Abstract: Silver nanoparticles (AgNPs) are widely used in the household, medical and industrial sectors due to their effective bactericidal activities and unique plasmonic properties. Despite the promising advantages, safety concerns have been raised over the usage of AgNPs because they pose potential hazards. However, the mechanistic basis behind AgNPs toxicity, particularly the sublethal effects at the organismal level, has remained unclear. In this study, we used a powerful in vivo platform Drosophila melanogaster to explore a wide spectrum of adverse effects exerted by dietary AgNPs at the organismal, cellular and molecular levels. Lethal doses of dietary AgNPs caused developmental delays and profound lethality in developing animals and young adults. In contrast, exposure to sublethal doses, while not deadly to developing animals, shortened the adult lifespan and compromised their tolerance to oxidative stress. Importantly, AgNPs mechanistically resulted in tissue-wide accumulation of reactive oxygen species (ROS) and activated the Nrf2-dependent antioxidant pathway, as demonstrated by an Nrf2 activity reporter in vivo. Finally, dietary AgNPs caused a variety of ROS-mediated stress responses, including apoptosis, DNA damage, and autophagy. Altogether, our study suggests that lethal and sublethal doses of AgNPs, have acute and chronic effects, respectively, on development and longevity by inducing ROS-mediated stress responses.

Abstract: An ultra-large voltage conversion ratio converter is proposed by integrating a switched-capacitor circuit with a coupled inductor technology. The proposed converter can be seen as an equivalent parallel connection to the load of a basic boost converter and a number of forward converters, each one containing a switched-capacitor circuit. All the stages are activated by the boost switch. A single active switch is required, with no need of extreme duty-ratio values. The leakage energy of the coupled inductor is recycled to the load. The inrush current problem of switched capacitors is restrained by the leakage inductance of the coupled-inductor. The above features are the reason for the high efficiency performance. The operating principles and steady state analyses of continuous, discontinuous and boundary conduction modes are discussed in detail. To verify the performance of the proposed converter, a 200 W/20 V to 400 V prototype was implemented. The maximum measured efficiency is 96.4%. The full load efficiency is 95.1%.

Abstract: In this letter, we report a one-step templating synthetic strategy to prepare aligned TiO2 nanotube and nanowire arrays on Si substrate from a solution at ambient temperature. The deposition of TiO2 and the selective-etching of the ZnO template proceeded at the same time through the careful control of process parameters. The different thickness of TiO2 sheaths, leading to the formation of nanotubes or nanorods, can be precisely controlled by the deposition time. The idea of selective etching and deposition is applicable to other oxide materials, and such a facile method is expected to find widespread applications.

Abstract: Experiments with amyloid-β (Aβ)-42-immunized transgenic mouse models of Alzheimer's disease have revealed amyloid plaque disruption and apparent cognitive function recovery. Neuropathological examination of patients vaccinated against purified Aβ-42 (AN-1792) has demonstrated that senile plaque disruption occurred in immunized humans as well. Here, we examined tissue histology and quantified and biochemically characterized the remnant amyloid peptides in the gray and white matter and leptomeningeal/cortical vessels of two AN-1792-vaccinated patients, one of whom developed meningoencephalitis. Compact core and diffuse amyloid deposits in both vaccinated individuals were focally absent in some regions. Although parenchymal amyloid was focally disaggregated, vascular deposits were relatively preserved or even increased. Immunoassay revealed that total soluble amyloid levels were sharply elevated in vaccinated patient gray and white matter compared with Alzheimer's disease cases. Our experiments suggest that although immunization disrupted amyloid deposits, vascular capture prevented large-scale egress of Aβ peptides. Trapped, solubilized amyloid peptides may ultimately have cascading toxic effects on cerebrovascular, gray and white matter tissues. Anti-amyloid immunization may be most effective not as therapeutic or mitigating measures but as a prophylactic measure when Aβ deposition is still minimal. This may allow Aβ mobilization under conditions in which drainage and degradation of these toxic peptides is efficient.

Abstract: This review reports on recent advances in state-of-the-art detection methods for microfluidic paper-based analytical devices (μPADs). The review commences by describing the materials, fabrication methods and driving mechanisms employed in μPADs. The review then explores the main detection methods and applications of μPADs proposed over the past five years. The discussions focus on seven detection technologies, namely (1) colorimetric detection, (2) fluorescent detection, (3) electrochemical detection and photoelectrochemical detection, (4) chemiluminescent detection, (5) electrochemiluminescent detection, (6) nanoparticle-based detection, and (7) spectrometry detection. Overall, the review provides a useful insight into recent advances in the μPAD detection field and serves as a useful source of reference for further research and innovation in the field.
