Asian Monsoon intensity changed in concert with Greenland temperature between 11,000 and 75,000 years before the present (yr. B.P.). Between 11,000 and 30,000 yr. B.P., the timing of changes in the monsoon, as established with 230Th dates, generally agrees with the timing of temperature changes from the Greenland Ice Sheet Project Two (GISP2) core, which supports GISP2's chronology in this interval. Our record links North Atlantic climate with the meridional transport of heat and moisture from the warmest part of the ocean where the summer East Asian Monsoon originates.

Abstract: Researchers worldwide with information about the Kirsten ras (Ki-ras) tumour genotype and outcome of patients with colorectal cancer were invited to provide that data in a schematized format for inclusion in a collaborative database called RASCAL (The Kirsten ras incolorectal-cancer collaborative group). Our results from 2721 such patients have been presented previously and for the first time in any common cancer, showed conclusively that different gene mutations have different impacts on outcome, even when the mutations occur at the same site on the genome. To explore the effect of Ki-ras mutations at different stages of colorectal cancer, more patients were recruited to the database, which was reanalysed when information on 4268 patients from 42 centres in 21 countries had been entered. After predetermined exclusion criteria were applied, data on 3439 patients were entered into a multivariate analysis. This found that of the 12 possible mutations on codons 12 and 13 of Kirsten ras, only one mutation on codon 12, glycine to valine, found in 8.6% of all patients, had a statistically significant impact on failure-free survival (P=0.004, HR 1.3) and overall survival (P=0.008, HR 1.29). This mutation appeared to have a greater impact on outcome in Dukes' C cancers (failure-free survival, P=0.008, HR 1.5, overall survival P=0.02, HR 1.45) than in Dukes' B tumours (failure-free survival, P=0.46, HR 1.12, overall survival P=0.36, HR 1.15). Ki-ras mutations may occur early in the development of pre-cancerous adenomas in the colon and rectum. However, this collaborative study suggests that not only is the presence of a codon 12 glycine to valine mutation important for cancer progression but also that it may predispose to more aggressive biological behaviour in patients with advanced colorectal cancer. ⌐ 2001 Cancer Research Campaign.

Abstract: A novel maximum-power-point-tracking (MPPT) controller for a photovoltaic (PV) energy conversion system is presented. Using the slope of power versus voltage of a PV array, the proposed MPPT controller allows the conversion system to track the maximum power point very rapidly. As opposed to conventional two-stage designs, a single-stage configuration is implemented, resulting in size and weight reduction and increased efficiency. The proposed system acts as a solar generator on sunny days, in addition to working as an active power line conditioner on rainy days. Finally, computer simulations and experimental results demonstrate the superior performance of the proposed technique.

Abstract: Although extended-spectrum beta-lactamases (ESBLs) hydrolyze cephalosporin antibiotics, some ESBL-producing organisms are not resistant to all cephalosporins when tested in vitro. Some authors have suggested that screening klebsiellae or Escherichia coli for ESBL production is not clinically necessary, and when most recently surveyed the majority of American clinical microbiology laboratories did not make efforts to detect ESBLs. We performed a prospective, multinational study of Klebsiella pneumoniae bacteremia and identified 10 patients who were treated for ESBL-producing K. pneumoniae bacteremia with cephalosporins and whose infecting organisms were not resistant in vitro to the utilized cephalosporin. In addition, we reviewed 26 similar cases of severe infections which had previously been reported. Of these 36 patients, 4 had to be excluded from analysis. Of the remaining 32 patients, 100% (4 of 4) patients experienced clinical failure when MICs of the cephalosporin used for treatment were in the intermediate range and 54% (15 of 28) experienced failure when MICs of the cephalosporin used for treatment were in the susceptible range. Thus, it is clinically important to detect ESBL production by klebsiellae or E. coli even when cephalosporin MICs are in the susceptible range (≤ 8 μg/ml) and to report ESBL-producing organisms as resistant to aztreonam and all cephalosporins (with the exception of cephamycins).

Abstract: We compute the branching ratios of $\stackrel{\ensuremath{\rightarrow}}{B}K\ensuremath{\pi}$ decays in the framework of the perturbative QCD factorization theorem. Decay amplitudes are classified into the topologies of tree, penguin, and annihilation amplitudes, all of which contain both factorizable and nonfactorizable contributions. These contributions are expressed as the convolutions of hard $b$ quark decay amplitudes with universal meson wave functions. It is shown that (1) matrix elements of penguin operators are dynamically enhanced compared to those employed in the factorization assumption, (2) annihilation diagrams are not negligible, contrary to common belief, (3) annihilation diagrams contribute large strong phases, and (4) the uncertainty of the current data of the ratio $R=\mathrm{Br}{(B}_{d}^{0}\ensuremath{\rightarrow}{K}^{\ifmmode\pm\else\textpm\fi{}}{\ensuremath{\pi}}^{\ensuremath{\mp}})/\mathrm{Br}{(B}^{\ifmmode\pm\else\textpm\fi{}}\ensuremath{\rightarrow}{K}^{0}{\ensuremath{\pi}}^{\ifmmode\pm\else\textpm\fi{}})$ and of $\mathrm{CP}$ asymmetries is too large to give a constraint of the unitarity angle ${\ensuremath{\varphi}}_{3}.$ Assuming ${\ensuremath{\varphi}}_{3}=90\ifmmode^\circ\else\textdegree\fi{}$ which is extracted from the best fit to the data of $R,$ predictions for the branching ratios of the four $\stackrel{\ensuremath{\rightarrow}}{B}K\ensuremath{\pi}$ modes are consistent with data.

Abstract: Dengue virus infection causes dengue fever (DF), dengue hemorrhagic fever (DHF), and dengue shock syndrome (DSS), whose pathogeneses are not clearly understood. Current hypotheses of antibody-dependent enhancement, virus virulence, and IFN-γ/TNFα-mediated immunopathogenesis are insufficient to explain clinical manifestations of DHF/DSS such as thrombocytopenia and hemoconcentration. Dengue virus infection induces transient immune aberrant activation of CD4/CD8 ratio inversion and cytokine overproduction, and infection of endothelial cells and hepatocytes causes apoptosis and dysfunction of these cells. The coagulation and fibrinolysis systems are also activated after dengue virus infection. We propose a new hypothesis for the immunopathogenesis for dengue virus infection. The aberrant immune responses not only impair the immune response to clear the virus, but also result in overproduction of cytokines that affect monocytes, endothelial cells, and hepatocytes. Platelets are destroyed by crossreactive anti-platelet autoantibodies. Dengue-virus-induced vasculopathy and coagulopathy must be involved in the pathogenesis of hemorrhage, and the unbalance between coagulation and fibrinolysis activation increases the likelihood of severe hemorrhage in DHF/DSS. Hemostasis is maintained unless the dysregulation of coagulation and fibrinolysis persists. The overproduced IL-6 might play a crucial role in the enhanced production of anti-platelet or anti-endothelial cell autoantibodies, elevated levels of tPA, as well as a deficiency in coagulation. Capillary leakage is triggered by the dengue virus itself or by antibodies to its antigens. This immunopathogenesis of DHF/DSS can account for specific characteristics of clinical, pathologic, and epidemiological observations in dengue virus infection.

Abstract: 2,3-Butanediol (2,3-BDL), which is very important for a variety of chemical feedstocks and liquid fuels, can be derived from the bioconversion of natural resources. One of its well known applications is the formation of methyl ethyl ketone, by dehydration, which can be used as a liquid fuel additive. This article briefly reviews the basic properties of 2,3-BDL and the metabolic pathway for the microbial formation of 2,3-BDL. Both the biological production of 2,3-BDL and the variety of strains being used are introduced. Genetically improved strains for BDL production which follow either the original mechanisms or new mechanisms are also described. Studies on fermentation conditions are briefly reviewed. On-line analysis, modeling, and control of BDL fermentation are discussed. In addition, downstream recovery of 2,3-BDL and the integrated process (being important issues of BDL production) are also introduced.

Abstract: The importance of aldosterone in the pathophysiology of chronic heart failure (HF) has been established in previous studies [1–4] and is emphasized by the findings of the RALES trial [5]. In this study, aldosterone blockade with spironolactone resulted in a 30% reduction in total mortality and a 35% reduction in hospitalizations for HF in patients with pre-existing chronic severe HF. Patients in the RALES trial also received standard therapy including an ACE inhibitor (if tolerated), a loop diuretic, and digoxin. While aldosterone receptor blockade has been proven beneficial in severe chronic HF due to systolic left ventricular (LV) dysfunction, its effects in patients with acute myocardial infarction (AMI) complicated by HF due to systolic left ventricular dysfunction are unknown. The pathophysiology of HF complicating AMI is complex. Factors such as the acute release of catecholamines, activation of the renin angiotensin aldosterone system, degree of ventricular remodeling, myocardial scar formation, extent of coronary artery disease, and residual ischemia may differ both quantitatively and qualitatively in patients with acute infarction compared to patients with chronic HF. Additionally, the extent of activation of cytokines, fibrinolytic balance, and activity of clotting factors may differ. Eplerenone was chosen for this study because of its demonstrated efficacy in an experimental model of AMI [6]. In clinical trials, eplerenone demonstrated efficacy similar to spironolactone in blocking aldosterone receptors, lowering blood pressure, and moderating hormonal and neurohormonal markers of HF [7,8]. However, eplerenone has significantly less affinity for androgen and progesterone receptors and should therefore be associated with a lower incidence of gynecomastia, breast pain and impotency in males, and diminished libido and menstrual irregularities in females [9–11]. While older patients suffering from refractory HF may tolerate these androgenic and progestational side effects, they may preclude widespread use of a nonspecific aldosterone antagonist in younger patients or in patients with less severe cardiac compromise. Since eplerenone is at least 100 times more specific in its affinity for aldosterone receptors than is spironolactone, if the hypothesis being tested in the EPHESUS trial proves correct, eplerenone has the potential to be used in a broad population to prevent progressive left ventricular remodeling, ventricular fibrosis, malignant arrhythmias, non-fatal AMI, and sudden cardiac death. We hypothesize that selective aldosterone receptor blockade with eplerenone will have a beneficial effect on survival and morbidity in patients with AMI complicated by HF due to systolic left ventricular dysfunction. This paper describes the background, design, and organization of a trial to test this hypothesis—the EPHESUS trial (Eplerenone Post-AMI Heart Failure Efficacy and Survival Study).

Abstract: Since the disposition of end-of-life home appliances has caused tremendous attention, Taiwan recently promulgated a Scrap Home Appliances and Computers Recycling Regulation that mandates manufacturers and importers to take back their products. Reverse logistics system planning shall become vital as the take-back rate increases and the service area expands in the future. This study utilizes a mixed integer programming model to optimize the infrastructure design and the reverse network flow. The proposed model attempts to minimize the total cost, which consists of transportation cost, operating cost, fixed cost for new facilities, final disposal cost and landfill cost, as well as the sale revenue of reclaimed materials. Results for various scenarios that consider various take-back rates and operating conditions are presented.

Abstract: Design and fabrication of microfluidic devices on polymethylmethacrylate (PMMA) substrates for analytical chemistry and biomedical-related applications using novel microfabrication methods are described. The image of microstructures is transferred from quartz master templates possessing the inverse image of the devices to plastic plates by using hot embossing methods. The microchannels on quartz master templates are formed by the combination of metal etch mask and wet chemical etching of a photomask blank. The micromachined quartz templates can be used repeatedly to replicate cheap and disposable plastic devices. The reproducibility of the hot embossing method is evaluated using 10 channels on different PMMA plastics. The relative standard deviation of the channel profile on the plastic chips is less than 1%. In this study, the PMMA microfluidic chips have been demonstrated as a microcapillary electrophoresis (μ-CE) device for DNA separation and detection. The capability of the fabricated chip for electrophoretic injection and separation is characterized via the analysis of DNA fragments ∅X-174-RF Hae III digest. Experimental results indicate that all of the 11 DNA fragments of the size marker could be identified in less than 2 min with relative standard deviations less than 0.4 and 8% for migration time and peak area, respectively. Moreover, with the use of a near-infrared (IR) dye, fluorescence signals of the higher molecular weight fragments (>603 bp in length) could be detected at total DNA concentrations as low as 0.1 μg/ml. In addition to DNA fragments ∅X-174-RF Hae III digest, DNA sizing of hepatitis C viral (HCV) amplicon is also achieved using microchip electrophoresis on PMMA substrates.

Abstract: The striatum is a crucial site of action for the motor effects of cannabinoids (CBs). However, the electrophysiological consequences of activation of CB receptors on the striatal neurons have not been established. Here we report for the first time that the cannabimimetic aminoalkylindole WIN 55,212-2 and the endogenous cannabinoid anandamide substantially depress corticostriatal glutamatergic synaptic transmission onto striatal neurons in the brain slice preparation. The selective CB1 receptor antagonist SR 141716 effectively reversed this inhibition. WIN 55,212-2 significantly increased the paired-pulse facilitation of synaptically evoked EPSCs, while having no effect on the sensitivity of postsynaptic neurons to [alpha]-amino-3-hydroxy-5-methylisoxazole-4-propionic acid. WIN 55,212-2 also reduced the frequency of spontaneous, action potential-dependent EPSCs (sEPSCs) without altering their amplitude distribution. Superfusion of WIN 55,212-2 elicited a membrane hyperpolarization accompanied by a decrease in input resistance. Both effects were blocked by intracellular caesium. In contrast, intracellular caesium failed to affect WIN 55,212-2-mediated synaptic inhibition. The WIN 55,212-2-mediated synaptic inhibition was blocked by the Gi/o protein inhibitor pertussis toxin (PTX), but not by the GABA(A) receptor antagonist bicuculline or GABA(B) receptor antagonist SCH 50911. Pretreatment with the N-type Ca2+ channel antagonist [omega]-conotoxin GVIA selectively abolished the WIN-55,212-2-mediated synaptic inhibition. These results suggest that cannabinoids depress the corticostriatal glutamatergic synaptic transmission through the activation of presynaptic CB1 receptors to inhibit N-type Ca2+ channel activity, which in turn reduces glutamate release. The presynaptic action of cannabinoids is mediated by a PTX-sensitive Gi/o protein-coupled signalling pathway.

Abstract: This paper describes a fast, low-cost but reliable process for the fabrication of microfluidic systems on soda-lime glass substrates. Instead of using an expensive metal or polisilicon/nitride layer as an etch mask, a thin layer of AZ 4620 positive photoresist (PR) is used for buffered oxide etching (BOE) of soda-lime glass. A novel two-step baking process prolongs the survival time of the PR mask in the etchant, which avoids serious peeling problems of the PR. A new process to remove precipitated particles generated during the etching process is also reported in which the glass substrate is dipped into a 1 M hydrochloride solution. A microfluidic channel with a depth of 35.95±0.39 µm is formed after 40 min BOE in an ultrasonic bath. The resulting channel has a smooth profile with a surface roughness of less than 45.95±7.96 A. Glass chips with microfluidic channels are then bonded at 580 °C for 20 min to seal the channel while a slight pressure is applied. A new bonding process has been developed such that the whole process can be finished within 10 h. To our knowledge, this is the shortest processing time that has ever been reported. In the present study, an innovative microfluidic device, a `micro flow-through sampling chip', has been demonstrated using the fabrication method. Successful sampling and separation of Cy5-labelled bovine serum albumin (BSA) and anti-BSA has been achieved. This simple fabrication process is suitable for fast prototyping and mass production of microfluidic systems.

Abstract: A total of 209 clinical isolates of Pseudomonas (193 Pseudomonas aeruginosa, 10 P. putida, 4 P. stutzeri, and 2 P. fluorescens isolates) with reduced susceptibilities to imipenem and/or ceftazidime were subjected to PCR assays with primers specific for blaIMP-1, blaIMP-2, blaVIM-1, and blaVIM-2 and sequence analysis to identify the metallo-β-lactamases (MBLs) prevalent among these organisms in Taiwan; and 21 isolates gave positive results. Five isolates including two P. putida and three P. stutzeri isolates were found to carry blaIMP-1, and six isolates including five P. putida and one P. stutzeri isolates harbored blaVIM-2. The remaining 10 isolates were P. aeruginosa, and all were found to carry a novel variant of blaVIM-2, designated blaVIM-3. There are only two nucleotide differences between blaVIM-2 and blaVIM-3, leading to two amino acid alterations. Our findings indicate that VIM-2 and its variant have become the most prevalent metalloenzymes in Pseudomonas in Taiwan. Southern hybridization with the blaVIM-2-, blaVIM-3-, and blaIMP-1 -specific probes revealed that only two VIM-2-producing P. putida isolates appeared to carry the MBL gene on plasmids. Pulsed-field gel electrophoresis showed that six VIM-3-producing P. aeruginosa isolates and two IMP-1-producing P. stutzeri isolates were genetically related, suggesting that the spread of these MBL genes in Taiwan could be due to clonal dissemination as well as genetic exchange between different clones.

Abstract: In vitro experiments have demonstrated that epidermal growth factor (EGF)-related peptides activate distinct subsets of ErbB receptors and differ in their biological activities. The implications of cross-talk among ErbB family receptors in human cancer, however, remain to be clarified. This cohort study was performed to examine the expression patterns of ErbB receptors by immunohistochemistry in primary human bladder cancer (n = 245) and compared with conventional biological indicators for their prognostic significance. Expression of individual EGF receptor (EGFR) and ErbB2, ErbB3, or ErbB4 receptors was detected in 72.2, 44.5, 56.3, and 29.8% of bladder cancer cases, respectively. Expression of two of the receptors varied from 14.7 to 42.4%, of three of the receptors between 11.0 and 22.0%, and of all four of the ErbB receptors by 8.6%. Important indicators in association with patient survival were tumor staging (P = 0.017), ErbB2 (P = 0.018), EGFR-ErbB2 (P = 0.023), and ErbB2-ErbB3 (P = 0.042). In the subset of grade-2 tumors, EGFR-ErbB2-ErbB3 and EGFR-ErbB2 predicted the development of second recurrence (P = 0.026 and 0.039, respectively), and ErbB2-ErbB3 tended to correlate with patient survival (P = 0.09). The results indicate that a combination of EGFR, ErbB2, and ErbB3 expression profile may be a better prognostic indicator than any family member alone. Given that ErbB2 is the preferred coexpression partner of ErbB family members, expression of other ErbB receptors may significantly affect the prognostic implication of ErbB2 for bladder cancer patients.

Abstract: The synthesis of Au/Pd bimetallic nanoparticles at various molar ratios by the coreduction of HAuCl4 and H2PdCl4 with hydrazine in the reverse micelles of water/sodium bis(2-ethylhexyl) sulfosuccinate/isooctane at 25 °C was investigated. Transmission electron microscopy photographs showed that the Au/Pd bimetallic nanoparticles obtained at the feeding compositions of [HAuCl4]/[H2PdCl4] = 9/1 to 1/9 essentially had uniform sizes and mean diameters of about 3 nm. Both the UV−vis absorption spectra and X-ray diffraction patterns for the bimetallic systems and the physical mixtures of individual metallic nanoparticles suggested the formation of bimetallic nanoparticles. The energy dispersive X-ray analysis on the single particle showed that the composition for each particle was roughly consistent with that of feeding solution. Abstract: We calculate the $B\to\pi,\rho$ transition form factors in the framework of perturbative QCD to leading power of $1/M_B$, $M_B$ being the $B$ meson mass. We explain the basic principle by discussing the pion electromagnetic form factor. It is shown that the logarithmic and linear singularities occurring at small momentum fractions of light meson distribution amplitudes do not exist in a self-consistent perturbative analysis, which includes $k_\perp$ and threshold resummations.

Abstract: This study tests middle managers’ perceptions on the association between supply chain quality management practices and organizational performance. Two instruments were used for the study: a revised version of Saraph et al.’s quality management instrument and Madu et al.’s organizational performance instrument. The data were classified into three groups based on the level of supply chain quality practices. It is observed that high quality‐tendency systems tend to perform better than low quality‐tendency systems on cost savings. High quality‐tendency organizations can be differentiated from medium quality‐tendency organizations in areas such as productivity, sales growth, and earning growth. Medium quality‐tendency systems seem to be differentiated from low quality‐tendency systems on indicators like employee satisfaction, productivity, and sales growth. Our results suggest that organizational performance could be enhanced through improved supply chain quality management.

Abstract: Polyphosphate- and polyhydroxyalkanoate (PHA)-accumulating traits of predominant microorganisms in an efficient enhanced biological phosphorus removal (EBPR) process were investigated systematically using a suite of non-culture-dependent methods. Results of 16S rDNA clone library and fluorescence in situ hybridization (FISH) with rRNA-targeted, group-specific oligonucleotide probes indicated that the microbial community consisted mostly of the alpha- (9.5% of total cells), beta- (41.3%) and gamma- (6.8%) subclasses of the class Proteobacteria, Flexibacter-Cytophaga (4.5%) and the Gram-positive high G+C (HGC) group (17.9%). With individual phylogenetic groups or subgroups, members of Candidatus Accumulibacter phosphatis in the beta-2 subclass, a novel HGC group closely related to Tetrasphaera spp., and a novel gamma-proteobacterial group were the predominant populations. Furthermore, electron microscopy with energy-dispersive X-ray analysis was used to validate the staining specificity of 4,6-diamino-2-phenylindole (DAPI) for intracellular polyphosphate and revealed the composition of polyphosphate granules accumulated in predominant bacteria as mostly P, Ca and Na. As a result, DAPI and PHA staining procedures could be combined with FISH to identify directly the polyphosphate- and PHA-accumulating traits of different phylogenetic groups. Members of Accumulibacter phosphatis and the novel gamma-proteobacterial group were observed to accumulate both polyphosphate and PHA. In addition, one novel rod-shaped group, closely related to coccus-shaped Tetrasphaera, and one filamentous group resembling Candidatus Nostocoidia limicola in the HGC group were found to accumulate polyphosphate but not PHA. No cellular inclusions were detected in most members of the alpha-Proteobacteria and the Cytophaga-Flavobacterium group. The diversified functional traits observed suggested that different substrate metabolisms were used by predominant phylogenetic groups in EBPR processes.
